New antibiotics from Actinoplanes strains. Structure of A 17002 C.
A 17002 C is a new metabolite produced by Actinoplanes strains, structurally related to the virginiamycin factor M. On the basis of physico-chemical data, MS, IR, 1H and 13C NMR, structure I is assigned to A 17002 C.
Heparin interferes with albumin determination by dye-binding methods.
Heparin, when added in vitro at rather high concentrations (400--1000 kilounits/L) as an anticoagulant, interferes with albumin determination by bromcresol green by forming an insoluble precipitate with this dye. The interference is much less with bromcresol purple. It can be abolished by addition of hexadimethrine bromide.
M.S. diagnostic test: a comparison of different electrophoretic and immunologic methods for CFS IgG estimation.
In order to select the most discriminative laboratory method of MS diagnosis, the results obtained with CSF IgG/total protein ratio, CSF-IgG Index, agarose and cellulose acetate electrophoresis were compared. The electrophoretic methods appear to be more discriminative for MS than IgG/total protein ratio or the CSF-IgG Index. Furthermore, cellulose acetate electrophoresis was found to be simpler and quicker to perform than agarose electrophoresis.
Synthesis and biological activity of new 2-nitroimidazole derivatives.
In an earlier paper we described the synthesis and the antitrichomonas activity of 2-nitro-alpha,alpha,1-trimethyl-1H-imidazole-5-methanol (2). Starting from this compound, several derivatives have been synthesized. Among these, the phenyl carbonate 8 has been shown to possess activity equal to that of 2 and to be less toxic. This compound therefore is interesting in comparison with some antitrichomonas agents currently in use clinically. Before undertaking an in-depth investigation, compound 8 was subjected to some studies to see whether it has any effects on the central nervous system (CNS). Preliminary results show that, at therapeutic doses, it might induce unwanted CNS effects to a lesser degree than metronidazole.
Synthesis and biological activity of some vinyl-substituted 2-nitroimidazoles.
In previous studies 1-methyl-2-nitro-1H-imidazole-5-carboxaldehyde and 1-methyl-2-nitro-5-vinyl-1H-imidazole were found to posses interesting antimicrobial activities. We have now prepared some 2-nitro-1H-imidazoles in which the 5-vinyl chain bears selected functional groups (CHO, COCH3, NO2) as well as nitrogen-condensation derivatives of the carbonyl functions. Furthermore, 5-methyl-2-nitro-1-vinyl-1H-imidazole has been synthesized. All the compounds, and some intermediates, have been assayed for antimicrobial activity. Several of them exhibited significant antibacterial and antitrichomonal activity in mice.
Synthesis and biological activity of two metabolites of 1-methyl-5-(1-methylethyl)-2-nitro-1 H-imidazole, an antiprotozoal agent.
5-(-Hydroxyl-1-methylethyl-)-1-methyl-2-nitro-1 H-imidazole (6) and 5-(1,2-dihydroxyl-1-methylethyl)-1-methyl-2-nitro-1 H-imidazole (9) are the principal metabolites found in urines of animals (mice, rats, and dogs) treated with 1-methyl-5-(1-methylethyl)-2-nitro-1 H-imidazole (1), an effective antitrichomonas agent. These two metabolites have been synthesized. Compound 6 was found to be less toxic than the parent compound 1 and to possess essentially the same activity against Trichomonas vaginalis in experimental infections. Compound 9 showed a low degree of in vivo activity.
1-Methyl-2-nitroimidazol-5-yl derivatives. IVth Communication.
The chemico-physical data and antimicrobial activities are described of a new series of 2-nitroimidazole derivatives prepared by condensing 1-methyl-2-nitroimidazole-5-carboxaldehyde with N-substituted hydroxylamines, N-aminopiperazines and N-aminopiperidines.
1-Methyl-2-nitroimidazol-5-yl derivatives. IIIrd communication.
A series of 1-methyl-2-nitroimidazole-5-carboxaldehyde derivatives (substituted hydrazones, oximes and Schiff bases) have been prepared and tested for their antimicrobial activity.
Synthesis and biological activity of some 2-aminoimidazoles.
A review of the literature showed that 2-amino-imidazoles have not benn taken into consideration as antimicrobial agents. Our preliminary data indicating that 2-amino-4(5)-phenylimidazole possessed a moderate in vitro activity prompted us to prepare a limited series of 2-amino-4(5)-arylimidazoles and of some related derivatives. In vitro atimicrobial screening gave evidence of a broad spectrum activity for some of them. Several compounds have been tested also against S. mutans. In particular, 2-amino-4(5)-(4-biphenylyl)-imidazole hydrochloride showed substantial activity in this test, confirmed also against defferent representative strains of S. mutans. When submitted to a plaque inhibition test, it strongly inhibited the plaque forming ability of a S. mutans strain.

Synthesis and preliminary pharmacological studies of some 3-substituted 5-amino-1,2,4-oxadiazoles.
Treatment of 4(5)-alkyl (or aryl)-2-aminoimidazoles with isoamyl nitrite in acetic acid gave rise to the corresponding 5(4)-substituted 2-amino-4(5)-hydroxyimino-4(5)H-imidazoles which were transformed into 3-acyl (or aroyl)-5-amino-1,2,4-oxadiazoles when treated in aqueous suspension. The results of a preliminary pharmacological investigation showed an interesting depressant activity on the CNS for one of these compounds, 5-amino-3-benzoyl-1,2,4-oxadiazole (III c). The pharmacological activities are correlated with the chemical substitutions on the oxadiazole ring.
[Some compounds correlated to pyracrimycin A].
Some biological characteristics of pyracrimycin A (I), a metabolic compound isolated from cultures of Streptomyces eridani are examined. Some other compounds related to I, obtained both by chemical reaction from the natural product and by synthesis are described. The in vitro antibacterial activity seems to be due to the presence of the unsaturated moiety --N equals (see article) --CH equals CH-- (see article) equals O.
