An anti-HIV 12mer oligonucleotide (ODN) conjugated to two different high molecular weight monomethoxy polyethylene glycols (MPEGs) has been tested for its antisense activity. The capacity of these conjugates to protect the MT-4 cells against HIV infection has been compared to the unmodified, native ODN, and the effect of the different structures of the supporting polymer has been discussed. It was found that only the ODN conjugated to the linear MPEG shows an anti-HIV activity in the investigated conditions. The same 12mer, when conjugated to a branched (MPEG)2, is fully inactive, as well as the native, unmodified ODN.
Preproenkephalin mRNA was analyzed in Neuroblastoma X Glioma, NG 108-15, cells as well as in respective parental cell lines, i.e. mouse Neuroblastoma N18 and rat Glioma C6, using a rat preproenkephalin cDNA as a probe. NG synthesize efficiently a preproenkephalin mRNA, similar in size to that of normal tissues. Of the two parental cell lines, Neuroblastoma seems to synthesize it as well as NG; conversely Glioma cell lines under the same conditions do not appear to synthesize preproenkephalin mRNA with the same efficiency as NG and N18.
The incidence of intestinal metaplasia (IM) was studied in 159 specimens from gastric mucosa. Rare in histologically normal specimens (4%), the IM showed the highest incidence in patients with gastric carcinoma (75%). Intermediate values were observed in chronic atrophic gastritis (25%) and peptic ulcer (21%). Histochemical data shows that type complete-IM was most common in benign conditions; type incomplete-IM and particularly subtype secreting sulphomucins revealed a higher frequence in the mucosa bordering carcinomas.
