Action of nicotinic acid (NA) on components of fibrinolysis in vitro and in vivo and on fibrinolytic activity in vivo was studied. It is shown that NA represses action of inhibitors as well as of fibrinolysis activators. Interaction of NA with plasma in vitro is associated with enhanced fibrinolytic activity on account of inhibition of antiplasmins. Action of NA of fibrinolytic activity of blood during intravenous administration depends on dose. Small doses of NA induce slight activation of fibrinolysis due to binding of antiplasmins. Administration of high doses of NA induces activation of fibrinolysis through thrombogenesis involving central nervous system.

Tricholysine (triase) and longolytin, isolated from cultural fluid of two similar lower fungi Trichothecium roseum and Arthrobotrys longa, exhibited anticoagulant and fibrinolytic properties after addition to blood plasma of experimental animals. Intravenous administration of the enzymes into animals caused the following effects: an increase in fibrinolytic activity, a slight decrease of fibrinogen concentration, an inadequate elongation of recalcification time and of thrombin time. Analysis of thromboelastograms demonstrated the anticoagulant effect: index of coagulation and maximal amplitude were decreased, time of coagulation was elongated. Preparation of tricholysine exhibited low toxicity; only doses, which exceeded 80-fold the thrombolytic dose, caused death in 34% of mice after intravenous administration.
An antiactivatory effect was estimated in blood plasma of human donors, rats and dogs on plates of stabilized fibrin using tissue plasminogen activator from pig heart. Inhibition of the tissue activator, used at definite concentrations, was expressed in per cents. The antiactivatory effect was distinctly higher in rat blood plasma, whereas it was similar in human and dog blood plasma, being about 3-fold decreased as compared with rats. The antiactivatory effect did not correlate with the antiactivator content in all the blood plasma preparations studied.
Fibrinolytic activity was studied in the renal vein of rats after electroshock. Electroshock increased distinctly the fibrinolytic activity in all parts of the venous system. The level of plasminogen activator was increased more in the renal vein and in the vena cava inferior, than in the jugular vein. In electroshock fibrinolytic activity was decreased 2-fold in kidney cortex; in the medullar layer of kidney it was unaltered.
Plasminogen activator isolated from cultural liquid of calf renal cells is an enzyme having a fibrinolytic action in vitro and in vivo, provoking a demonstrable thrombolytic effect, which depends on the drug dose, thrombus size and animal's body weight.
Intravenous administration of tissue activator of plasminogen obtained from pig heart into the healthy rats led to increase in the fibrinolytic activity in euglobulin fraction of blood plasma and to elevation in concentration of the products of fibrin degradation /fibrinogen/. Concentration of fibrinogen, the total fibrinolytic activity, inhibitors of fibrinolysis and the length of recalcification were unaltered distinctly in the experimental conditions described.
Fibrinolytic system was studied in rats under various conditions of intravenous administration of the plasminogen tissue activator (RTA). Two administrations of the activator with an interval of 10 min did not cause an additional activation of fibrinolysis as compared with single treatment. Increase in amount of administrations up to four times within a day (an interval for 30 min) as well as chronic administrations of PTA within 5 days did not cause also any significant and long-term action on the animal fibrinolytic system. Efficiency of blood plasma antiactivator was decreased after repeated and chronic PTA administrations. Insensitivity of the healthy animal fibrinolytic system to repeated and chronic administration of PTA depended apparently on binding of the activator to inhibitors and on its rapid excretion from circulation.
The patients with chronic heart disease (265 subjects) and with little active rheumatic fever (145 subjects) received two variants of isocaloric diets which differed only in protein quota. The former diet contained the physiological norm of protein (100 g) and the latter one a higher quota (130 g). As a result of the treatment the subjective sensations and objective status of all the patients improved. Evaluation of the time course of lipid metabolism, the system of hemostasis, capillary permeability and immunological reactivity showed that the diet containing the physiological norm of protein appeared the most effective in patients with ischemic heart disease, whereas that with a higher protein quota turned out the most adequate in patients with rheumatic fever.
Fibrinolytic and thrombolytic properties of morikrase, a complex of proteinases obtained from hepatopancreatic tissues of king crab (Paralithodes camtschatica), were studied in vivo and in vitro. High fibrinolytic activity and dose-dependent activating properties were detected on standard and heated fibrin plates. The preparation maintained its properties during long-term storage at 4 degrees and 22 degrees C. The thrombolytic activity of the drug, affecting the preformed thrombus, was observed both in vitro and in vivo, in rats with experimental thrombosis. Two-phase alteration of the fibrinolytic system and moderate hypocoagulation were found after intravenous administration of morikrase.
An ointment containing, 1% morikrase (a complex of proteinases isolated from hepatopancreatic tissues of king crab Paralithodes camtschatica) exhibited the wound healing properties and did not irritate the skin tissues during long-term treatments of rats. Only slight hypocoagulation was observed in intact animals after external application of the ointment. At the same time, the drug caused an increase in fibrinolytic activity in blood of rats with purulent wounds due to activation of the plasminogen activator.
Forty patients with coronary heart disease (CHD) concurrent with hyperlipidemia were divided into 2 groups according to the baseline fibrinolytic activity. The group with the depressed fibrinolytic system displayed the highest atherogenicity index and levels of cholesterol, low and very low density lipoprotein cholesterol and half levels of high density lipoproteins. After 45-day ingestion of docanol, a new Russian food additive containing docosahexanoic and eicosapentaenoic acids there were positive changes in the lipid spectrum in the both groups. The fibrinolytic changes were heterodirectional: activator and plasma activities increased in the group of fibrinolytic depression while in the group of baseline normal fibrinolysis the activity of plasminogen activator fell.
