Molecular forms of acetylcholinesterase in the developing chick visual system.

The developmental profiles of the enzyme acetylcholinesterase, and of some of its quaternary structural forms, characterized by discrete sedimentation coefficients, have been comparatively analyzed in chick retina and optic tectum, between embryonic day 8 and day 10 after hatching. Four molecular species of AChE have been characterized in both retina and tectum during this developmental period: two of them with sedimentation coefficients of 11S and 6S, accounting together for 94-99% of the AChE activity in the initial homogenate, can be easily extracted by homogenization in a buffer containing 1% Triton X-100 and 1 M NaC1, at 4 degrees C. The other two, however, are not extractable by such treatment, but can be released by collagenase from the residue left after the detergent-salt extraction; they have apparent sedimentation coefficients of 21.5S and 16.5S and represent, together, less than 2% of activity in the initial homogenate. All four forms of the enzyme show distinctive patterns of change during the developmental period considered, with significant differences between retina and tectum. These differences are discussed in the context of the specific roles of retina and tectum in the visual process.

Structure and assembly of phage phi29.

Bacteriophage phi29 is a small, morphologically complex, virus with a DNA of molecular mass 12 X 10(6). The most likely structure of the head of phi29 consists of two fivefold symmetric end-caps based on T = 1 icosahedral symmetry, separated by an equatorial row of 5 hexamers. The eighteen genes identified in phi29 genome have been mapped and, in some cases, the gene products have been identified. Five linked genes, four coding for structural proteins (G, A, E, H) and one coding for a non-structural protein (J), are essential to determine the normal shape of the capsid. Protein pJ may be a scaffolding protein. An account of the effects of mutations in phi29 genes is given.

Fluorescent berberine binding as a marker of internal glycosaminoglycans sulfate in bovine oocytes and sperm cells.

The use of berberine as a biological marker of glycosamineglycans sulfate was employed to corroborate the presence of heparin in mammalian oocytes and sperm and its distribution in all the structures, or only in some specialized zones, of the male and female gametes. Oocytes and sperms were treated with 1.8 mM berberine for the presence of heparin and examined 10, 30, 60, and 120 minutes later. We have found that heparin is homogeneously distributed in all the zones of bovine oocytes and in sperm cells. When sperm cells are first treated with 80 microM of heparin and then berberine, 40% of them display in their post acrosomal region an intense yellow fluorescence. This may be in relation to the high amount of heparin binding sites due to the presence of the reticular membranous like system in this sperm region and in its possible role whereby gametes recognize and adhere to one another. Therefore, the use of berberine as a fluorescent marker of heparin represents clear proof of the presence of GAGs and their binding sites in the outside and inside of mammalian gametes, reinforcing the importance they play in the events of the process of fertilization.

DNA unpacking in guinea pig sperm chromatin by heparin and reduced glutathione.

The kinetics of nuclear decondensation and DNA unpacking induced by the action of a physiological concentration of heparin and glutathione of guinea pig spermatozoa was studied. Sperm (acrosomeless) suspensions were incubated at several different temperatures (37, 40, 43, and 46 degrees C), with a constant concentration of either heparin (50 microM) or reduced glutathione (12.5 mM) and increasing concentrations of the other reagent. Nuclei spermatozoa remained highly condensed when incubated in the medium alone or in either GSH or heparin alone for up to 72 h. Swelling of nuclei spermatozoa was initially observed during the first 20 min of incubation. The sperm nuclei initiate decompaction at the central part of the nuclear structure while at the periphery there remain numerous residues of densely packed chromatin. The swollen chromatin pattern presents the characteristic organization into "hub-like" nuclear bodies that measured 10-100 nm diameter joined by a network of chromatin fibers. At full nuclei decondensation chromatin end fibers are loose, probably meaning that DNA is not organized into loop domains. DNA presence was verified by the use of ethidium bromide and acridine orange.

Iron metabolism, inflammation and anemia in critically ill patients. A cross-sectional study.

INTRODUCTION: For critically patients, enteral immunonutrition results in notable reductions in infections and in length of stay in hospital, but not on mortality, raising the question as to whether this relate to the heterogeneous nature of critically ill patients or to the absence of the altered absorption of specific nutrients within the immunonutrient mix (e.g. iron). Immune-associated functional iron deficiency (FID) is not only one of the many causes or anaemia in the critically ill, but also a cause of inappropriate immune response, leading to a longer duration of episodes of systemic inflammatory response syndrome and poor outcome. OBJECTIVE: This prospective cross-sectional study was undertaken to assess the prevalence of FID in critically ill patients during their stay in intensive care (ICU) in order to find the more appropriate population of patients that can benefit from iron therapy. METHOD: Full blood cell counts, including reticulocytes (RETIC), serum iron (SI), transferring levels (TRF) and saturation (satTRF), serum TFR receptor (sTfR), ferritin (FRT) and C-reactive protein (CRP) were measured in venous blood samples from 131 random patients admitted to the ICU for at least 24 h (Length of ICU stay, LIS; min: 1 day; max: 38 days). RESULTS: Anaemia (Hb < 12 g/dL) was present in 76% of the patients (Hb < 10 g/dL in 33%), hypoferremia (SI < 45 microg/dl) in 69%; satTRF < 20% in 53%; FRT < 100 ng/mL in 23%; sTfR > 2.3 mg/dL in 13%; and CRP > 0.5 mg/dL in 88%. Statistically significant correlations (r of Pearson; *p < 0.05, **p < 0.01) were obtained for serum CRP levels and WBC**, Hb*, TRF**, satTRF*, and FRT**. There was also a strong correlation between TRF and FRT (-0.650**), but not between FRT and satTRF or SI. LIS correlated with Hb*, CRP**, TRF*, satTRF* and FRT**. CONCLUSIONS: A large proportion of critically ill patients admitted to the ICU presented the typical functional iron deficiency (FID) of acute inflammation-related anaemia (AIRA). This FID correlates with the inflammatory status and the length of stay at the ICU. However, 21% of the ICU patients with AIRA had an associated real iron deficiency (satTRF < 20; FRT < 100 and sTfR > 2.3). Since oral supplementation of iron seems to be ineffective, all these patients might benefit of iv iron therapy for correction of real or functional iron deficiency, which in turn might help to ameliorate their inflammatory status.

[Analysis of some variables in patients with hydatidosis surgically treated at the Regional Hospital of Valdivia, Chile, 1976-1986].

A survey of surgically treated cases of hydatidosis in the province of Valdivia, Chile, was done. Data were provided by the Regional Hospital of Valdivia, and a period of eleven years was covered (1976-1986). Disease, treatment and related data from each patient were consigned. For the analysis, the information was grouped according to: sex, age, cyst location, condition of the cyst, concomitance with another pathology, condition at time of discharge, hospitalization period and number of repeat hospitalizations. In the period studied, 197 cases of hydatidosis were surgically treated. The sex distribution was 53.3% for males and 46.7 for females; the age group 10-19 years was the most affected by the disease (19.3%). The most frequent locations of hydatid cysts were the liver (58.9%) and lungs (36.5%); the 38.6% of the patients showed complicated cysts, mainly with bacterial infections (11.7%); also, 20% of the cases presented a concomitant liver-related pathology. After the operation, 153 (77.7%) clinically recovered and 27 (13.7%) non-recovered patients left the hospital and 7 (3.5%) cases died while hospitalized. The average hospitalization period was 29 days. During 1976-1986, 24 patients were rehospitalized.

[Human hydatidosis in Valdivia, Chile: retrospective survey at the Valdivia Regional Hospital. 1987-1991].

A retrospective hospital survey of surgically confirmed cases of hydatidosis in the province of Valdivia was carried out. Data were provided by the Valdivia Regional Hospital and a five year (1987-1991) period was covered. Morbidity, mortality and case fatality rates were calculated. Descriptive epidemiological characteristics of the disease were determined. In the period studied, 137 new cases were registered. The incidence rate for the period was 9.1 per 100,000 inhabitants. The mortality rate was 0.2 per 100,000 inhabitants and the average fatality rate was 2.2%. The over-all sex distribution was 51.8% for females and 48.2% for males. The age group 10-19 years was the most affected by the disease (30.7%). The distribution according to activity revealed a higher proportion of house-wives (29.2%) and students (23.2%) with the disease. Hydatidosis was more frequent in Valdivia (23.4%), Panguipulli (17.5%) and La Unión (15.5%) and in patients from rural areas (73.7%).

[Accumulation and depuration of aldrin in the oyster Crassostrea rhizophorae, of the Ciénaga Grande de Santa Marta (Colombian Caribbean)].

Laboratory experiments were conducted at different salinities (35, 26 and 17/1000) to determine accumulation and depuration rates of the organochlorine compound aldrin by the oyster Crassostrea rhizophorae, Ciénaga Grande de Santa Marta (Colombian Caribbean). Residual levels were detected in tissues after 24 hours of exposure, showing increases directly related to the exposure concentration. For all the exposure concentrations the highest retention values were measured at 26/1000 salinities, followed by 17/1000, and the lowest values at the highest salinity. The highest bioconcentration values were 96115.5 (at 26/1000), 37938.9 (at 17/1000), and 22605.1, (35/1000) times the environmental concentration. The depuration process varied across salinity levels. At 26/1000 there was a clear decrease in the residual levels during the first five days, eliminating in this period the same amount as oysters kept at 17/1000 for 20 days and surpassing the amount eliminated by those kept at 35/1000 for 20 days. A rapid decrease in the amount of residues is followed by a slow elimination of the aldrin remaining in tissues.
