Gastrodia anti-fungal protein from the orchid Gastrodia elata confers disease resistance to root pathogens in transgenic tobacco.

Diseases of agricultural crops are caused by pathogens from several higher-order phylogenetic lineages including fungi, straminipila, eubacteria, and metazoa. These pathogens are commonly managed with pesticides due to the lack of broad-spectrum host resistance. Gastrodia anti-fungal protein (GAFP; gastrodianin) may provide a level of broad-spectrum resistance due to its documented anti-fungal activity in vitro and structural similarity to insecticidal lectins. We transformed tobacco (Nicotiana tabacum cv. Wisconsin 38) with GAFP-1 and challenged transformants with agriculturally important plant pathogens from several higher-order lineages including Rhizoctonia solani (fungus), Phytophthora nicotianae (straminipile), Ralstonia solanacearum (eubacterium), and Meloidogyne incognita (metazoan). Quantitative real-time PCR and western blotting analysis indicated that GAFP-1 was transcribed and translated in transgenic lines. When challenged by R. solani and P. nicotianae, GAFP-1 expressing lines had reduced symptom development and improved plant vigor compared to non-transformed and empty vector control lines. These lines also exhibited reduced root galling when challenged by M. incognita. Against R. solanacearum expression of GAFP-1 neither conferred resistance, nor exacerbated disease development. These results indicate that heterologous expression of GAFP-1 can confer enhanced resistance to a diverse set of plant pathogens and may be a good candidate gene for the development of transgenic, root-disease-resistant crops.

[The histamine provocation test: are 1 second capacity and specific conductance competing or complementary measurement values?].

We tested criteria for the decision, whether a provocation challenge with histamine is positive. The combination of spirometry and bodyplethysmography by the condition sGaw less than or equal to 0.04 x PC-FEV overcomes the contradictions due to conventional criteria as PC-FEV greater than or equal to 20% or PC-sGaw greater than or equal to 50%.

Performance and perception in a simulated clinical encounter.

The feasibility of using data arising from a simulated patient encounter to determine the nature of the relationship between patient satisfaction and examinee performance during a clinical examination is explored. Satisfaction (on dimensions of sensitivity, participation and thoroughness) is shown to be related to aspects of the physical examination and the history taking.

Immunological comparison of ovarian and colonic CEA.

Ovarian and colonic CEA were compared immunologically by means of antisera prepared against each of them. CEAs of both origins were found identical by immunodiffusion methods. In radioimmunological experiments, slight differences were observed between some but not all ovarian CEAs and colonic CEAs and also between different preparations of colonic CEA: no organ specificity of ovarian CEA could be demonstrated. Finally, CEA level was measured in 41 sera of patients with ovarian carcinoma by two radioimmunoassays, one using colonic CEA as tracer and standard and anti-colonic CEA serum, the other using ovarian CEA and anti-ovarian CEA serum: the values given by the two assays were highly correlated (rs = 0.8107), meaning that an organ specific assay for ovarian CEA is not needed.

[Stability of liquid ergotalkaloid preparations as a function of the solvent polarity (author's transl)].

The degradation of dihydroergotamine-, dihydroergocornine-, dihydroergocryptine- (both isomers) and dihydroergocristine-mesilate in solution was studied as a function of the temperature and the dielectric constant of the composition. The degradation reactions of the studied hydrogenated ergopeptide alkaloids gave pseudo first-order kinetics. The pseudo first-order rate constants at room temperature were calculated using the Arrhenius equation. It was found that the stability of the studied hydrogenated ergopeptide alkaloids in solution is a function of the dielectric constant of the composition, and that stable solutions can be obtained by the use of water-alcohol mixtures having dielectric constants between 30 and 45. Solutions of the hydrogenated ergot alkaloids in predominantly organic media have the advantage that the degradation to the corresponding aci-forms, hydrolysis products and oxidation products is reduced. For this reason it is not necessary to carry out the filling operation under an inert gas atmosphere in order to protect the hydrogenated ergot alkaloids from oxidation. These results present an important technological advance.

Determination of dexamethasone in milk by high pressure liquid chromatography.

A rapid and sensitive high pressure liquid chromatographic method for the analysis of dexamethasone (9 alpha - fluoro - 11 beta, 17 alpha, 21 - trihydroxy - 16 alpha - methylpregna - 1,4-diene-3,20-dione) in milk has been developed. The corticosteroid can be quantitated at 20 ppb, with the limit of detectability estimated at 5 ppb. Recoveries ranged from 72 to 94%. After dexamethasone was injected into 5 Holstein-Friesian cows, no residues were dound in milk, even after the first milking.
