Behavioral, electroencephalographic, and biochemical alterations have been studied in rats with porta-caval shunt, up to 45--50 days after the operation. No behavioral or electroencephalographic changes have been observed, while modifications of various amino acids both in plasma and brain have been found. Among all the amino acids considered of particular significance are the plasma and brain increases of tyrosine, phenylalanine, and tryptophan, since they are the precursors of biogenic amines. In fact, the increase of 5-HIAA found in regions of CNS which are richest in serotoninergic synapsis could indicate an increased turnover of 5-HT. On the other hand no significant alterations of turnover of catecholamines have been found.
A peptic-tryptic-cotazym (PTC) digest of a crude wheat gliadin preparation was obtained under experimental conditions simulating in vivo protein digestion and then fractionated into 10 peaks by ion-exchange chromatography. PTC-gliadin digest and one of its subfractions (coded as fraction 9 according to its elution pattern) were very active in inhibiting in vitro development and morphogenesis of small intestine from 17- and 18-day-old rat fetuses, whereas they were harmless for the culture of jejunum from 21-day-old fetuses. PTC-digest also induced extensive tissue degeneration and necrosis of in vitro cultured small intestinal mucosa from patients with active celiac disease (gluten-induced entheropathy), but did not cause any detectable effect on histologically normal human small intestinal mucosa. Some wheat albumin and gliadin fractions were also tested on in vitro developing small intestine from 17-day-old rat fetus. Among all the tested protein fractions, only one gliadin fraction (coded as alpha 10-gliadin from its gel electrophoretic mobility) exhibited a toxic effect; morphologic alterations induced by alpha 10-gliadin were similar to those induced by PTC-digest and fraction 9.
Measurement of free amino acids in seminal plasma from fertile and infertile oligozoospermic subjects revealed a quantitative reduction of these molecules in infertile patients. Study of the effect of acidic media on seminal plasma appears to suggest the primary role of free amino acids in the "protection" of spermatozoa in the vagina, and the failure of sperm progression in the female genital tract may be due to a decrease of these molecules in semen of infertile patients.

An experimental scheme for the evaluation of two parameters of validity of an analytical method, precision and accuracy, has been reported. Particular emphasis has been given to the problem of comparison between two methods. In the description a validity limit of the scheme has been given and certain critical points, commonly encountered in the measurement of the above mentioned parameters during the preparatory as well as the experimental phase, has been underlined. Further, with the help of tables and graphs, an example for electronic elaboration of the data obtained during the course of standardization of an automatic method for the determination of uric acid in serum according to the above scheme, has been given.
A centrifuge speed analyzer, used for triglyceride, cholesterol, glucose and serum uric acid determinations, was tested as to linearity, precision, steadiness and accuracy requirements. The results were compared to those obtained by other techniques. Automatic techniques were proved to comply with reliability and practicability requirements, necessary during the screenings for preventive medicine. The centrifuge speed analyzer seems to be particularly fit for the primary prevention trials of coronary heart disease, since the follow-up studies require repeated laboratory examinations.
The method of Butts and Coll. for determination of serum thiocyanate--parameter correlated to smoke assumption--has been modified in order to improve sensibility and has been characterized as regards precision and accuracy. The improved method has been successively utilized for determination of serum thiocyanate of 493 subjects during a screening for prevention from coronary heart disease. A linear correlation (r = 0,916) between the number of smoked cigarettes and serum thiocyanate has been found. Assuming 40 mumol/l as a cut-off point to distinguish cigarette smokers and non smokers, 1,8% false positive and 5% false negative resulted.
Plasmatic concentrations of amino acids valine, methionine, isoleucine, leucine, tyrosine and phenylalanine, in 4100 newborns of age varying between 1 and 18 days have been analyzed. The samples were randomly taken among the newborns of Lazio, Abruzzi and Molise subjected to neonatal screening for aminoacidopathies at the Centro delle Oligofrenie Dismetaboliche, Istituto di Neuropsichiatria Infantile. Average concentrations in mumoles/100 ml of the aminoacids and their correlations with weight and age have been studied. A comparison of the results of this research with the already published data is made, and the cut off points in mass screening for aminoacidopathies are calculated.

An automatic manifold for the organic phosphorus mineralization is described. The device at issue does consist in an electrically heated aluminium blok, in which 80 holes are drilled, in order to hold 80 hydrolysis vials. The heating is automatically controlled by an independent unity. The ortophosphate determination is performed photometrically with the previously described automatic apparatus.
