Isolation and characterization of cDNAs encoding viscotoxins of mistletoe (Viscum album).

Viscotoxins have been isolated from leaf homogenates of European mistletoe (Viscum album L.) and purified to apparent homogeneity. Antisera raised against these polypeptides were used to screen a cDNA expression library in lambda gt11. Two positive clones have been isolated, one encoding a full-length preprotein of viscotoxin A3 and the other encoding the precursor of viscotoxin B. Besides the viscotoxin domain the precursor contained a signal sequence and an acidic polypeptide domain. Similar higher molecular mass precursor proteins have been described for thionins of leaves and seeds of barley. Even though the acidic part of the viscotoxin precursor is much shorter than the corresponding domain of the precursors of the leaf and seed thionins of barley, both the negative charge and the number and the relative position of cysteine residues have been conserved within the acidic domain. This result is consistent with our proposal that the acidic domain of the thionin precursor may play an important role in keeping the thionin inactive within the plant cell.

The recollection of affect.

A group of psychiatric in-patients significantly over-estimated the intensity of their depression when asked to recollect how depressed they had been one week earlier. The initial level of depression affected the accuracy of recall. Unexpectedly, patients who were more depressed initially had more accurate recall, particularly for biological and symptomatically negative items of the Zung Self-Rating Depression Scale.

Do psychosocial factors predict weight loss following gastric surgery for obesity?

This study was undertaken to determine whether psychosocial factors determined during a pre-operative semi-structured psychiatric interview were associated either with the amount of weight loss following obesity surgery or with dropping out from follow-up after surgery. Multiple regression and discriminant function analysis of weight loss at six, twelve, twenty four and thirty six months showed no correlation between psychosocial variables and the amount of weight lost or with dropping out from follow-up. It is concluded that the type of psychiatric interview performed in this study does not yield information which reliably predicts weight loss after surgery for obesity.

Intracellular thionins of barley. A second group of leaf thionins closely related to but distinct from cell wall-bound thionins.

Leaf thionins of barley have been identified as a novel class of cell wall proteins, toxic to plant pathogenic fungi, and possibly involved in the defense mechanism of plants (Bohlmann, H., Clausen, S., Behnke, S., Giese, H., Hiller, C., Reimann-Philipp, U., Schrader, G., Barkholt, V., and Apel, K., (1988) EMBO J. 7, 1559-1565). In the present work a second subfraction of thionins has been detected within the leaf cell, mainly in the vacuole. Thionins of both groups are closely related to each other. They are toxic to phytopathogenic fungi as well as to plant protoplasts, they share similar amino acid sequences, and their synthesis in etiolated seedlings of barley is down-regulated by light. Despite these similarities each of the two subfractions of thionins could be clearly distinguished by its subcellular distribution. In ultrathin sections of embedded etiolated leaf material, cell wall thionins could be immunogold labeled specifically by an antiserum raised against a fusion protein of Escherichia coli beta-galactosidase and the 15,000 Mr precursor polypeptide of thionins. This antiserum did not react with intracellular thionins. Inversely, intracellular thionins were recognized specifically by an anti-serum raised against soluble leaf thionins. The possible function of intracellular thionins as part of a defense mechanism has been discussed.

[Polar medical research from the viewpoint of public health].

The increasing interest in polar researches is the reason for numerous foundations of antarctic stations during the last years. In 1987 the German Democratic Republic founded a research station on the 6th continent, working all the years round. The medical polar research, hitherto done only sporadically has got a greater importance now and a working group "Polar Medicine" was established in 1989. Special hygienic problems in Antarctica and future research are discussed.

Dysfunctional attitudes in former psychiatric inpatients.

In order to test the hypothesis that patients who are predisposed to depression have an enduring cognitive style, dysfunctional attitudes (Dysfunctional Attitudes Schedule), neuroticism and extraversion (Maudsley Personality Inventory), and severity of depression (Levine-Pilowsky Depression Questionnaire) were measured in a survey of former patients with previous diagnoses of either depressive or nondepressive psychiatric conditions. We found that there were no significant differences in dysfunctional attitudes between these groups of patients and that their scores were similar to those reported for normal populations. There was, however, a correlation between introversion and high dysfunctional attitude scores. Possible implications regarding interactions between cognitive style, personality, and predisposition toward depression are discussed as well as a suggestion that a history of a suicide attempt may predict a poor response to cognitive psychotherapy.

The usefulness of DSM-III axis IV and axis V assessments.

DSM-III multiaxial diagnoses were given to 257 patients. Analysis of axis I diagnoses and axis IV and axis V assessments generally revealed the expected associations between particular psychiatric disorders and levels of psychosocial stress and social and occupational functioning. However, a major difficulty affecting the usefulness of axis IV and axis V data when they are routinely gathered according to the current DSM-III instructions is the stipulation that the clinician judge the etiological significance of a stressor before it is coded.

Medical students' attitudes towards psychiatry: a conceptual shift.

Based on the Nevid & Morrison Libertarian Mental Health Ideology Scale, the present study shows a conceptual/ideological shift in medical students' attitudes towards psychiatry after a 6-week psychiatry course. The resultant attitudes were more in line with those of teaching staff than those of the clinical team. Overall, the students developed a more positive view of psychiatry.

Narcolepsy, paranoid psychosis, major depression, and tardive dyskinesia.

This report describes a man with narcolepsy, paranoid psychosis, major depression, and tardive dyskinesia. The case illustrates the treatment difficulties such a patient presents and also raises questions about interactions between the putative neurotransmitters involved in each of these conditions. It is suggested that the presence of narcolepsy may facilitate the appearance of unwanted effects of antidepressants and neuroleptics such as psychosis and depression.
