Immunocytochemical and ultrastructural findings in a mature retroperitoneal teratoma.

Report is made of a mature retroperitoneal teratoma in a 32-year-old man. Investigation of the tumor revealed cells immunoreactive for ACTH, Met-enkephalin, beta-LPH, serotonin, FSH, BPP, S100, Neuron-specific-enolase. These cells were mainly present in the glandular epithelium, lining the cysts of the tumor. Ultrastructurally, neuro-secretory granules were demonstrated in the cytoplasm of the tumoral endocrine cells. At no time did the patient display endocrine symptoms.

Postnatal development of growth hormone and prolactin cells in male and female rat pituitary. An immunocytochemical light and electron microscopic study.

Localization and ultrastructural maturation of prolactin (PRL) and growth hormone (GH) cells were studied in pituitaries from neonatal, immature (4-6 weeks old), and adult rats (2-3 months old) by light and electron microscopic immunocytochemistry. The distribution pattern of these cells did not change with age. Both cell types were concentrated laterodorsally, with PRL cells adjacent to the intermediate lobe and GH cells nearer the center of the pars distalis. Labeling density of the immunogold reaction was highest for both hormones in immature rats. In neonatal and immature rats, one PRL cell type with granules 200 nm in diameter was present. In adult rats, two types of PRL cells were present: one containing polymorphous granules measuring about 500 nm (prevalent in female rats), the other with spherical granules about 200 nm (prevalent in male rats). No changes were detected in GH cells during maturation.

Selective complement fixation by pancreatic B-cells after binding to islet cell surface antibodies.

A two wavelength immunofluorescence system was used to demonstrate islet cell surface antibodies (ICSA) from juvenile diabetics and complement on the same viable rat islet cell. When incubated with rat islet cells at 4 C and at 37 C, ICSA specific for B-cells proved capable of binding complement. In contrast, ICSA specific for both B and non-B cells were able to fix complement on all cell types at 4 C but only on B-cells at 37 C. This study provides further evidence for the B-cell specific cytotoxic effect of ICSA in the presence of complement at physiological temperature. In addition, the ultrastructural events leading to B-cell destruction after ICSA and complement binding were studied by transmission electron microscopy (TEM).

A psychophysical investigation of the behavioural relevance of neurophysiological feature detectors (S cells).

The aim of the study was to establish whether monaural auditory stimulation (a nonretinal perceptual variable) affects the class 1 oblique effect (a behavioural manifestation of simple cells). The left or right monaural stimulus was a pure tone, 1000 Hz, 70 dB(A), delivered continuously throughout the experimental sessions. The left or right monocular stimulus was a thin red phosphorescent bar the orientation of which was manipulated. In order to determine the oblique effect differential orientation thresholds for principal meridians were compared to those for oblique orientations. The results, indicating an interaction effect of the monaural and monocular stimulation on the magnitude of the oblique effect, are a further demonstration that the oblique effect is not as simple as some theories (derived from extrapolation from neurophysiological findings) would imply.

Islet cell surface antibodies from insulin-dependent diabetics bind specifically to pancreatic B cells.

Viable rat islet cells were used to detect islet cell surface antibodies (ICSA) in the sera of diabetic and control patients. ICSA were present in almost all recent-onset insulin-dependent diabetics younger than 30 yr (15/16); their incidence in other diabetics (6/22) was also higher than in normal controls (1/18) or in patients with autoimmune thyroiditis (1/12). The varying specificity of the ICSA for the different islet cell types led to the recognition of class I sera, whose ICSA bind exclusively to B cells, class II sera, binding only to A and pancreatic polypeptide (PP) cells and class III sera, reacting with the three islet cell types but not with D cells. Most recent-onset insulin-dependent diabetics younger than 30 contained class I-ICSA, which is consistent with an autoimmune basis of their disease and with an involvement of surface antibodies in the B cell destruction. The presence of class II ICSA in three older diabetics and in one normal control raises the question whether autoimmune reactions against A and PP cells exist and are associated with a distinct entity in islet disease. It is concluded that the autoimmune form of diabetes mellitus represents a heterogeneous group, in which ICSA-positive patients can be distinguished on the basis of their ICSA-binding to one or more islet cell types. Three techniques can be used for the further identification of circulating ICSA, namely binding experiments with purified A or B cells, electron microscopical analysis of ICSA-binding islet cells purified by fluorescence-activated cell sorting, and the immunocytochemical characterization of ICSA-positive cells.

Ultrastructural aspects of asthenospermia.

In fifty cases of proven asthenospermia semen samples were examined for ultrastructural abnormalities. In 32 of the cases the sperm cells appeared either completely normal, or presented only minor abnormalities. In 15 cases severe teratospermia was observed. In the 3 remaining cases, specific and hitherto undescribed tail abnormalities were detected.

Pleasingness vs interestingness of visual stimuli with controlled complexity: their relationship to looking time as a function of exposure time.

Ss take more time to perceive interesting/displeasing stimuli than uninteresting/pleasing ones. This is consistent with the results of former experiments. However we used a different operationalization of looking time, based on binocular rivalry. Each of six stimulus pairs was presented in a stereoscope. One member of each pair was interesting but displeasing in comparison to the other member. Stimulus complexity was under control. Due to binocular rivalry Ss perceived only one pattern a time. 20 Ss were asked to indicate which pattern they actually saw by pushing two buttons. For each stimulus pair was registered how long each button was pushed during each of six successive minutes. Unlike other operationalizations this one is less dependent on S's determination of what stimulus will be looked at or for how long. It has the advantage that it is bound up more exclusively with relations of similarity and dissimilarity between stimulus elements. It allows manipulation of exposure time in a systematic and continous way. There is no significant interaction between looking and exposure time.
