The effect of microorganisms, necrotic tissues and a foreign body on the development of festering process in rat wounds was studied. It was found that the presence of necrotic tissues is the necessary and sufficient condition for a clinical manifestation of infection in rat wounds. Additional infection of the wounds and introduction of a foreign body did not appreciably change the clinical picture. A model is suggested of a festering wound in rats without artificial infecting. In the festering wound, one could observe the replacement of the gram-positive coccal microflora by the gram-negative one. Meanwhile the gram-positive coccal microflora was predominant in the non-festering wound. The level of potential biochemical indicators of infection in the blood of animals with festering wounds was higher than in the blood of those with non-festering wounds. The morphology, and the content of microbial cells, the content of nucleic acids, and total proteolytic activity in festering wound tissues were examined over time.

Autoradiography and scanning electron microscopy were-used to assess new Soviet dressing materials in experiments on rats. It was shown that the two-layer nonwoven atraumatic material, caproic gauze, a modified surface active agent and kombutek-11 exert favourable effects on the regenerative processes in the wounds and promote more rapid wound healing as compared with a conventional cotton-gauze bandage. It is advisable to devise dressing materials possessing wound-healing action similar to kombutek-11.
The possibilities of the scanning electron microscopy (SEM) technique were evaluated in the studies of functional properties of dressing materials. It was shown that the use of SEM enables the disclosing of the effect of structural characteristics of the dressing materials on their atraumaticity, the appraisal of the absorption capacity of the dressings, and the exploration of the time course of wound healing under the dressings.
A study was made of the effect of potassium orotate on wound healing. The duration of wound healing was 3 days less in animals given potassium orotate as compared to controls, with the rate of prolin-containing protein synthesis by fibroblasts and the rate of their egress into the intercellular space being increased. No excess fibrillogenesis was noted. Hydroxyprolin content in the granulation tissue of wounds differed little in control and in experimental animals.
A study was made of the effect of glycine given in doses approximating the physiological ones on the repair of processes in rat wound tissues. It was disclosed that in the early periods of wound healing, glycine administration leads to the increased content of cAMP and cAMP/cGMP ratio in the wound muscle and then in the granulation tissue, which appears to promote the intensification of the repair processes manifesting in the changes in tissue metabolism (DNA, collagen), in anti-inflammatory action, as well as in a more rapid maturation of the granulation tissue and wound reduction. It was also found that the doses of glycine tested do not affect the content of insulin and hydrocortisone in the blood of experimental animals.
A study was made of the effect of imidazole on wound healing. The duration of wound healing was 2 days less in animals given imidazole as compared to controls, with the rate of prolin-containing protein synthesis by fibroblast and the rate of their egress into the intercellular space being increased. No excess fibrillogenesis was noted. Hydroxyproline content in the granulation tissue of wounds differed little in control and in experimental animals.
Experiments were made to examine atraumatic properties of 3 samples of dressing material. A dressing made of cotton wool and gauze served as control. It was shown that atraumatic properties of dressing material can be reliably evaluated by cytological examination of wound prints. Atraumatic properties of dressings are determined by structural features of materials used for the manufacture of dressing.
An experimental study was made of the effect exerted by prednisolone in a dose of 4 mg/kg body weight on wound healing. It was shown that prednisolone reduces the period of inflammation, accelerates the formation of granulation tissue and epidermis. All these effects are accompanied by diminution of the mitotic activity of fibroblasts and dramatic increase in the intensity of RNA synthesis in them. Dislocation of newly synthetized RNA from the nucleus to the cytoplasm proceeds at a greater rate as compared to control. The differentiation of fibroblasts is accelerated.
Studies on the effect of cGMP on the proliferation of granulation tissue and collagenogenesis in experimental wound healing have shown that cGMP promoted more rapid formation and differentiation of fibroblasts rose thereby promoting an accelerated formation of collagen. Histological and biochemical findings correlated with wound planimetry data. In the course of stimulation the wound healing surfaces diminished more quickly as compared to the control.
Electron microscopic examinations of the white blood in 7 patients with purulent wounds and clinical manifestations of sepsis and in 1 patient with purulent wounds complicated by the development of bacterial shock as well as of the ultrastructure of bacteria isolated from the wound content of the patients were carried out. The controls consisted of electron microscopic examinations of the blood from two donors. The blood of patients with sepsis was found to contain free microbial cells and those phagocytized by neutrophilic leukocytes. Free bacteria formed large colonies enveloped in a strong membrane in the blood. Some leukocyte-phagocytized microbes underwent lysis, the others, surrounded by an envelope were located in large vacuoles in the blood cell cytoplasm (incomplete phagocytosis). Numerous microbial cells both free and phagocytized were particularly frequently found in bacterial shock accompanied by the clinical picture of most acute septicemia.
The intensity of uridine-53H and proline-3H incorporation into fibroblast nuclei and cytoplasm as well as into intercellular spaces was studied in wound healing of mice given potassium orotate and in the control animals. It was shown that proline-3H incorporation was more intensive than that of uridine-53H. At the same time a significant increase in RNA synthesis in fibroblasts under the action of potassium orotate, and a less pronounced rise in proline-containing protein synthesis were noted in similar conditioelationships correlate with the fibroblast ultrastructural changes in the process of their differentiation.
The rate of uridine-5 3H incorporation into the skin-muscle wound fibroblasts of control and experimental animals, and also into the fibroblasts of flocculent connective tissue of intact animals was studied. The character of incorporation and distribution of the isotope in the nuclei and cytoplasm in the flocculent connective tissue fibroblasts differed in intact mice from those in the wound fibroblasts only by intensity. Potassium orotate RNA synthesis stimulation in the fibroblast nuclei closely correlated with the increased rate and intensity of the labeled RNA passage from the cell nuclei to the sites of collagen synthesis.
The changes in the DNA synthesis in the granulation tissue fibroblasts in the wounds of adult animals given potassium orotate and in controls were studied. For this purpose a daily autoradiographic examination of the wound areas were conducted in the both groups of animals. It appeared that under the effect of potassium orotate (0.2 ml of 2% solution) there was more frequent involvement of fibroblasts into the DNA synthesis; under conditions of the wound process stimulation there occurred an increase of the proliferative activity of these cells.
