DNA flow cytometry of histological material from human gastric cancer.

DNA flow cytometry was performed on fixed embedded histological material from 56 radical gastrectomies for human gastric cancer. The ploidy and proportion of cells in the DNA synthetic phase of the cell cycle (S phase fraction) were estimated and the results correlated with histological features. DNA aneuploidy was encountered in 73 per cent of cases. Aneuploid and diploid tumours both had significantly higher fractions of cells in S phase than normal mucosa. S phase fractions for aneuploid tumours were higher than diploid tumours. There was a trend for tumours exhibiting an infiltrating mode of growth and poor tubule formation to have a diploid DNA content and low S phase fraction. No difference was observed between the results obtained from tumours with or without lymph node metastases. Early uniform fixation greatly improved the quality of the flow cytometry results measurable by a reduction in the mean coefficient of variation.

Extraskeletal myxoid chondrosarcoma: a histochemical and immunohistochemical study.

In reviewing a large series of soft tissue sarcomas, nine cases of extraskeletal myxoid chondrosarcoma have been retrieved. These tumours, which principally presented in middle-aged adults, have been examined histochemically to determine the heteroglycan content of their myxoid matrix and immunohistochemically for the presence of S-100 protein. The principal mucopolysaccharides identified were chondroitin-4 and 6-sulphate and keratan sulphate; each of the tumours was S-100 positive. The relevance of these findings to the histogenesis and differential diagnosis of these uncommon neoplasms is discussed.

Prognostic importance of DNA flow cytometry in non-Hodgkin's lymphomas.

Eighty one cases of non-Hodgkin's lymphoma were examined by DNA flow cytometry, using fixed embedded histological tissue. The frequency of detection of DNA aneuploidy and the values for S phase fractions depended on the histological subtype and grade of lymphoma. Twenty two of the patients with low grade centroblastic/centrocytic non-Hodgkin's lymphoma had repeat biopsies. Eleven of these patients remained histologically and cytometrically stable, but the remaining eleven transformed into high grade non-Hodgkin's lymphoma. The mean value for the S phase fraction in the initial biopsy specimens from patients which transformed was higher than that for patients whose lymphomas remained stable (p less than 0.001). It is proposed that estimates of S phase fraction prospectively identify patients with low grade non-Hodgkin's lymphoma at risk from transformation.

Anxiety of non-rooming-in parents of hospitalized children.

This study examined levels of anxiety in 51 parents who did not room in with their young hospitalized children. The Spielberger State-Trait Inventory was used to measure parental anxiety twice, with a 3-day interval. Levels of anxiety were significantly higher in parents with a greater number of children at home, less education, and lower social status. Findings of clinical significance were: the longer the child's hospitalization the more anxious parents became, especially if there were children at home; hospitalization of a child was more stressful for fathers because they maintain lower trait anxiety than mothers; and as parental visits to their hospitalized child decreased, parental anxiety levels increased.

A new monoclonal antibody to epithelial membrane antigen (EMA)-E29. A comparison of its immunocytochemical reactivity with polyclonal anti-EMA antibodies and with another monoclonal antibody, HMFG-2.

Two polyclonal rabbit antibodies to epithelial membrane antigen (EMA), two mouse monoclonal antibodies (E29 and HMFG-2), and a "cocktail" of these two monoclonals have been compared using an indirect immunoperoxidase technique. Sections from 25 tissues (17 malignant and 8 benign), were examined. The distribution of staining with each of these reagents was similar, but the polyclonal antibodies produced stronger staining in colorectal carcinomas and lactating breast, whereas staining with the monoclonal antibodies was stronger in non-neoplastic pleural mesothelium and in pulmonary alveolar cells. When the two monoclonals were mixed there was no increase in staining intensity. E29 gave a "cleaner" result than HMFG-2, with better discrimination between cells and stroma, and is highly suitable for routine diagnostic histopathology.

Impact of a province-wide nurses' strike on medical care in a regional referral centre.

A 4-week, province-wide nurses' strike in Alberta in 1982 caused the closure of 57% of the acute care beds, including 47% of the intensive care beds, in Calgary. The effects of the strike on patient care at Foothills Provincial General Hospital, where nurses did not strike, were assessed. The number of emergency admissions, severity of illness and rate of death in the intensive care unit increased. On the other hand, the rate of death, length of stay and number of unexpected deaths on the medical wards were similar to those in the control periods before and after the strike. A subjective perception by hospital personnel of deteriorating patient care caused much anxiety; however, the results of analysis of measurable aspects of care suggested that the patients admitted to hospital received care during the strike that was comparable to care given before or after the strike. The inconvenience and potential harm to the patients not admitted because they had less severe illness were not measured.

Humoral antibodies to anaerobic bacteria isolated from patients with pulpal-periapical disease.

The purpose of this investigation was to determine if anaerobic bacteria isolated from pulpal-periapical infections elicit a humoral immune response in the host. Results indicated that patients with pulp and periapical disease do not have significantly raised levels of IgG or IgM in their serum and that in the acute sera the precipitin antibody titer to the anaerobic bacteria was significantly higher in the patient group than in the control group. However, there was no significant difference in antibody titer for the convalescent sera in the patient and control groups or the acute and convalescent sera for the patient group.

Synthesis and evaluation of the male antifertility properties of a series of N-unsubstituted sulfamates.

A series of six aliphatic and one carbocyclic N-unsubstituted sulfamates have been synthesized and evaluated as potential male antifertility agents. Three of the aliphatic sulfamates, 1,2-ethanediyl sulfamate (1), 1,3-propanediyl sulfamate (2), and 1,4-butanediyl sulfamate (3), when administered orally to male rats caused a decrease in the number of pregnant females and/or implantation coupled with increased embryonic and fetal resorption. The compounds were prepared by treating the appropriate glycol salt with sulfamoyl chloride or by the cleavage of a tert-butylsulfamate with trifluoroacetic acid.
