[Interrelationship between the thiamine content, thiamine diphosphate-dependent enzyme activity and reduced glutathione level in the rat liver].

Changes in the amount of thiamine, reduced glutathione, thiamine diphosphate-dependent dehydrogenase activity has been traced after thiamine injection to thiamine-deficient rats and oxythiamine to normal rats. The obtained data show that a drop in reduced glutathione level was a primary reason of the alpha-keto-acid dehydrogenase activity reduction under conditions of the thiamine deficiency. The existence of immediate connection between thiamine and glutathione metabolism is supposed.

[Activity of certain redox enzymes of rat liver mitochondria at different levels of dietary vitamin E].

Taking into account the found earlier relation of vitamin E to the ubiquinone functioning and metabolism, the authors studied the enzymic activity of succinate dehydrogenase, NADH-dehydrogenase and cytochrome-c-oxidase--coenzyme Q binding sites of the respiratory chain of the rat liver mitochondria. The experiments were carried out with female rats who received a vitamin-E-deficient diet for 6 months. The enzymic activities and the ubiquinone content in the liver mitochondria of these animals are shown to be considerably lower as compared to the animals received a vitamin E diet; alpha-ocopherol, alpha-tocopheronolactone and ubiquinone 3h after administration manifest a clearly pronounced normalizing effect relative to both the enzymic activity and the ubiquinone content. An assumption is advanced that the effect of alpha-tocopherol and its metabolite is associated with controlling the level of functionally active ubiquinone in the mitochondria. Other mechanisms of the membrane-bound enzymes control by the compounds under study are also discussed in connection with the alpha-tocopherol effect on the mitochondrial membranes.

[Tocopherol-binding proteins in membranes of rat liver mitochondria].

The use of the adsorption chromatography on the hydroxyl apatite makes it possible to yield and partly purify the triton X-100-solubilized mitochondrial protein fraction of the rat liver able to bind specifically [3H]-alpha-tocopherol. The method permits removing simultaneously both free detergent and [3H]-alpha-tocopherol from the protein mixture without disturbance of the established equilibrium. When compared with methods used for the removal of free hydrophobic ligands in the in vitro binding experiments, the applied method is the most effective.

[Solubilization and various properties of NAD-binding receptor protein from rat brain synaptic membranes].

The NAD-binding receptor protein has been solubilized from the synaptic membranes of the rat brain by different detergents. Digitanin proved to be the most effective detergent which exerted no action on the specific binding of [14C]NAD with the solubilized receptor protein. Kinetic parameters of the soluble ligand-receptor complex were studied. The affinity of the solubilized receptor protein to NAD did not change as compared to the protein of native membranes. The specific binding of [14C]NAD was saturated at Kd = 0.53 microM, Bmax = 0.011 nmol per 1 mg of protein. The molecular weight of the soluble NAD-receptor complex determined under native conditions was equal to 115 kDa.

[The role of a protein factor in the effect of alpha-tocopherol on mitochondrial respiration].

It is shown that alpha-tocopherol in vitro stimulates respiration of the liver mitochondria in E-hypovitaminosis rats only in the presence of the specific protein factor isolated from the liver cytosol. The action of alpha-tocopherol on mitochondria in the presence of NAD and a protein factor is not accompanied by an increase in the NADH level, that evidences for the absence of the direct redox interaction between NAD and tocopherol.

[Characteristics of thiamine-binding protein from rat brain synaptosomes].

The thiamine-binding protein was obtained from rat brain synaptosomes by affinity chromatography and gel-filtration on Sephadex G-200. The protein is homogeneous by the data of SDS gel-electrophoresis, anode electrophoresis and isofocusing between pH 3.5-9.0. The isoelectric point of this protein is near pH 4.8-5.0. The binding nature of the protein with [14C] thiamine was studied. It is shown that metal ions, especially Na+ and Ca2+, increase the thiamine-binding activity. The binding process is of a saturation character at the thiamine concentrations of 10(-7)-10(-5) M. Thiamine possesses two binding sites with KD1 = 3.1 microM and KD2 = 30 microM. Out of the tested thiamine analogues and antagonists of vitamin B1 thiamine-monophosphate and pyrithiamine were the most competitive.

[The role of vitamin E in the body].

The basic experimental data obtained at the Department of Coenzymes' Biochemistry of the A. V. Palladin Institute of Biochemistry of the Ukr. SSR Academy of Sciences as to the biological role of vitamin E are analyzed. Vitamin E, selenium and methionine are found to induce peculiar changes in the activity of glutathione-peroxidase, metabolism of sulphur-containing amino acids, biosynthesis of adenine nucleotides, proteins and nucleic acids. Participation of alpha-tocopherol and its active derivatives in the control of biosynthesis and intertransformation of ubiquinone and its cyclic isomer, ubichromenol, in the animal organism, is proved, which determines to a considerable extent the biological role of vitamin E in the bioenergy processes. It is substantiated in experiments that the detected wide range of the biological effect of vitamin E is associated with the control of RNA biosynthesis. Under these conditions the effect of vitamin E on the RNA synthesis does not depend on the manifestation of antioxidant properties of its molecule and in this sense it is a specific one. The results obtained are discussed for their significance in explanation of the molecular mechanism of the vitamin E action and in substantiation of the possibility to use the results in practical medicine and animal husbandry.

[The role of vitamin E in the biosynthesis of the isoprenoid region of the ubiquinone molecule in the rat liver].

A decrease in the CoA and acetyl-CoA amount in the rat liver tissue by 34.8 and 29.4%, respectively, as well as inhibition of the biosynthesis rate of mevalonic acid from [I-14C] acetyl-CoA in the postmitochondrial liver fraction by 17.9% as compared to the control are found against a background of E-hypovitaminosis. The last change is not associated with the inhibition of the 3-oxy-3-methylglutaryl-CoA-reductase activity and may not be one of reasons which cause the biosynthesis disturbances in the isoprenoid part of the ubiquinone molecule in this organ. alpha-Tocopherol activates the ubiquinone biosynthesis from [2-14C] sodium acetate in the liver of rats with E-hypovitaminosis under conditions of 30 min preincubation and is not efficient when added in combination with actinomycin D. Probably, such an effect of alpha-tocopherol is realized at the level of RNA synthesis and is associated with the biosynthesis activation of short-living RNA.

[In vitro incorporation of [14CH3]methionine into liver phosphatidylcholine in rats with vitamin E deficiency].

The content of phosphatidyl choline, a terminal product of phosphatidyl ethanolamine methylation as well as in intensity in vitro incorporation of [14CH3]methionine into phosphatidyl choline of rat liver with E-hypovitaminosis are found to considerably decrease as compared to the norm. In this case the total content of phospholipids is practically the same.

[Phenylalanine-4-hydroxylase and tyrosine aminotransferase activities in rat liver tissue under different vitamin E content in the organism].

Enzymic activity of tyrosine aminotransferase is unchanged in subcellular fractions of the liver tissue of control, E-hypovitaminotic rats and of the same animals subjected to a single vitamin E administration. Enzymic activity of phenyl alanine-4-hydroxylase in the supernatant fraction determined in the incubation medium without biopterin in animals with E-hypovitaminosis is 39% lower than in the control rats. The coefficient of phenyl alanine-4-hydroxylase activation with biopterin in vitro in E-hypovitaminotic animals is thrice as high as that in the control rats. A single vitamin E administration produces a 1.8-fold decrease in the activation coefficient. The data obtained give reason to suggest the possible influence to vitamin E on the biopterin level in the liver.

[E-vitamin activity of vitamin E derivatives with experimental encephalomalacia in chicks].

When adding pharmacopoeian alpha-tocopherylacetate, short-chain alpha-tocopherylacetate, alpha-tocopherylquinine, short-chain alpha-tocopherylquinone and alpha-tocopheronolactone to E-avitaminotic rations pharmacopoeian alpha-tocopherylacetate and alpha-tocopheronolactone manifest the highest E-vitamin activity in preventing encephalomalacia in chickens. The action of alpha-tocopheronolactone is not directly associated with changes in the content of vitamin E and ubiquinone in the brain and liver tissues. All the studied derivatives are effective in increasing resistance of erythrocytes to osmotic hemolysis. The data obtained evidence for a nonspecific function of vitamin E in preventing alimentary encephalomalacia in chickens as well as for the absence of disturbances in ubiquinone metabolism under conditions of the E-hypovitaminosis experimental model.
