Polyurea-functionalized multiwalled carbon nanotubes: synthesis, morphology, and Raman spectroscopy.

An in situ polycondensation approach was applied to functionalize multiwalled carbon nanotubes (MWNTs), resulting in various linear or hyperbranched polycondensed polymers [e.g., polyureas, polyurethanes, and poly(urea-urethane)-bonded carbon nanotubes]. The quantity of the grafted polymer can be easily controlled by the feed ratio of monomers. As a typical example, the polyurea-functionalized MWNTs were measured and characterized in detail. The oxidized MWNTs (MWNT-COOH) were converted into acyl chloride-functionalized MWNTs (MWNT-COCl) by reaction with neat thionyl chloride (SOCl2). MWNT-COCl was reacted with excess 1,6-diaminohexane, affording amino-functionalized MWNTs (MWNT-NH2). In the presence of MWNT-NH2, the polyurea was covalently coated onto the surfaces of the nanotube by in situ polycondensation of diisocyanate [e.g., 4,4'-methylenebis(phenylisocyanate)] and 1,6-diaminohexane, followed by the removal of free polymer via repeated filtering and solvent washing. The coated polyurea content can be controlled to some extent by adjusting the feed ratio of the isocyanato and amino groups. The structure and morphology of the resulting nanocomposites were characterized by FTIR, NMR, Raman, confocal Raman, TEM, EDS, and SEM measurements. The polyurea-coated MWNTs showed interesting self-assembled flat- or flowerlike morphologies in the solid state. The signals corresponding to that of the D and G bands of the carbon nanotubes were strongly attenuated after polyurea was chemically tethered to the MWNT surfaces. Comparative experiments showed that the grafted polymer species and structures have a strong effect on the Raman signals of polymer-functionalized MWNTs.

Functional analysis of promoter activity of murine beta-1,6-N-acetylglucosaminyltransferase.

In the mouse, beta-1,6-N-acetylglucosaminyltransferase (IGnT) forms branches in poly-N-acetyllactosamines, which are good scaffolds for oncodevelopmental cell-surface antigens and recognition markers. There are two isoforms of IGnT, IGnT A and B, which are produced by alternative splicing of the IGnT gene; the unique portion is encoded by exon 1 and common portion is encoded by exons 2 and 3. Thus, the expression of each isoform is controlled by a different promoter. Here, we identified the regulatory regions of the mouse IGnT A and B genes. The promoter regions for IGnT A and B did not contain putative TATA or CAAT boxes, but each contained GT boxes. The upstream regulatory region of each gene was examined by transient luciferase reporter gene transfection experiments and gel mobility shift assay. Promoter activity for each gene was detected in F9 embryonal carcinoma cells, which express IGnT A and B, but not in N2a cells, which do not express the gene. Deletion analysis demonstrated that the regions 308 bp upstream from the transcriptional initiation site of IGnT A and 430 bp upstream from the transcriptional initiation site of IGnT B showed minimal promoter activity. Mutation of the single GT box in IGnT A and two GT boxes in IGnT B caused marked reduction of the promoter activity. These findings provided strong evidence that the GT boxes play crucial roles in transcriptional regulation of the genes.

Glypican-2 binds to midkine: the role of glypican-2 in neuronal cell adhesion and neurite outgrowth.

Cell-surface heparan sulfate proteoglycans participate in molecular events that regulate cell adhesion, migration, and proliferation. The present study was performed to elucidate whether glypican-2 plays a role in interactions of neurons with midkine (MK), a heparin-binding neuroregulatory factor. MK bound to heparan sulfate chains of glypican-2 in a manner similar to syndecan-3. Microbeads coated with MK or poly-L-lysine induced clustering of glypican-2 as well as syndecan-3. Substratum-bound MK or poly-L-lysine induced cell adhesion of N2a neuroblastoma cells, while only MK promoted neurite outgrowth of these cells. Ligation of cell-surface glypican-2 with MK or an antibody against epitope-tagged glypican-2 induced cell adhesion and promoted neurite outgrowth. These results verified that cell-surface glypican-2 bound to MK and suggested that MK-glypican-2 interactions participate in neuronal cell migration and neurite outgrowth. In addition, we observed different localization of epitope-tagged glypican-2 and syndecan-3 on the surface of N2a cells; the result suggested that they may play different roles in MK-mediated neural function.

A new 3-D display method for 12-lead ECG.

A new three-dimensional (3-D) 12-lead electrocardiogram (ECG) display method is presented which employs a 3-D rectangular coordinate system to display the 12-lead cardiac electric signals in two 3-D graphs. The 3-D graph consists of a temporal axis representing the time domain of the cardiac signals, a spatial axis representing the lead positions, and an amplitude axis representing the voltages of the cardiac signals. The six horizontal plane leads and the other six frontal plane leads were displayed in two 3-D graphs, respectively. The voltages of the cardiac signals were represented in rainbow-like colors. Cubic interpolation was employed to insert interconnecting points between neighboring leads on each plane and to smooth the surface of the 3-D ECG graphs. The 3-D ECG graphs of a normal subject, a patient with myocardial infarction, and a patient with left bundle branch block were presented in this paper. This new display method could not only be used as a complementary display method to the 12-lead ECG, but also provide physicians with an overall integral view about the spatial distribution of the cardiac signals.

[Environmental investigation in areas where Anopheles anthropophagus distributed in Hubei Province].

OBJECTIVE: To investigate the natural and socio-economical environment in the areas where Anopheles anthropophagus distributed in Hubei Province. METHODS: 5% of the villages in distribution area of An. anthropophagus were randomly selected for the investigation which included vegetation, soil, water, temperature, farming, resting places of the mosquito. RESULTS: The vegetation of An. anthropophagus area consisted of forest, bush, sod and crops. The area was densely covered with river, ditches and ponds, the water pH being 6.1-7.7. The soil texture was either yellow, yellow brown, grey or black, contained 2.72% organic material. There were one or two seasons of rice plantation in the area and the amount of insecticide used was 0.828 kg/mu in the rice field. The annual average temperature was 16.9 degrees C and the humidity was 76.9%. The breed period of An. anthropophagus was from June to September. There are two peaks of mosquito population in area with two seasons of rice plantation, and one peak in one rice season. The composition of An. anthropophagus resting in households, cowsheds and pigpens was 80.9%, 12.2% and 6.0%, respectively. CONCLUSION: The distribution of An. anthropophagus was in point, flaky or belt form in low hill, hillock, shallow hilly plain. These areas were full of vegetation and source of water, the principal crop was single cropping of rice, and soil texture is yellow or yellow brown. The major resting place of the mosquito was human dwellings.

[Study on the essential oil components in Thymus quinquecostatus Celak].

AIM: To analyse the essential oil components of Thymus quinquecostatus Celak. METHODS: Gas chromatography/mass spectrometry were used. RESULTS: A total of 78 components were separated. 34 of them were identified, which accounted for 95.93% of the total peak area. The main constituents are o-cymene, carvacrol, caryophyllene, 2-isopropyl-1-methoxy-4-methylbenzene and gamma-terpene. CONCLUSION: Thymus quinquecostatus Celak has perfume and medical development value.

The impact of overall treatment time on outcomes in radiation therapy for non-small cell lung cancer.

PURPOSE: A retrospective study was carried out to evaluate the impact of overall treatment time (OTT) on the results of radiation therapy for non-small cell lung cancer (NSCLC). MATERIALS AND METHODS: From Jan. 1990 to Dec. 1996, 256 patients with stages I-IIIb NSCLC entered this analysis. All patients received definitive radiotherapy. Biologically effective dose (BED) was used to standardize the irradiation effects. The correlation between OTT and local progression-free survival was analyzed by linear-regression and Cox proportional hazard models. The prognostic variables for survival and distant metastasis were also briefly studied. RESULTS: OTT had been shortened in 64 patients because of an accelerated hyperfractioned irradiation, while OTT was prolonged i n 114 patients due to interruptions of irradiation courses. The main ca uses of interruption were machine breakdown or delayed preparations of c errobend block for boost fields (55%), holidays (11%) and treatment toxi city and side effects (34%). Patients tre ated with prolonged OTT (> 45 days) had significant poorer local progression-free survival than whom with OTT of </=45 days, 1, 3 and 5 year actuarial local progression-free survivals being 49, 17 and 15% for the former, and 74, 35 and 25% for the latter, respectively (P<0.001). BED-T that contained the factor of OTT correlated directly to local controls, which implied that BED-T represented radiobiological effects accurately, in other words, OTT had played a role in determining the radiobiological effects. Linear-regression on 103 cases treated with BED of 80-85 Gy(10) showed that 3 year local progression-free survival decreased by 9% per week with prolongation of OTT, or vice versa it increased by 9% per week with shortening OTT in an OTT range of 30-76 days. Cox multivariate analyses confirmed that OTT was an independent prognostic factor for local controls. CONCLUSION: OTT may have played an important role in determining local controls in radiotherapy for NSCLC. One should always keep in mind to make the OTT as short as possible, provided the patients can tolerate it, and to reduce irradiation interruptions for whatever reasons to a minimum.

A novel variant form of murine beta-1, 6-N-acetylglucosaminyltransferase forming branches in poly-N-acetyllactosamines.

A novel form of murine beta-1,6-N:-acetylglucosaminyltransferase that forms branches in poly-N:-acetyllactosamines (designated as IGnT B) was cloned based on sequence homology to the known IGnT (designated as IGnT A). When expressed as proteins, IGnT B showed higher specific activity than IGnT A. The C-terminal 1/4 of IGnT B was identical to that of IGnT A, while the rest of the predicted sequences showed 63% identity. Genomic analysis indicated that IGnT A and IGnT B were derived by alternative splicing; the unique portion was encoded by exon 1, and the common portion was encoded by exons 2 and 3. IGnT B showed an expression profile closely related to that of IGnT A and was strongly expressed in the liver, kidney and intestine, and moderately in the mammary gland, submaxially gland, embryonic stem cells, and embryonal carcinoma cells. The specificity of IGnT B examined using various substrates was indistinguishable from that of IGnT A, which is classified as the central acting IGnT (cIGnT). Thus, IGnT B acted on Galbeta1-4GlcNAcbeta1-3Galbeta1-4Glc, but not on GlcNAcbeta1-3Galbeta1-4Glc. It formed branches in both of the internal galactosyl residues of Galbeta1-4Glc-NAcbeta1-3Galbeta1-4GlcNAcbeta1-++ +3Galbeta1-4Glc, and prolonged incubation resulted in production of the di-branched oligosaccharide. Although addition of sialic acid to the terminal galactosyl residue did not abolish the acceptor activity, alpha2-6 sialylation was a preferred one as compared to alpha2-3 sialylation.

Biphasic changes in autonomic nervous activity during pregnancy.

To understand the sequential response of the autonomic nervous system to pregnancy, we studied heart rate variability in 23 first trimester, 23 second trimester and 21 third trimester pregnant women. Twenty non-pregnant women were recruited as controls. Time and frequency domain measures of heart rate variability in three recumbent positions were compared. We found that normalized high-frequency power in the supine position increased significantly in the first trimester (42.2 (95% confidence interval (CI) 5.4) nu (normalized unit); P < 0.05) compared with non-pregnant controls (33.0 (6.0) nu), and then decreased progressively in the second (27.3 (6.7) nu) and third (21.8 (6.0) nu; P < 0.05) trimesters. The low-/high-frequency power ratio in the supine position decreased significantly in the first trimester (0.8 (0.3); P < 0.05) compared with that of non-pregnant controls (1.1 (0.3)) and increased progressively in the second (1.5 (0.4)) and third (2.1 (0.8); P < 0.05) trimesters. When the position was changed from the supine to the right lateral decubitus, the percentage change in normalized high-frequency power correlated significantly and negatively with normalized high-frequency power in the supine position in non-pregnant controls (r = -0.56, P = 0.01) and in pregnant women in the first (r = -0.44, P = 0.034), second (r = -0.68, P < 0.001) and third (r = -0.68, P < 0.001) trimesters. These results indicate that autonomic nervous activity shifted towards a lower sympathetic and higher vagal modulation in the first trimester, and changed towards a higher sympathetic and lower vagal modulation in the third trimester as gestational age increased. The balance between the haemodynamic changes of pregnancy and aortocaval compression caused by the enlarging gravid uterus may be responsible for the biphasic changes in autonomic nervous activity during pregnancy.

Effect of different recumbent positions on spectral indices of autonomic modulation of the heart during the acute phase of myocardial infarction.

OBJECTIVE: To examine which recumbent position can lead to the highest vagal modulation in patients during the acute phase of myocardial infarction. DESIGN: Descriptive study. SETTING: Intensive care unit in a medical center. PATIENTS: A total of 52 patients admitted to the intensive care unit because of acute myocardial infarction (AMI), 41 patients with coronary artery disease (CAD), and 28 patients with patent coronary arteriogram. INTERVENTIONS: None. MEASUREMENTS AND MAIN RESULTS: Heart rate variability analysis was performed in patients with AMI, patients with CAD, and patent coronary controls in supine, left lateral decubitus, and right lateral decubitus positions in random order. In the right lateral decubitus position, the vagal modulation was the highest and the sympathetic modulation was the lowest among three recumbent positions in three groups of patients. When the position was changed from supine to right lateral decubitus, the increase in vagal modulation was greater in patients who had more severely depressed vagal modulation in the supine position and the rate of increase was the greatest in patients with AMI, followed by patients with CAD and patent coronary controls. Detailed analysis showed that the vagal enhancing and sympathetic suppression effect of the right lateral decubitus position applied to patients with Q wave myocardial infarctions. CONCLUSIONS: The right lateral decubitus position can lead to the highest vagal modulation and the lowest sympathetic modulation among three recumbent positions in patients with Q wave myocardial infarction. The right lateral decubitus position can be used as an effective vagal enhancer in patients with Q wave myocardial infarction but without severe bradycardia or atrioventricular block.

Sequential changes in heart rate variability after coronary artery bypass grafting.

Heart rate variability (HRV) decreased soon after coronary artery bypass grafting and returned to the preoperative level within 2 months; however, HRV did not exceed the preoperative level, even 6 months after coronary artery bypass grafting. Although myocardial ischemia can be improved by coronary artery bypass grafting, HRV did not benefit from coronary artery bypass grafting within 6 months.

Comparison of supine and upright positions on autonomic nervous activity in late pregnancy: the role of aortocaval compression.

In order to understand the effect of aortocaval compression on autonomic nervous activity, we compared the effects of supine and upright postures on measures of heart rate variability in late pregnancy. Thirty-two women in late pregnancy and 23 nonpregnant age-matched women were studied. Both time and frequency domain heart rate variability measures were compared between the supine and upright positions, and between pregnant women and controls. The pregnant women had smaller mean RR interval, standard deviation of RR intervals and normalised high-frequency power, and a larger low- to high-frequency power ratio than nonpregnant women in the supine position. When the position was changed from upright to supine in nonpregnant women, the percentage change in the mean RR interval and normalised high-frequency power were increased, whereas the percentage change in the coefficient of variation of RR interval and low- to high-frequency power ratio were decreased. In contrast, the percentage change in normalised high-frequency power was decreased and the percentage change in low- to high-frequency power ratio was increased in pregnant women when the position was changed from upright to supine. These results indicate that the vagal enhancing effect seen in normal subjects when changing position from upright to supine was reversed in women in late pregnancy. Aortocaval compression in the supine position in pregnant women might be responsible for this reversal in the autonomic response to lying down.

Return of autonomic nervous activity after delivery: role of aortocaval compression.

We have compared measures of heart rate variability (HRV) in 15 women in late pregnancy before and 3 months after delivery and in 20 non-pregnant controls, to study the effect of aortocaval compression on autonomic nervous activity. Normalized high-frequency power was used as an index of vagal activity and the low-/high-frequency power ratio as index of sympathovagal balance. We found that the decrease in vagal activity and increase in sympathovagal balance before delivery returned to normal 3 months after delivery. Percentage changes in vagal activity and sympathovagal balance 3 months after delivery in the left lateral decubitus position were least among three recumbent positions. The relief of aortocaval compression as a result of delivery of the fetus may be the main cause of return of autonomic nervous activity 3 months after delivery. Postpartum hormonal changes may also contribute to return of autonomic nervous activity after delivery.

Comparison of three recumbent positions on vagal and sympathetic modulation using spectral heart rate variability in patients with coronary artery disease.

Patients with coronary artery disease (CAD) have depressed vagal modulation. Because the mortality risk from acute myocardial infarction is lower in patients with higher vagal modulation, methods that can increase vagal modulation are desirable for patients with CAD. The right lateral decubitus position has been found, in young healthy subjects, to lead to the highest vagal modulation among 3 recumbent positions using spectral heart rate variability (HRV) analysis. This study investigated which recumbent position can give rise to the highest vagal modulation in patients with severe CAD. Twenty-seven patients scheduled for coronary artery bypass graft surgery and 17 patients with angiographically normal coronary arteries were studied. Spectral HRV analysis was performed in random order on these patients in 3 recumbent positions: namely, the supine, left lateral decubitus, and right lateral decubitus positions. Normalized high-frequency power was the highest, whereas normalized low-frequency power and low/high-frequency power ratio in the right lateral decubitus position were the lowest, among the 3 recumbent positions in both groups of patients. The lower the normalized high-frequency power in the supine or left lateral decubitus position, the higher the increase in the normalized high-frequency power when the position was changed from supine or left lateral decubitus to right lateral decubitus in patients with severe CAD. Right lateral decubitus position can lead to the highest vagal modulation and the lowest sympathetic modulation among the 3 recumbent positions in patients with severe CAD. Therefore, the right lateral decubitus position can be used as an effective physiologic vagal enhancer in patients with severe CAD.

Energy dissipation as a key factor for electroporation of protoplasts.

Energy dissipation (epsilon) during electroporation was theoretically determined to be epsilon = 0.5CV02 for the various combinations of capacitance (C) and initial voltage (V0). Experiments on asparagus protoplasts established that electroporation efficiency (EE) and survival rate were directly proportional to energy dissipation during electroporation. A positive linear relationship exists between energy dissipation per unit volume and EE, whereas energy dissipation per unit volume and survival rate of protoplasts are related in a negative linear manner. At the same energy level, longer time constants were more effective at increasing EE. This suggests that energy dissipation approximating rectangular waveforms is more important than that dissipated as sharply decaying exponential waveforms. With energy as the key parameter, the optimization of electrical parameters for efficient electroporation is greatly simplified, is not machine-dependent, and generally applies to all species.
