[Cold preservation of human sperm].

The authors report the development and investigation of a new technique for freezing and storage of human spermatozoa. Utilizing isolated container of different kind the average rate of freezing was between 1,3 grd/min and 14 grd/min in the sphere from 5 degrees C to--10 degrees C and between 2,9 grd/min to 55 grd/min in the sphere from--10 degrees C to--60 degrees C. The technical expense was low. Semen specime were provided by 33 andrological patients. A total of 53 samples of human semen were frozen and stored for periods of time up to 25 days. The authors used the cryoprotective medium from Behrman and Sawada. The best results were obtained in the mode of freezing at 2,8 grd/min respectively 12 grd/min in ejaculates of original motility about 80%. The recovery of motility and fertilizability of spermatozoa was 50% of original motility, in cases of oligospermia only 34%. The results of this new technique of freezing and storage of human spermatozoa are similar to other new reports in literature.

[Occult blood in stool. A field trial in cancer screening (author's transl)].

4000 male probands aged 45 to 69 were investigated in a field trial according to random distribution for undetected intestinal bleeding with the Haemoccult - method. With a participation of 28% only 39 out of a total of 65 positive cases could be evaluated. Four cases of colon or rectal carcinoma and four cases of colonic polyps were ascertained. In view of complete lack of experience with false negative test results the Haemoccult test is at present a useful addition to the diagnostic possibilities. However, in suspected cases even a repeatedly negative test cannot replace complete colo-rectal investigation.

[The reliability of myocardial scintigraphy with thallium201, A comparison with lacvocardiography and perfusion scintigraphy (author's transl)].

Eighteen patients were investigated by laevocardiography, perfusion scintigraphy and with a 201thallium scan. In four patients with normal contractility of the left ventricle, normal uptake of 202thalium was found in every patient. Hypokinesia of the anterior wall or apex of the ventricle showed normal uptake on the 201thallium scintigram in four out of five patients. It follows that it is not usually possible to diagnose hypokinetic mural portions on 201thallium scintigrams without quantitative analysis. Our experience up to date with areas of akinesia has shown that it is possible to demonstrate areas of scarring (as negative contrast) in seven out of nine cases.

Molecular and cellular effects of antisickling concentrations of alkylureas.

Alkylureas are capable of inhibiting sickling in vitro and the gelation of solutions of hemoglobin S at concentrations between 0.05 and 0.1 M with increasing effectiveness that is directly proportional to the length of the alkyl chain (butyl greater than propyl greater than ethyl greater than methyl). 6The inhibitory effect is independent of pH between 6.5 and 7.5 and is a process driven by entropy. The alkylureas at concentrations of 0.1 M have minimal effects on several erythrocyte functions. Oxygen equilibria, osmotic fragility, reduced glutathione content, and glutathione reductase activity are totally unaffected, while pyruvic kinase activity is decreased only by butylurea by about 20%, and glucose-6-phosphate dehydrogenase activity is decreased progressively to a maximum of 30% in direct proportion to the length of the alkyl chain. Alkylureas not only inhibit sickling but are also capable of desickling erythrocytes that have been maintained in the deoxygenated state. They have little effect on several erythrocyte functions at antisickling concentrations, but their toxicity must be evaluated before they can be examined as potential therapeutic agents for the treatment or prevention of acute episodes in sickle cell anemia.

[Metabolism of essential and endogenous serum fatty acids in healthy persons, coronary disease and patients with lipid metabolism disorder. Partial aspect of high risk problem patients with age-related lipid and fatty acid concentrations in serum and regulation of long-chain products from essential and endogenous precursors in healthy subjects].

Total lipid, free and esterified cholesterol and triglycerides significantly increase in concentration with increasing age. Palmitic, palmitoleic, oleic and linoleic acid behave in the same way. Free fatty acids, phospholipids and stearic acid show the same concentrations in all groups of age. C20 fatty acids demonstrate a competitive regulatory mechanism of desaturase activity in their age dependant distribution. While the 20:3(n-9), a product of the endogenous (n-9) family, increases, the 20:3(n-6) and 20:4(n-6), both desaturation and chain elongation products of the essential n-6 family, decrease. Such behavior has been demonstrated in many feeding studies (12, 13, 15, 16, 17, 29) and is interpreted as a result of competitive substrate interaction on desaturase activity.
