Abundant GFP expression and LTP in hippocampal acute slices by in vivo injection of sindbis virus.

Virus-mediated gene transfer into neurons is a powerful tool for the analysis of neuronal structure and function. Recombinant sindbis virus has been previously used to study protein function in hippocampal neuron cultures as well as in hippocampal organotypic slice cultures. Nevertheless, some concern still exists about the physiological relevance of these cultured preparations. Acute hippocampal slices are a widely used preparation for the study of synaptic transmission, but currently recombinant gene delivery is usually achieved only through time-consuming transgenic techniques. In this study, we show that a subregion of the CA1 area in acute hippocampal slices can be specifically altered to express a gene of interest. A sindbis virus vector carrying an enhanced green fluorescent protein (EGFP) reporter was injected in vivo into the hippocampus of adult rats. After 18 h, rats were killed, and acute hippocampal slices, infected in the CA1 field, were analyzed morphologically and electrophysiologically. Infected slices showed healthy and stable electrophysiological responses as well as long-term potentiation. In addition, infected pyramidal cells were readily recognized in living slices by two-photon imaging. Specifically, the introduction of an EGFP-Actin fusion protein greatly enhanced the detection of fine processes and dendritic spines. We propose this technique as an efficient tool for studying gene function in adult hippocampal neurons.

Evaluation of flexible cloth electrodes for electrodermal activity recording.

BACKGROUND: Instrument selection for recording physiological data in flight studies requires careful attention to subject comfort and non-interference with aircrew activities. Several electrode types and recording sites may be used to examine electrodermal activity (EDA). Placement of electrodes on the foot minimizes interference with physical activity and reduces motion artifacts; however, use of conventional, hard-plastic-encased metal (PEM) electrodes within a flight boot can produce discomfort and pressure-induced artifacts. HYPOTHESIS: When applied with proper electrolyte gels, thin, flexible, silver-impregnated cloth electrodes should acquire EDA signals qualitatively similar to those acquired using conventional, PEM electrodes. METHODS: EDA responses evoked by light flashes, auditory stimuli and valsalva maneuvers were recorded with cloth and PEM electrodes simultaneously from both feet of 4 male subjects. Performance of cloth vs. PEM electrodes and variability of signals recorded with the same electrode type were examined by placing pairs of selected electrodes on each foot of the subjects. Placements were balanced with respect to age and handedness of the subject and the number of trials with each electrode type placed on the left or right foot. RESULTS: Qualitatively similar signals were recorded with cloth and PEM electrodes. Cloth electrodes showed more variability between electrodes of the same type. CONCLUSION: For EDA recording, cloth electrodes can perform at least as well as PEM electrodes, making it practical to take advantage of the cloth electrodes' flexibility and lower profile.

Usefulness of dobutamine stress echocardiography in detecting coronary artery disease in end-stage renal disease.

The cardiovascular evaluation of patients with end-stage renal disease (ESRD) has been hampered by the suboptimal sensitivity and specificity of currently employed diagnostic tests. Dobutamine stress echocardiography (DSE) is a recently developed technique which is accurate for the diagnosis of coronary artery disease (CAD) in general populations. The purpose of this study was to assess its diagnostic accuracy and prognostic implications in patients with ESRD. Patients with ESRD (n = 97) underwent DSE as part of a preoperative evaluation before being listed for renal transplantation. Patients were followed for 12 +/- 6 months (range 1 to 24) after the study. Rest and dobutamine stress echocardiograms were analyzed for regional and global function. Coronary angiography was performed in 30 patients, and 25 underwent renal transplantation in the follow-up period. DSE had a sensitivity of 95% (92% for 1-vessel, 100% for > or = 2-vessel disease), specificity of 86%, and accuracy of 90% for the detection of CAD. During the follow-up period, 6 patients died; DSE revealed inducible ischemia in 4, and catheterization before death revealed multivessel CAD in 2. Conversely, a normal DSE identified a very low risk population, with a 97% probability of being free of cardiac complications or death during the follow-up period. We conclude that DSE accurately identifies CAD in patients with ESRD and identifies a cohort of patients at low risk for cardiac complications.

[Echocardiographic diagnosis of hematoma compressing the right atrium in post-operative heart surgery].

This is a report of a patient presenting with a hematoma compressing the right atrium in the immediate post-operative period and the diagnosis by echocardiography allowed prompt surgical treatment with reversibility of the hemodynamic failure. Echocardiography is an important tool in the diagnosis of post-operative complications and transesophageal echocardiography can be very helpful in this setting, where technically difficult transthoracic exams are the rule.

Standardization of M-mode echocardiographic left ventricular anatomic measurements.

To improve standardization of echocardiographic left ventricular anatomic measurements, echographic left ventricular dimensions and mass were related to body size indexes, sex, age and blood pressure. Independent normal populations comprised 92 hospital-based subjects (64 women, 28 men) and 133 subjects from a population sample (55 women, 78 men). All measurements of chamber size, wall thickness and mass differed between men and women in both series (p less than 0.01 to p less than 0.001). Left ventricular mass was related most closely to body surface area among measurements of body size (r = 0.37, p less than 0.01 to r = 0.57, p less than 0.001) in all four groups. Indexation by body surface area eliminated sex differences in wall thicknesses and internal dimension, but a significant sex difference in left ventricular mass index persisted (89 +/- 21 g/m2 in men versus 69 + 19 g/m2 in women in the entire series, p less than 0.0001). The 97th percentile of left ventricular mass index was identical in both groups of men (136 and 132 g/m2) and women (112 and 109 g/m2). A highly significant difference in lean body mass, estimated from 24 hour urine creatine excretion, was observed between men and women (58 +/- 15 versus 40 +/- 13 kg, p less than 0.001) and no sex difference existed in left ventricular mass indexed by lean body mass (3.4 +/- 1.3 versus 3.5 +/- 1.5 g/kg). Weak correlations were observed between left ventricular mass/lean body mass and systolic or diastolic blood pressure (r = 0.25, p less than 0.05 and r = 0.28, p less than 0.01, respectively) but not age (18 to 72 years).(ABSTRACT TRUNCATED AT 250 WORDS)

[Echography in posterior scleritis (author's transl)].

Following a brief clinical description of posterior scleritis its echographical criteria are described with reference to a case history: broad scleral echos with high reflectivity and poor sound attenuation in the A-scan are characteristic, as well as clear demarcation of the thickened posterior sclera from the orbital tissue and edema in Tenon's space in the B-scan.

[Ultrasonography of orbital rhabdomyosarcoma in children (author's transl)].

We report on 4 children suffering from orbital rhabdomyosarcoma. The ultrasonographical findings in both A and B mode are: irregular structure, poorly outlined borders, very low reflectivity, weak sound attenuation, hard consistency and little mobility of the tumor. There was no correlation between the ultrasonographical findings and the histological type of rhabdomyosarcoma.

[B-scan echography in eye and orbital diseases].

After a short description of ultrasound A- and B-scan-techniques, the ultrasonic findings in the different diseases of the eye and orbit are discussed. The results, as presented in this paper, show that ultrasound proves to be a useful aid in the evaluation and differential diagnosis of ocular pathology.

Increased cardiac performance in mild essential hypertension. Left ventricular mechanics.

To analyze cardiovascular dynamics in essential hypertension, 81 subjects with untreated mild essential hypertension (mean blood pressure, 112 +/- 14 mm Hg) and 87 normotensive subjects from the same working population were studied by echocardiography and simultaneous blood pressure determination. Hypertensive subjects had significantly higher pulse pressure, stroke volume index, cardiac index, left ventricular internal dimension, end-systolic pressure/volume ratio, end-systolic stress, left ventricular mass index, and relative wall thickness than normotensive subjects. Among both normotensive and hypertensive subjects, cardiac performance was closely dependent on afterload, as indicated by close inverse linear relationships between left ventricular fractional shortening and log end-systolic stress (r = -0.83 and -0.78 respectively; both, p less than 0.001). However, 19 of 81 hypertensive patients (23%) fell above the 95% confidence limits of this relationship in normotensive subjects (p less than 0.001 compared with that in normotensive subjects), with a bimodal distribution of fractional shortening as a percent of predicted in relation to end-systolic stress among patients with essential hypertension. This subgroup of hypertensive subjects, with increased resting cardiac performance independent of afterload, was similar in age to the remaining hypertensive subjects but had higher fractional shortening (41 +/- 5% vs 35 +/- 7%; p less than 0.001) and cardiac index (4.3 +/- 1.3 L/min/m2 vs 3.4 +/- 1.0 L/min/m2; p less than 0.005) and lower total peripheral resistance (1257 +/- 502 dyn sec cm-5 vs 1582 +/- 584 dyn sec cm-5 p less than 0.05) and left ventricular relative wall thickness (0.34 +/- 0.06 vs 0.42 +/- 0.10; p less than 0.005). Thus, analysis of cardiac mechanics detected a subset of patients with essential hypertension in whom increased cardiac function cannot be attributed either to relative youth or to supercompensatory left ventricular hypertrophy.

Neuronal NT-3 is not required for synaptic transmission or long-term potentiation in area CA1 of the adult rat hippocampus.

Neurotrophic factors, including BDNF and NT-3, have been implicated in the regulation of synaptic transmission and plasticity. Previous attempts to analyze synaptic transmission and plasticity in mice lacking the NT-3 gene have been hampered by the early death of the NT-3 homozygous knockout animals. We have bypassed this problem by examining synaptic transmission in mice in which the NT-3 gene is deleted in neurons later in development, by crossing animals expressing the CRE recombinase driven by the synapsin I promoter to animals in which the NT-3 gene is floxed. We conducted blind field potential recordings at the Schaffer collateral-CA1 synapse in hippocampal slices from homozygous knockout and wild-type mice. We examined the following indices of synaptic transmission: (1) input-output relationship; (2) paired-pulse facilitation; (3) post-tetanic potentiation; and (4) long-term potentiation: induced by two different protocols: (a) two trains of 100-Hz stimulation and (b) theta burst stimulation. We found no difference between the knockout and wild-type mice in any of the above measurements. These results suggest that neuronal NT-3 does not play an essential role in normal synaptic transmission and some forms of plasticity in the mouse hippocampus.
