[Analysis of monoclonal protein in 72 cases of multiple myeloma].

Serum and urinary monoclonal proteins (M protein) were measured in 72 cases of multiple myeloma (MM) using rate nephelometry. In IgG and IgA types of MM, the level of immunoglobulin (Ig) corresponding to the malignant isotype was significantly higher and that of Ig uncorresponding to the malignant isotype lower than the normal level. The light chain corresponding to the malignant isotype in serum was increased and the light chain uncorresponding to that in serum was decreased either kappa-IgG, IgA types or lambda-IgG, IgA types. Either kappa-LC or lambda-LC type of MM, the serum light chain corresponding to the malignant isotype was in the normal range and uncorresponding to that was decreased, and the corresponding light chain in urine was significantly elevated. Kappa/lambda ratio in serum and urine was all significantly abnormal in IgG, IgA, and LC types of MM. Our data suggest that any quota among kappa light chain > 20 g.L-1 or < 5 g.L-1, lambda light chain > 10 g.L-1 or < 2 g.L-1 in serum and kappa/lambda ratio > 5 or < 0.75 in serum and urine has an important value for diagnosing MM.

[The cytogenetical studies of fertility restoration of some male sterile lines of alloplasmic wheat].

The frequency of cells with univalent (FCU) at MI and with lagging chromosomes and chromosomal bridge (FCLB) at AI in PMC of F1 hybrids of 4 alloplasmic 1BL:/1RS and non-1BL/1RS wheat male sterile lines with Aegilops kotschyi, Ae. variabilis, Ae. ventricosa and Ae. bicornis cytoplasms were systematically investigated, and the relationship between FCU at MI, FCLB at AI and F1 selfed seed set was analyzed. The results were as follows: (1) The frequency of abnormal chromosomes at MI and AI was higher in 1BL/1RS hybrids than in non-1BL/1RS hybrids; (2) 4 alien cytoplasms had positive effects on the FCU at MI in non-1BL/1RS hybrids; (3) The effect of 1B.1BL/1RS hetero-nucleus on meiosis was more obvious than that of alien cytoplasms in 1BL/1RS hybrids; (4) The selfed seed set of 1BL/1RS hybrids was not correlated to FCU, but was negatively correlated to FCLB; (5) The chromosomes' deeds at meiosis was more steady in non-1BL/1RS hybrids than in 1BL/1RS hybrids. The non-1BL/RS hybrids were easy to restore and restoring degree was high, and they had good prospects to be used.

[The vitronectin anchors on placenta tissue of hypertensive pregnancy].

Using immunohistochemistry staining and Western-blot techniques, we detected the distribution and content of vitronectin antigen in placenta tissue of one case of hypertensive pregnancy in order to explore pathogenesis of the development of hypertensive pregnancy. The results were that vitronectin expressions on hypertensive pregnant placenta were obviously higher than those on normal placenta, which suggests that fibrin deposition on placenta tissue was associated with suppression of fibrinolysis.

[The features and significance of immunophenotyping in adult acute leukemia].

Immunophenotypic assay was determined by using indirect immunofluorescent method in 120 adult patients with acute leukemia. Eight monoclonal CD antibodies consisting of anti-CD2, -CD7, -CD10, -CD19, -CD13, -CD33, -CD14, and anti-HLA-DR monoclonal antibodies were applied conventionally. The results were that immunophenotyping had high corresponding rate to FAB morphology typing in diagnosing acute leukemia. Also there were some patients who had cross or inverse CD marker expression or no specific CD antigen expression. There were some immunophenotypic features in AML-M3, ALL-L3, and B-Lineage and T-Lineage acute lymphocytic leukemia respectively. Prognostic significance was observed in the patients with specific immunophenotypes, that is acute myeloid leukemia patients with lymphoid antigen (Ly+ AML) and some cases with inverse immunophenotypes.

[Effects of rhGM-CSF and rhG-CSF on growth of CFU-F derived from acute leukemia (AL) patient bone marrow in vitro].

By using the cultural method of CFU-F derived from bone marrow in the patient with acute leukemia in vitro, we dissected the effects of rhGM-CSF and rhG-CSF on growth of CFU-F derived from AL patient bone marrow. The results showed that rhGM-CSF and rhG-CSF enhanced the number of colonies of CFU-F and its proliferation activity. Both factors had a synergy on the above effects. The effects of rhGM-CSF on CFU-F were significantly greater than that of rhG-CSF. The data indicate that rhGM-CSF and rhG-CSF might regulate hemopoiesis more effectively by stimulating the growth of fibroblast and improving bone marrow microenvironment.

[Frequency of prothrombin gene G20210A variant in the 3'-untranslated region in Chinese people with ischemic stroke].

OBJECTIVE: To ascertain the frequency of prothrombin(F II) gene 3'-untranslated region(3'-UT) G20210A variant and to explore whether this mutation is related to arterial thrombosis in Chinese people. METHODS: We studied 49 patients with thrombosis and 46 healthy Chinese persons. The patients had a first ischemic stroke. The G20210A mutant allele of the prothrombin gene in all blood specimens was investigated by DNA extraction, polymerase chain reaction amplification, Hind III digestion and polyacrylamide gel electrophoresis (PAGE). RESULTS: The patients and normal control subjects were all homozygous for the normal G20210G allele, and there was no any F II G20210A variant. CONCLUSION: Factor II gene 3'-UT G20210A mutant allele is absent in Chinese patients with ischemic stroke and normal subjects; its mutation may not be a major risk factor for thrombogenesis in Chinese people.

[Studies on identification of circumsporozoite protein genotyping of Plasmodium vivax].

OBJECTIVE: To develop a new method of genotyping circumsporozoite protein (CSP) gene for identification of field isolates of Plasmodium vivax. METHODS: Improved Chelex-100 ion-exchange method was used to extract DNA from blood filter paper samples, nested PCR and allele-specific PCR techniques, agarose gel electrophoresis analysis and dot/southern blotting-probe hybridization were employed for amplification, resolution and identification of the diagnostic fragments. RESULTS: Using the nest-allele-specific PCR assay reported here, small amounts of DNA extracted from a piece of blood filter paper sample were amplified which produced three different size ranges of diagnostic bands: 650-770 bp PV-species-specific band, 170-230 bp diagnostic band for temperate zone family and 588 bp band for PV type-2. The sizes and patterns of the bands produced by the reference strains were consistent with those of designed target sequences. Of 59 examined isolates from 6 provinces of China, 42 temperate zone family strains, 15 tropical zone family strains and two PV type-2 strains were identified. CONCLUSION: 1, Three genotype strains of P. vivax mentioned above could be identified by this method with only two rounds of PCR and without probe hybridization. 2, The preliminary results showed that PV type-1 including temperate zone family strains and tropical zone family strains as well as PV type-2 strains are present in China. In addition, another CSP genotype with both sequence characteristics of temperate zone and tropical zone family might also be present in China.

The influence of surgical sites on early postoperative hypoxemia in adults undergoing elective surgery.

UNLABELLED: To determine the influence of the surgical sites on early postoperative hypoxemia, we studied postoperative hypoxemia in 994 patients, ASA physical status I or II, aged 18-68 yr, scheduled for various types of elective surgery. Patients were divided into three groups on the basis of the surgical sites: Group 1 = elective superficial plastic surgery (n = 288); Group 2 = upper abdominal surgery (n = 452); and Group 3 = thoracoabdominal surgery (n = 254). Anesthesia was maintained with 1%-2% enflurane and 67% nitrous oxide in oxygen; thiopental or fentanyl was given IV as required. SpO2 levels were recorded while patients breathed room air shortly after arrival in the recovery room (0 min) and 5, 10, 15, 20, 30, 40, 50, 60, 120, and 180 min thereafter. The results showed that during the early postoperative period, the degree of arterial desaturation and the incidences of hypoxemia (SpO2 86%-90%) and severe hypoxemia (SpO2 85%) were closely related to the operative sites and were greatest for thoracoabdominal operations, less for the upper abdominal operation, and least for the peripheral surgery. The incidence of hypoxemia and severe hypoxemia in the recovery room was 7% and 0.7%, respectively, in Group 1, 38% and 3% in Group 2, and 52% and 20% in Group 3. Mild airway obstruction and hypothermia in the postanesthesia recovery unit (PAR) were the predictive factors of early postoperative hypoxemia. We conclude that during the early postoperative period, there were significant differences in SpO2 levels and incidences of hypoxemia and severe hypoxemia among the three groups. IMPLICATIONS: We found that the severity of arterial desaturation and the incidence of hypoxemia during the early postoperative period are closely related to the surgical sites and are strongest for thoracoabdominal surgery, less for upper abdominal surgery, and least for peripheral surgery.

[Effects of pentagastrin with intracerebroventricular injection on blood pressure and heart rate in rats].

Experiments were carried out on 84 artifially ventilated SD rats anaesthetized with urethane and immobilized with gallamine triethiodide. Injection of pentagastrin (G5) into the rat lateral ventricle resulted in a marked elevation in arterial blood pressure which reached its peak at 3 min (from 12.2 +/- 1.5 to 14.1 +/- 1.7 kPa) and also a significant increase in heart rate from 435 +/- 53.9 to 471.6 +/- 53.2 beats/min. This effect of G5 could be partially antagonized by injection of phentolamine (alpha-receptor antagonist, 10 micrograms) into the lateral ventricle, but not by propanolol (beta-receptor antagonist, 4 micrograms) or atropine (M-receptor antagonist, 4 micrograms). These findings indicate that the central effect of G5 on blood pressure and heart rate is partly mediated by alpha-receptor.

Murine heparin cofactor II: purification, cDNA sequence, expression, and gene structure.

Heparin cofactor II (HCII) is a glycoprotein in human plasma that inhibits thrombin rapidly in the presence of dermatan sulfate or heparin. Unexpectedly, we found that HCII activity in murine plasma is present in two proteins of 68 and 72 kDa. The two proteins have the same N-terminal amino acid sequence, and both react with an antibody raised against the C-terminal nine amino acid residues of murine HCII predicted from the cDNA sequence. Treatment of the two proteins with peptide-N4-(N-acetyl-beta-glucosaminyl)asparagine amidase yields a single 54-kDa band. Thus, murine plasma contains two forms of HCII that appear to have identical amino acid sequences but differ in the composition of their N-linked oligosaccharides. HCII cDNA clones isolated from a murine liver library include a 1434 bp open reading frame following the first Met codon, a TAA stop codon, and 580 bp of 3'-untranslated sequence terminating in a poly(A) tail. The amino acid sequence deduced from the cDNA contains the N-terminal sequence of purified murine plasma HCII preceded by a 23-residue hydrophobic sequence presumed to be the signal peptide. The amino acid sequence of murine HCII is 87% identical to that of human HCII, the greatest variability occurring in the N-terminal portion of the protein. Northern blot analysis reveals a 2.3-kb HCII mRNA in murine and human liver, but no HCII mRNA is detectable in heart, brain, spleen, lung, skeletal muscle, kidney, testis, placenta, pancreas, or intestine. Southern blot analysis of restriction fragment length polymorphisms in progeny on interspecific and intersubspecific crosses indicates that mice have a single HCII gene (designated Hcf2), which maps to chromosome 16 between Prm-1 and Igl. The murine HCII gene is approximately 7.1 kb in size and consists of at least four exons and three introns. The intron/exon organization is identical to that of the human HCII gene except at the 5' end, where the murine gene may lack a large intron in the 5'-untranslated region. Our results indicate that HCII is more highly conserved than the human and murine homologues of other serpins such as alpha 1-antitrypsin and alpha 1-antichymotrypsin.

[RBC Na(+)-Li+ countertransport and Na(+)-K+ cotransport in patients with essential hypertension].

The correlation analyses were performed in 60 patients with essential hypertension (EHT) and 77 normotensives (NT) between RBC Na(+)-Li(+)-countertransport (SLC), Na(+)-K+ cotransport (SPT) and blood pressure, Quetelet index (QI), serum insulin, blood lipids, uric acid, blood glucose, etc. Results showed: (1) The SLC in EHT patients were higher, while SPT lower than NT group; (2) The difference between EHT and NT group was much prominent in SLC of male, while prominent in SPT of female; (3) There was no sex difference in SLC and SPT, and the SLC was correlated positively with QI3; (4) By multiple variants stepwise regression analysis, the SLC was shown to be associated positively with systolic and diastolic blood pressure; while the SPT inversely with systolic blood pressure. It suggests that SLC and SPT exert an independent effect on blood pressure.

[Study on risk factors involving train traffic accidents--effects of human factors on accidents].

This article analysed human factors involving train traffic accidents by using log-linear model. The results showed that lower degree of culture (OR = 2.571) and weak nervous type (OR = 2.099) were risk factors involving train traffic accidents. Interactions were shown to exist between nervous type and degree of culture, average sleeping time everyday, and visuomotor reaction time.

Experimental studies of the injurious effects of Q-switched Nd:YAG lasers and their outdoor applications.

Comparison of the dose-response relationship of retinal damage induced by Q-switched (Q-sw) Nd:YAG lasers was conducted for rabbits and monkeys. Experimental results indicated that the two probit regression lines were parallel to each other, and the damage threshold (ED50) ratio was approximately 1:3.57. Observations of the injurious effects of Q-sw Nd:YAG rangefinders--the total energy level being about 10-100 mJ--and their injurious distances are reported in this paper. A brief description is given of coagulative and hemorrhagic pathological changes under different exposure conditions and at various distances.

[Clinical and experimental observations of jinqiancao gao on preventive and therapeutic effects in cholelithiasis].

This paper reports the clinical treatment of 120 patients with cholelithiasis for 2 months with Jinquiancao Gao. The Results showed that the clinical effective rate was 92.5%, among which 29 cases were cured, 42 cases markedly improved and 40 cases ineffective. Animal experiments showed that this medicine can prevent gallstone growth. The gallstone growth rate was 78.5% in the control group and 42.8% in the experimental group (P less than 0.05). The mechanism of the decrease of gallstone growth rate suggests that the said medicine could inhibit the activity of beta-Glucuronidase in the bile, diminish the non-combined bilirubin and prevent the bile from forming bilirubin stones. The result of cultured liver cells in vitro showed that the cholesterol synthesis was markedly inhibited by Jinqiancao Gao. Thus the cholesterol concentration of bile was decreased. These effects in turn prevented the bile from forming cholesterol stones.
