Response of rat aortic adventitial fibroblasts to oestradiol treatment in vivo. Role of the cell tubulin system.

The fine morphological response was studied of rat aortic adventitial fibroblasts to oestradiol treatment in vivo. Fibroblasts from control rats revealed a well-developed Golgi complex, rough endoplasmic reticulum and cytoplasmic microtubules. Golgi-related vacuoles, elongated and flattened rough endoplasmic reticulum cisterns and microtubules associated with both these organelles were found in these cells. In contrast, dilated rough endoplasmic reticulum cisterns mainly of the vacuolar type were a conspicuous finding in fibroblasts from oestradiol treated rats. The Golgi complex was less expressed, and these cells contained no microtubules. The cytoplasmic microtubules and/or membrane bound tubulin (rough endoplasmic reticulum cisternal membrane bound tubulin) are suggested to be the cell targets on which oestradiol exerts its primary, cholchicine-like effect, resulting in the cellular response of rat aortic adventitial fibroblasts, in a dilatation of the cisterns of rough endoplasmic reticulum.

[Ultrastructural similarities of secretory processes in arterial smooth muscle cells and elastic cartilage chondrocytes. vacuolar apparatus and microtubules (author's transl)].

A highly developed vacuolar apparatus is found in aorta and pulmonary trunk smooth muscle cells and elastic cartilage chondrocytes, and its relationship with cytoplasmic microtubules is described. The Golgi complex as well as the rough endoplasmic reticulum (RER) may be the intracellular sources of the components of the vacuolar apparatus. Some vacuoles, Golgi-related only, showed an association with microtubules. Smooth muscle cells which contained vacuoles derived from RER did not possess Golgi vacuoles and microtubules. In chondrocytes, both Golgi-related and RER vacuoles coexisted in a single cell. 2 main secretory pathways are suggested to operate in smooth muscle cells and chondrocytes: Golgi and RER pathway. The former requires an intact cell tubulin system (cytoplasmic microtubules and membrane-bound tubulin), while the latter is probably microtubule-independent.

Fine morphological aspects of the secretory process in arterial smooth muscle cells. II. Role of microtubules.

Preliminary results are presented regarding the role of the microtubular system in the secretory process in the rabbit aorta and pulmonary trunk smooth muscle cells. The results are based on electron microscopic findings of the effect of colchicine in vivo. On colchicine treatment an accumulation of secretory granules versus a vacuolar type of dilatation of the rough endoplasmic reticulum cisternae distinguishes two groups of smooth muscle cells. It is suggested that two secretory pathways operate in these cells, and that the cell microtubular status plays a key role in triggering one or the other pathway.
