[Immune system in women with genital inflammatory diseases caused by Chlamydia and virus infection].

Immunologic studies were carried out in 29 women with clinical manifestations of chronic inflammations of the internal genitals. Dot-hybridization with biotin-labeled DNA probes revealed chlamydial DNA in the cervical cells of 14 women, Herpes simplex type 2 and/or cytomegaloviral DNA in 15 women. Cell-mediated immunity was assessed using monoclonal antibodies CD3, CD4, CD8, and CD19 and by Facscan flow cytofluorometer (Becton Dickinson). Immunoglobulins were measured by nephelometry with Abbott kits and TDx device. No appreciable changes in the mean values of the parameters characterizing T-cellular immunity were revealed in women with genital inflammations and chlamydial infection (p < 0.05). The counts of CD19+ lymphocytes varied to a certain degree, and IgM levels were increased in patients with chlamydial infection, this pointing to activation of B-lymphocytes with Chlamydia. A viral infection of the genitals was associated with a reduction of T-cellular immunity parameters and an imbalance in the ratio of immunoregulatory cells at the expense of reduction of the share of lymphocytes expressing CD4 antigen.

[Evaluation of the effects of methods of suppurative-inflammatory complications prevention after cesarean section on cellular and humoral components of the immune system].

The immune status of 73 pregnant women and puerperae, administered proteolysis inhibitors or common antibacterial therapy to prevent pyoseptic complications, was assessed before cesarean section and 1 and 7 days after it. The results indicate certain shifts in the immunity status of puerperae of all groups, which were the most expressed 1 day after surgery (reduced content of CD3 lymphocytes, IgG and IgA, and a marked increase of the content of C-reactive protein) and normalized by day 7 postoperation. These changes are most likely to be caused by the surgical intervention but not the infectious inflammatory process, which is proved by the absence of pyoseptic complications after prophylactic measures taken. Hence, proteolysis inhibitors, along with the traditional methods, may be recommended for the prevention of pyoseptic complications after cesarean section.

Detection of antisperm antibodies on the surface of living spermatozoa using flow cytometry: preliminary study.

OBJECTIVE: To assess the use of flow cytometry (FCM) analysis of living spermatozoa subjected to indirect immunofluorescence staining with the mouse anti-human immunoglobulin (Ig)G monoclonal antibodies (FCM test) as an objective method for the detection of antisperm antibodies in semen and serum. DESIGN: The comparative studies were conducted with the direct and indirect mixed antiglobulin reaction (MAR) and FCM test performed by FCM analyzer FACSCAN (Becton Dickinson Immunocytometry Systems, Mountain View, CA). SETTING: Clinical infertile service. PATIENTS: Seventy-six semen and 20 sera samples of infertile patients were analyzed. RESULTS: For IgG in semen there was a highly significant correlation between the direct MAR and FCM test. Six of 12 IgG-positive samples revealed also IgA, and just one sample, which showed negative MAR, was positive for IgM in FCM test. Flow cytometry test and indirect MAR showed also a highly significant correlation of the results of IgG detection in sera. CONCLUSIONS: Flow cytometry test may be used to verify that IgG antisperm antibodies are on the surface of living spermatozoa either retrieved directly from an ejaculate or after exposure to serum and to determine proportion of antisperm antibodies-positive spermatozoa, titer, and quantity of antibodies bound to the cell surface.

[The effect of a natural complex of cytokines on oxygen-dependent function of rat vaginal phagocytes].

Our investigation consisted in 10-days treatment of vaginal mucous membrane wounds in white not inbred rats by introduction of natural cytokines complex into the vagina. At the same time the dynamics of vaginal phagocytes oxygen-depended function change was studied. We have ascertained the activating influence of the cytokines complex on the phagocytes chemiluminescence (CL) that found expression in 2.5-7 times fold augmentation of the CL intensity in comparison with control level (an equal portion of Medium--199 was introduced in the vagina of control-group rats). Having ended the application of the cytokines complex we have observed a rather fast normalization of the cells' activity (by the 15-17 days), consequently the cytokines did not have a distant effect on the oxygen-depended phagocytes function. We consider the activation of the phagocytes in wound as well as the bactericidal effect of the oxygen free radicals generated by the phagocytes as positive result.

[Chronic viral diseases of the liver and pregnancy].

Sixty patients with chronic active hepatitis and liver cirrhosis of viral etiology had in the course of the disease a total of 130 pregnancies, that eventuated in labor in 62 (48%), in spontaneous abortions in 13 (10%), and in artificial abortions in 55 (42%) cases. The incidence of preterm labor was 18%, of late gestosis 46%; early detachment of a normally positioned placenta occurred in 2 women, and 3 women suffered an abnormal blood loss. Cesarean sections were carried out in 9% of cases. Perinatal mortality has made up 64.5%. Two babies died within the first months of life of congenital cerebral paralysis. The pregnancy and its outcome did not result in 78% of cases in deterioration of the patients' health status and did not influence the disease development. The authors discuss the policy of management of patients with chronic active hepatitis and liver cirrhosis of a viral origin in pregnancy and labor.

[Tumor necrosis factor in acute inflammatory diseases of the internal genitalia].

Blood serum cytotoxic activity of women with acute inflammatory diseases of the internal genitals was studied over the course of combined therapy, including immunomodulators, making use of target cells sensitive to the tumor necrosis factor (TNF). The levels of TNF activities varied within a wide range at the moment of the patients' admittance to hospital. TNF levels were high in the majority of patients or were increasing over the course of treatment. A low cytotoxic activity or its stable level over the course of treatment may be explained by a reduction of the immunocompetent cells functional activity.

[Ultrasonic study of the fetal heart in a pregnant woman with antiphospholipid antibodies].

Analysis of the findings of ultrasonic examinations of the fetal heart in pregnant women with antibodies to phospholipids has shown changes in this parameter in 42% of the examinees, in 3 cases antenatal fetal death was revealed. The detected changes were of different nature, but in the majority of cases they were of a compensatory type. Marked hypertrophy of the right ventricular myocardium, whose work in the antenatal period is largely responsible for the viability of the fetus, was found.

[Neutrophil chemiluminescence in active cytomegalovirus infection].

The effect of cytomegalovirus infection (CMVI) on the phagocytic activity of leukocytes was studied in patients by the method of luminol-dependent chemiluminescence (CL) used to evaluate the production of oxygen radicals by the peripheral blood neutrophils in phagocytosis of opsonized zymosan (OZ) in 10 female patients with active CMVI and 10 women of the control group. In the patients with CMVI, chemiluminescence in OZ phagocytosis was found to be reduced nearly by 1.5-fold as compared with the controls (pt < 0.05).

[The effect of dalargin on stress-induced changes in the 5'-nucleotidase activity of the macrophages and in the level of endogenous blood cortisol in mice].

The action of dalargin, synthetic analogue of leuenkephaline, on stress-induced changes in 5'-nucleotidase activity and endogenous hydrocortisone levels was investigated in mice. It was found that there is a direct relation between the activity of 5'-nucleotidase and the level of hydrocortisone in CBA mice. For C57Bl/6 mice the relation was inverse. Dalargin is able to change the dependence from direct to opposite in CBA mice.
