Microdontin A and B: Two New Aloin Derivatives from Aloe microdonta.

The leaf juice of ALOE MICRODONTA Chiov. is used in Somali traditional medicine as a remedy for jaundice and for topical treatment of skin diseases. Mucilage was precipitated from the fresh juice by addition of ethanol and the supernatant chromatographed on Amberlite XAD-2 to yield a fraction containing anthraquinone derivatives. Flash chromatography on silica gel followed by preparative TLC yielded aloin A and a new compound. Spectral data (mass, (1)H- and (13)C-NMR) identified this compound as a mixture of aloin A and B where the glucose of both compounds is esterified with 4-hydroxycinnamic acid at the 6'-position. The two compounds were denoted as microdontin A and B, respectively.

Bioassay-guided isolation of serotonin from fruits of Solanum tuberosum L.

An ethanol extract of the fruits of Solanum tuberosum was partitioned between water and petroleum ether. The aqueous fraction had contractive effect on the isolated guinea-pig ileum. Chromatography of this fraction on Sephadex LH-20 and silica gel was monitored by the guinea-pig ileum test and resulted in isolation of serotonin. HPLC analysis of the content of serotonin in fruits, leaves and tubers of the plant, showed that the fruits and leaves contained 7.5 micrograms and 2 micrograms, respectively, of serotonin/g fresh weight. In agreement with previous observations, no serotonin was detected in the tubers.

[Dementia is not the only mental problem among the elderly. Long-term perspective is important for care].

The entire 1902 and 1903 birthyear cohorts in the catchment area of Dalby Community Health Centre in southern Sweden have been followed with health examinations and interviews every second year since 1969-70. Of the total of 192 subjects, complete data are available for 153. Of 65 subjects surviving at the age of 83, 6 per cent were advanced dementia cases, 11 per cent were in the early stages of dementia, and a further 17 per cent were afflicted with other mental problems requiring help and care. Of those who died before the age of 83, 11 per cent were afflicted with dementia, with a further 4 per cent in the early stages, and 17 per cent had other mental problems. Thus, the overall risk of dementia is at least 16 per cent, but may eventually turn out to be as high as 30 per cent, while those with other forms of mental dysfunction, serious enough to require help in ADL, account for a similar proportion. The annual incidence of dementia is 7.5 per mill (10.6 per mill including those in early stages). The risk of institutionalisation and death was much higher among the dementia victims, most of whom had had little support from the social services, and some none at all, before being institutionalised. Dementia victims came into contact with the medical services much earlier than did mentally healthy patients with other, often vague and more varied symptoms. It is argued in the article that current concern with dementia should not be allowed to divert our attention from the needs, diagnosis and care of the many elderly patients with other mental problems.

Inventory of plants used in traditional medicine in Somalia. I. Plants of the families Acanthaceae-Chenopodiaceae.

Thirty-eight plants are listed, which are used by traditional healers in the central and southern parts of Somalia. For each species are listed: the botanical name with synonyms, collection number, vernacular name, medicinal use, preparation of remedy and dosage. Results of a literature survey are also reported including medicinal use, substances isolated and pharmacological effects.

T-cadinol: a pharmacologically active constituent of scented myrrh: introductory pharmacological characterization and high field 1H- and 13C-NMR data.

Fractionation of an ethyl acetate extract of scented myrrh (resin of Commiphora guidottii Chiov., Burseraceae), using the guinea pig ileum test to monitor pharmacological activity, resulted in isolation of the sesquiterpene (+)-T-cadinol. High field NMR spectroscopy yielded new detailed 1H- and 13C-NMR data for the compound. T-cadinol was shown to have a concentration-dependent smooth muscle relaxing effect on the isolated guinea pig ileum and a dose-dependent inhibitory effect on cholera toxin-induced intestinal hypersecretion in mice.

Stability and change of personality in old age and its relation to survival.

Stability of personality characteristics with increasing age was analyzed in a Swedish longitudinal study using a modified version of the Rod-and-Frame Test. This test was analyzed in a serial manner in order to emphasize cognitive style. Two important findings emerged. First, during the 6 years between the ages of 67 and 73, 80% of the study participants showed stability in the field independence-dependence dimension. Second, stability of the field independence-dependence trait between ages 67 and 73 predicted survival to age 83, whereas destabilization (i.e., changes of Rod-and-Frame Test classification) was significantly associated with earlier death. In most cases, destabilization meant that field-independent subjects became field dependent. The results are discussed in relation to dispositional and situational factors. Causative factors for destabilization are hypothesized.

Acclimation of Photosynthetic Light Reactions during Induction of Inorganic Carbon Accumulation in the Green Alga Chlamydomonas reinhardtii.

Cells of the unicellular green algae Chlamydomonas reinhardtii were grown in high dissolved inorganic carbon (DIC) concentrations (supplied with 50 milliliters per liter CO(2)[g]) and transferred to low DIC concentrations (supplied with </= 100 microliters per liter CO(2)[g]). Immediately after transfer from high to low DIC the emission of photosystem II related chlorophyll a fluorescence was substantially quenched. It is hypothesized that the suddenly induced inorganic carbon limitation of photosynthesis resulted in a phosphorylation of LHCII, leading to the subsequent state 1 to state 2 transition. After 2 hours of low-DIC acclimation, 77 K fluorescence measurements revealed an increase in the fluorescence emitted from photosystem I, due to direct excitation, suggesting a change in photosystem II/photosystem I stoichiometry or an increased light harvesting capacity of photosystem I. After 5 to 6 hours of acclimation a considerable increase in spillover from photosystem II to photosystem I was observed. These adjustments of the photosynthetic light reactions reached steady-state after about 12 hours of low DIC treatment. The quencher of fluorescence could be removed by 5 minutes of dark treatment followed by 5 minutes of weak light treatment, of any of four different light qualities. It is hypothesized that this restoration of fluorescence was due to a state 2 to state 1 transition in low-DIC acclimated cells. A decreased ratio of violaxanthin to zeaxanthin was also observed in 12 hour low DIC treated cells, compared with high DIC grown cells. This ratio was not coupled to the level of fluorescence quenching. The role of different processes during the induction of a DIC accumulating mechanism is discussed.

Smooth muscle relaxing flavonoids from Alchornea cordifolia.

Alchornea cordifolia (Schum. & Thonn.) Muell. Arg. (Euphobiaceae) is widely distributed throughout tropical Africa, where it is used extensively in traditional medicine. Conditions for which the plant has enjoyed wide use are: coughs, gonorrhoea, yaws, ulcer, rheumatic pains, fever and bronchial troubles. This paper reports isolation of smooth muscle relaxing flavonoids from the leaves of the plant.

Immunoglobulin allotypes are normally distributed in Swedish AIDS patients.

Immunoglobulin allotypes G1m(a), G1m(x), G3m(b) and Km(1) were determined in 83 Swedish AIDS patients. Twenty of the patients had Kaposi's sarcoma and 29 had Pneumocystis carinii pneumonia. The distribution of the Gm and Km allotypes did not significantly differ between these different disease categories and the general Swedish population.

Induction of Inorganic Carbon Accumulation in the Unicellular Green Algae Scenedesmus obliquus and Chlamydomonas reinhardtii.

The induction of a dissolved inorganic carbon (DIC) accumulating mechanism in the two algal species Scenedesmus obliquus (WT) and Chlamydomonas reinhardtii (137 c+) was physiologically characterized by monitoring DIC uptake kinetics at a low and constant DIC concentration (120-140 micromolar), after transfer from high-DIC culturing conditions. A potentiometric titration method was used to measure and calculate algal DIC uptake. Full acclimation to low-DIC conditions was obtained within a period of 90 min, after which time the DIC uptake had been increased 7 to 10 times. Experiments were also conducted in the presence of inhibitors against DIC accumulation. The inhibitor of extracellular carbonic anhydrase (CA), acetazolamide (50 micromolar), inhibited the adaptation partly, while the inhibitor of both extra- and intracellular CA, ethoxyzolamide (50 micromolar) totally inhibited the acclimation. Cycloheximide (10 micrograms per milliliter), which inhibits protein synthesis on cytoplasmic ribosomes, and vanadate (180 micromolar), which inhibits the plasmamembrane bound ATPase, also inhibited the acclimation totally. These results taken together suggest that the algae are dependent on intracellular CA, plasmamembrane bound ATPase, and de novo protein synthesis for DIC accumulation. Also, these components are more important than extracellular CA for the overall function of the DIC-accumulating mechanism.

Ending one's life in a nursing home: a note on Swedish findings.

Statistics on place of death, validated against longitudinal evidence on the entrance into nursing homes, show the "final" rate of institutionalization to have risen in the decades between 1938 and 1975 in Sweden. In a local study, 15 percent entered a nursing home between the ages of sixty-seven and eighty, which seems representative of national trends. The "final" rate is estimated to be between 22 percent and a maximum of about 40 percent. Currently, the socially and economically deprived dominate in Swedish nursing homes just as they did in the poor house of the past. Yet, rising rates may indicate raised welfare for the poorest in society. Issues concerning who is institutionalized and why appear more important than precise measurement of rates of institutionalization.

Proton magnetic resonance characterization of phoratoxins and homologous proteins related to crambin.

The mistletoe protein toxins ligatoxin, phoratoxins A and B, and viscotoxins A3 and B have been investigated by 1H NMR spectroscopy at 300 and 600 MHz. The five polypeptides define a set of closely related homologues, containing 46 amino acid residues each, in a structure constrained by three cystine bridges. Their methyl and aromatic spectra were analyzed and a number of signals identified and assigned via comparative criteria, two-dimensional chemical-shift correlated spectroscopy, acid-base titration, and proton Overhauser experiments in 1H2O. The spectra indicate a compact globular conformation and a common folding pattern for the toxins. In particular, use was made of well-resolved aliphatic and aromatic resonances in order to compare the mistletoe proteins with the thionins, a set of homologous toxins from gramineae, and with crambin, a closely related polypeptide from a crucifer, which we have previously studied by NMR. We observe that while all the investigated proteins have very similar secondary and tertiary structures, they differ widely in their dynamic characteristics as probed by the amide NH 1H-2H exchange kinetics in deuteriated solvents; thus, while crambin and the thionins exhibit very fast isotope exchange, the kinetics for the mistletoe toxins are slow, with some NH groups showing exchange half-lives that extend up to several days at pH* 5.8 or that are too long to be measurable at ambient temperature. The temperature dependence of the 1H NMR spectrum also indicates that the toxins are endowed with a thermally very stable native (ground-state) structure, with little evidence of large amplitude structural breathings up to approximately 370 K, although irreversible chemical degradation (denaturation) becomes evident at temperatures greater than or equal to 350 K. It is concluded that the mistletoe toxins afford valuable rigid structures for NMR conformational studies.

Elderly handicapped persons in a primary care district: changes during a 5-8 year follow-up.

643 handicapped persons born in 1918 or earlier were found at an inventory in 1975-78. In a follow-up 5-8 years later the mortality was 46%, which was significantly higher compared to the general mortality in the same age groups. There was no relationship between social class and mortality. Based on data from the first study, the deceased showed, compared to the surviving, impaired ADL and they used more medical and social care. The surviving population showed impairments in primary and secondary ADL during the follow-up. Elderly handicapped persons constitute a group at risk as they have an increased mortality and a great need of medical and social care. They should therefore be known to the medical profession.

The Susceptibility of Photosynthesis to Photoinhibition and the Capacity of Recovery in High and Low Light Grown Cyanobacteria, Anacystis nidulans.

The susceptibility of photosynthesis to photoinhibition and the rate of its recovery were studied in the cyanobacterium Anacystis nidulans grown at a low (10 micromoles per square meter per second) and a high (120 micromoles per square meter per second) photosynthetically active radiation. The rate of light limited photosynthetic O(2) evolution was measured to determine levels of photoinhibition and rates of recovery. Studies of photoinhibition and recovery with and without the translation inhibitor streptomycin demonstrated the importance of a recovery process for the susceptibility of photosynthesis to photoinhibition. We concluded that the approximately 3 times lower susceptibility to photoinhibition of high light than of low light grown cells, significantly depended on high light grown cells having an approximately 3 times higher recovery capacity than low light grown cells. It is suggested that these differences in susceptibility to photoinhibition and recovery depends on high light grown cells having a higher turnover rate of photosystem II protein(s) that is(are) the primary site(s) of photodamage, than have low light grown cells. Furthermore, we demonstrated that photoinhibition of A. nidulans may occur under physiological light conditions without visible harm to the growth of the cell culture. The results give support for the hypotheses that the net photoinhibitory damage of photosystem II results from the balance between the photoinhibitory process and the operation of a recovery process; the capacity of the latter determining significant differences in the susceptibility of photosynthesis to photoinhibition of high and low light grown A. nidulans.
