Transcriptomic responses of Porphyrophora sophorae larvae during licorice root colonization reveal coordinated remodeling of translation, mitochondrial energy metabolism and defense-related genes.

BACKGROUND: Porphyrophora sophorae is a subterranean piercing-sucking scale insect that damages licorice (Glycyrrhiza uralensis) roots, but the molecular responses associated with larval root colonization remain insufficiently defined. METHODS: We compared non-parasitic larvae (NP) and root-colonizing larvae (RC) using six RNA-seq libraries, de novo transcriptome assembly, DESeq2-based differential expression analysis, GO/KEGG enrichment, annotation-based candidate gene screening, and RT-qPCR validation of selected genes. RESULTS: Sequencing yielded 260.91 million clean reads, and de novo assembly produced 60,794 non-redundant transcripts. DESeq2 identified 703 FDR-significant DEGs, including 49 upregulated and 654 downregulated genes in RC larvae. Upregulated genes were mainly associated with translation- and ribosome-related processes, whereas downregulated genes were enriched in mitochondrial, oxidation-reduction, energy metabolism, and oxidative phosphorylation-related functions. Annotation-based screening identified 75 FDR-significant candidate genes associated with chemosensation, defense-related responses, and energy metabolism, with mitochondrial energy metabolism-related genes forming the largest module. RT-qPCR validation based on the raw Ct data showed concordant expression directions for ten selected transcript targets. CONCLUSIONS: Root colonization in P. sophorae larvae was associated with coordinated transcriptional remodeling involving selective activation of translation-related processes, adjustment of mitochondrial energy metabolism, and changes in defense-related gene expression. These results provide candidate molecular targets for future functional studies of host contact, feeding establishment, and physiological adjustment in this subterranean scale insect.

Density functional theoretical investigations on various nanostructural zeolite surfaces.

Through density functional calculations, the Brønsted acidities on various nanostructural ZSM-5 zeolite surfaces were studied as well as the hydrogen exchanging processes with adsorbed H(2)O monomer or dimer. The Brønsted acidities on the four nanostructural surfaces show differences, although slightly, with their strengths increasing as (100) < (210) < (410) < (001). For hydrogen exchanging processes with H(2)O monomer or dimer, the reaction rate increases in the order (210) < (100) < (001) < (410) or (210) < (410) < (001). No transition-state structure is present on H(2)O dimer/(100) surface system. The introduction of a second H(2)O molecule accelerates the hydrogen exchanging processes and meanwhile influences the nanostructural geometries such that they are more evident. Besides the activation barrier, the adsorption energy and reaction heat display differences from one surface to another, which results in the preference of catalytic reactions to a specific nanostructural zeolite surface, such as the hydrogen exchanging processes studied in this paper.

H2 adsorption on Fe/ZSM-5 zeolite: a theoretical approach.

The hydrogen reduction of the red-ox Fe/ZSM-5 zeolite is an essential step for catalyst preparation. In this letter, various adsorption modes of the H(2) molecules on the Fe(III)/ZSM-5 zeolite were first explored by density functional theory, with their exact configurations provided. The adsorption energies revealed that the two configurations with Fe(III) at the sextet states are the main ones, consistent with the results of inelastic neutron scattering (INS) experiments. On such basis, the vibrational and orbital analyses were made, which will be valuable for the future studies on the Fe/ZSM-5 systems.

The effects of work experience, lift frequency and exposure duration on low back muscle oxygenation.

BACKGROUND: Previous studies have shown changes in low back muscle oxygenation after static muscle contractions or short-term dynamic lifting exertions. The aim of this study was to document the changes in low back muscle oxygenation during prolonged lifting activity over an entire workday as of function of work experience and lift frequency. METHODS: Four novice and six experienced subjects participated in a lifting study in which they lifted load with a given weight at one of five different frequencies (2, 4, 8, 10, 12 lifts/min) for an 8-h period. Oxygen saturation of the left and right erector spinae was measured continuously and non-invasively using near-infrared spectroscopy during each lifting session. FINDINGS: Exposure duration had a statistically significant effect on muscle oxygenation level (P<0.0001). Oxygen saturation in the erector spinae increased during the 8-h lifting period. As lift frequency increased, back muscle oxygenation in experienced subjects also increased. In general, the increase in muscle oxygenation for experienced subjects was less than that for novice subjects. INTERPRETATION: This study suggested that the requirement of oxygen for the low back muscle in a typical industrial lifting job increased over time and experienced workers responded differently from the novice subjects. These findings may provide more insight into the physiological changes of the working muscle and the potential risks of developing muscle injury.

The effect of alpha-adrenergic antagonists in chronic prostatitis/chronic pelvic pain syndrome: a meta-analysis of randomized controlled trials.

The effectiveness of alpha-adrenergic antagonists on patients with chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS) has not been supported by well-evaluated study. The meta-analysis was performed to supply the best evidence about use of this class of drugs in CP/CPPS. A fully recursive literature search to June 2005 was conducted in PubMed, EMBASE, the Cochrane Controlled Trials Register, and the Chinese Biomedicine Database to identify potentially relevant randomized controlled trials. RevMan4.2 was used for statistical analysis. Nine studies with 734 patients were included. Combined analysis showed a significant reduction of total NIH-CPSI or I-PSS in patients with treatment duration of more than 3 months. There were also valuable results in urinary symptom alleviation. Alpha-adrenergic antagonists did not show benefit in pain. The meta-analysis revealed that the use of alpha-adrenergic antagonists was warranted in CP/CPPS, and the treatment duration should be long enough (more than 3 months).

Estimation of the aqueous solubility of weak electrolytes.

Jain and Yalkowsky [Jain, N., Yalkowsky, S.H., 2001. Estimation of the aqueous solubility. I. Application to organic non-electrolytes. J. Pharm. Sci. 90, 234-252.] demonstrated that the general solubility equation (GSE) can be used to estimate the aqueous solubility of organic non-electrolytes. In this study the applicability of the GSE was extended to weak electrolytes. It is demonstrated that the GSE estimates the aqueous solubility of 949 compounds, including 367 weak electrolytes with an AAE of 0.58. It is also shown that the intrinsic solubilities of weak acids for which the pK(a)+log S(w)<or=0 and for weak bases for which pK(a)-log S(w)<or=14 are within a factor of 2 of the total solubilities.

Polyaniline/Au composite hollow spheres: synthesis, characterization, and application to the detection of dopamine.

Polyaniline (PANI)/Au composite hollow spheres were successfully synthesized using polystyrene/sulfonated polystyrene core/shell gel particle templates. The PANI shell thickness and the number of Au nanoparticles decorating the PANI could be controlled effectively by adjusting the experimental conditions. The morphology, composition, and optical properties of the resulting products were characterized by scanning electron microscopy, transmission electron microscopy, X-ray diffraction, thermogravimetric analysis, X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy, and ultraviolet-visible absorption spectra. It was found that the electrical conductivity of the PANI/Au composite hollow spheres was more than 3 times higher than that of the pure PANI hollow spheres. Furthermore, PANI/Au composites were immobilized on the surface of a glassy carbon electrode (GCE) and applied to construct a sensor. The obtained PANI/Au-modified GCEs showed one pair of redox peaks and high catalytic activity for the oxidation of dopamine. The possible formation mechanism of the PANI/Au composite hollow spheres was also discussed.

Spinal loading during manual materials handling in a kneeling posture.

Stooped, restricted, kneeling, and other awkward postures adopted during manual materials handling have frequently been associated with LBP onset. However, lift assessment tools have focused on materials handling performed in an upright, or nearly upright standing posture. Unfortunately, many of the tools designed to analyze standing postures are not easily adapted to jobs requiring restricted postures. Therefore, the objective of this study was to evaluate spinal loading during manual materials handing in kneeling postures and determine if those loads can be predicted using simple regression. An EMG-driven biomechanical model, previously validated for upright lifting, was adapted for use in kneeling tasks. Subjects knelt under a 1.07m ceiling and lifted luggage of six weights (6.8, 10.9, 15.0, 19.1, 23.1 and, 27.2kgf) to one of four destination heights (0, 25.4, 53.3, 78.7cm). Spine loading was significantly affected by both destination height and load weight. Destination height increased compression, AP shear and lateral shear by an average of 14.5, 3.7 and 6.6N respectively per cm height increase. Load weight increased compression, AP shear and lateral shear by an average of 83.8, 27.0 and 13.1N respectively per kgf lifted. Regression equations were developed to predict peak spine loading using subject height, load weight and destination height with R(2) values of 0.62, 0.51 and 0.57 for compression, AP and lateral shear respectively.

[Change of substance P in portal vein during rectoanal inhibitory reflex].

OBJECTIVE: To investigate the change pattern of substance P (SP) in the portal vein during the rectoanal inhibitory reflex (RAIR), and its physiologic significance; the influence of external splanchnic nerve of rectum and anal to the RAIR. METHODS: The rats were divided into seven groups, among them there were six groups, which were first divided into two big groups according to whether the external splanchnic nerve to the rectum and anal were cut off, one is no cut-off external splanchnic nerve group, the other is cut-off external splanchnic nerve group. Each group were further divided, according to the distance of the balloon-sac on Foley's tube in the rectum away from anal verge, into 2, 4, 6 centimeter groups; A control group with Foley's tube put into the rectum, but the balloon-sac on Foley's tube did not pumped up with water. Measure and compare the value and change of SP in the portal vein during the RAIR. RESULTS: The comparison of SP in portal vein, among the 2, 4 centimeter groups with cut-off external splanchnic nerve, all groups with intact external splanchnic nerve supply and control group, had no statistic difference (P>0.05). The comparison between the 6 centimeter group with intact external splanchnic nerve group and the 2, 4 centimeter groups with cut-off external splanchnic nerve, P<0.01, the statistic difference was significant. The comparison between 6 centimeter group of intact and cut-off external splanchnic nerve, P<0.01, the difference was significant. CONCLUSION: The reason for the stimulation on upper rectum dose not induce the RAIR is related with this stimulation result in the release of SP, the exciting mediator to internal sphincter. The external splanchnic nerve supply of rectum and anal canal have influence on the change of SP of the portal vein during RAIR.

Study on conformation interconversion of 3-alkyl-4-acetyl-3,4-dihydro-2H-1,4-benzoxazines from dynamic NMR experiments and ab initio density functional calculations.

Variable-temperature NMR experiments and ab initio density functional calculations were carried out to investigate the conformation interconversion of novel chiral 3-alkyl-3,4-dihydro-2H-benzo[1,4]oxazine derivatives. With CDCl3 as the solvent, the coalescence temperatures of H2, H3, H11, and H19 of product 1 are about 289, 304, 292, and 316 K, with the corresponding activation free energies at 58.0 +/- 6.7, 60.9 +/- 7.1, 58.3 +/- 6.8, and 59.6 +/- 6.9 kJ.mol(-1), respectively. When dimethyl sulfoxide (DMSO-d6) was used as the solvent, 1H and 13C NMR signals were completely assigned at 375 K. The effects of solvent and temperature were investigated through a polarizable continuum model. At each theoretical level (MP2 or B3LYP), the changing tendencies of the calculated activation free energies and interconversion rates agree well with those of the NMR results. In addition, the interconversion rate at each specified temperature was calculated to be about 1.5 times faster in DMSO-d6 than in CDCl3. Accordingly, we failed to observe the coalescence phenomena of H3 and H19 in DMSO-d6 by NMR measurements from 296 to 375 K. The substitution effect at the R1-R5 positions was considered using density functional calculations, with the activation barriers decreasing as follows: product 6 > 3 > 1 > 7 > 2. This sequence is consistent with that of the reaction heats, except for product 7, implying that the interconversion processes may be thermodynamically controlled. Surprisingly, the substituted groups near the acetyl group in product 2 and 7 do not elevate the activation barrier but, instead, lower it somewhat, with the possible reasons for this provided in the paper.

Clinical evidence of growth hormone for patients undergoing abdominal surgery: meta-analysis of randomized controlled trials.

AIM: To assess the effectiveness and safety of perioperative growth hormone (GH) in patients undergoing abdominal surgery. METHODS: We searched the following electronic databases: MEDLINE, EMBASE, the Cochrane Controlled Trials Register, Chinese Bio-medicine Database. The search was undertaken in February 2003. No language restrictions were applied. Randomized controlled trials (RCT) comparing GH with placebo in patients undergoing abdominal surgery were extracted and evaluated. Methodological quality was evaluated using the Jadad scale. RESULTS: Eighteen trials involving 646 patients were included. The combined results showed that GH had a positive effect on improving postoperative nitrogen balance (standardized mean difference (SMD) = 3.37, 95%CI (2.46, 4.27), P<0.00001), and decreasing the length of hospital stay (weighted mean difference (WMD) = -2.07, 95%CI (-3.03, -1.11), P = 0.00002), and reducing the duration of postoperative fatigue syndrome (SMD = -1.83, 95%CI (-2.37, -1.30), P<0.00001), but it could increase blood glucose levels (WMD = 0.91, 95%CI (0.56, 1.25), P<0.00001). CONCLUSION: GH for patients undergoing abdominal surgery is effective and safe, if blood glucose can be controlled well. Further trials are required with a sufficient size to account for clinical heterogeneity and to measure other important outcomes such as infection, morbidity, mortality, fluid retention, immunomodulatory effects, and tumor recurrence.

Application of dietary fiber in clinical enteral nutrition: a meta-analysis of randomized controlled trials.

AIM: To evaluate the effects of dietary fiber (DF) as a part of enteral nutrition (EN) formula on diarrhea, infection, and length of hospital stay. METHODS: Following electronic databases were searched for randomized controlled trials about DF: Chinese Biomedicine Database (CBM), MEDLINE, EMBASE and Cochrane Controlled Trials Register. RevMan 4.1 was used for statistical analysis. RESULTS: Seven randomized controlled trials with 400 patients were included. The supplement of DF in EN was compared with standard enteral formula in five trials. Combined analysis did not show a significant reduction in occurrence of diarrhea, but there were valuable results for non-critically ill patients. Combined analysis of two trials observing the infection also did not show any valid evidence that DF could decrease the infection rate, though the length of hospital stay was reduced significantly. CONCLUSION: Based on the current eligible randomized controlled trials, there is no evidence that the value of DF in the diarrhea can be proved. Though length of hospital stay was shortened by the use of DF, there is no available evidence in preventing infection by DF. Further studies are needed for evaluating the value of DF in EN.

Microwave solid-state synthesis of LiV(3)O(8) as cathode material for lithium batteries.

A novel and economical microwave route has been developed for the synthesis of electrochemically active LiV(3)O(8) material by using a domestic microwave oven. The heating behavior of the designed reaction system guided the preparation of LiV(3)O(8) at a suitable irradiation power (i.e. heating rate), reaction time, and temperature. At the lowest irradiation power, the conversion fraction of reactants was mainly controlled by reaction temperature. Characterization results of X-ray diffraction (XRD), Fourier transform infrared (FTIR) and Raman spectroscopy, scanning (SEM) and transmission (TEM) electron microscopy, and BET surface areas indicated that the phases of samples prepared by microwave and traditional methods were in good agreement. Nevertheless, the crystallinity, crystallite configuration, and morphology of the samples were different, and were affected by the irradiation time and power. A floppy superposition structure of nanosheets (the size of one nanosheet was about 4.5 microm x 1.2 microm x 3 nm) was preferentially grown at the lowest irradiation power, and this effect on structure was more in evidence as the nanorods formed at the highest irradiation power. Electrochemical studies on ionic conductivity, electrochemical impedance spectroscopy (EIS), and charge-discharge capacity were carried out. It was found that the conductivity, first discharge capacity, and cycle performances of the samples were affected by the crystal size, crystallinity, and crystal configuration and defection concentration. The sample L30 prepared at the lowest irradiation power and the shortest time (30 min) showed the highest discharge capacity (335 mAh/g), but its discharge capacity decreased rapidly. By comparison, the sample L100 had a floppy superposition structure of nanosheets and a high surface area, provided a good two-dimensional channel for the transition of Li(+) ions, and was stable during the intercalation/deintercalation process of Li(+) ions, therefore the high ionic conductivity, high discharge capacity, and good cycle performance were presented. The relationship between the electrochemical properties and the irradiation power was discussed.

Heating behavior and crystal growth mechanism in microwave field.

A simple microwave solid-state reactor was designed on the basis of a domestic microwave oven by using graphite powder as heating medium. The heating behavior of the reactor was studied by using an on-line computer to monitor the real-time temperature during irradiation. It was found that the temperature (T) was related to the time (t) and that microwave power depended on the duty cycle (x) of microwave irradiation. Two empirical equations were proposed and could be applied to the similar microwave solid-state reactors. Four inorganic layered materials, LiV(3)O(8), KNb(3)O(8), KTiNbO(5), and KSr(2)Nb(3)O(10), were successfully synthesized in the designed reactor at a suitable heating rate and temperature that were fully controlled by the empirical equations. Characterization results of X-ray diffraction (XRD), Fourier transform infrared (FTIR) and Raman spectroscopy, and scanning (SEM) and transmission (TEM) electron microscopy indicated that the phases of samples prepared by traditional and microwave methods were in good agreement; nevertheless, the heating nature and the morphologies of products were quite different. The samples synthesized in the microwave field had crystallographic defects and showed an incompactly stacking structure of nanosheets. Due to the rapid formation of crystallites and different extended growth rate along the crystal axis of the products in microwave field, the crystal growth mechanism of layered metal oxides was not according to that of the traditional method and is briefly discussed.

Clinical evidence of growth hormone, glutamine and a modified diet for short bowel syndrome: meta-analysis of clinical trials.

This study assessed the safety and efficacy of growth hormone (GH) and glutamine (GLN) combined with a modified (high-carbohydrate-low-fat, HCLF) diet in patients with short bowel syndrome. A meta-analysis of all the relevant clinical trials was performed. Clinical trials were identified from the following electronic databases: MEDLINE, EMBASE, the Cochrane Controlled Trials Register, Chinese Bio-medicine Database. The search was undertaken in May 2004. Language was restricted to Chinese and English. Literature references were checked at the same time. Clinical trials were extracted and evaluated by two reviewers independently of each other. The statistical analysis was performed by RevMan4.2 software which was provided by the Cochrane Collaboration. A P value of < 0.05 was considered statistically significant. Thirteen trials involving 258 patients were included. The combined results showed that GH, GLN and HCLF diet had positive treatment effect on body weight (weighted mean difference [WMD] = 2.44, 95%CI [1.62, 3.27], P<0.00001), stool output (WMD = -376.49, 95%CI [-600.35, -152.63], P=0.001), lean body mass (WMD = 2.16, 95%CI [0.91, 3.41], P=0.0007), absorption of carbohydrates (WMD = 6.21, 95%CI [5.27, 7.15], P< 0.00001), absorption of nitrogen (WMD = 10.83, 95%CI [5.22, 16.44], P=0.0002), absorption of D-xylose (WMD = 0.37, 95%CI [0.29, 0.44], P<0.00001), and off TPN (total parenteral nutrition) (odds ratios [OR] = 64.63, 95%CI [15.51, 269.22], P<0.00001). But there were no improvements in fat mass (WMD = -1.50, 95%CI [-3.48, 0.48], P=0.14), absorption of energy (WMD = 7.48, 95%CI [-7.22, 22.17], P=0.32), and absorption of fat (WMD = 7.16, 95%CI [-2.95, 17.28], P=0.17). Most patients had side effects that are known to occur during treatment with high doses (0.14 mg/kg/day) of GH. No serious adverse effects occurred during active treatment with low doses (< or =0.1 mg/kg/day) of GH. Treatment with a combination of low-dose GH, GLN and HCLF diet is effective without any major adverse effects in patients with short bowel syndrome. Further trials are required, especially in children, with sufficient size and rigorous design.

Combined effect of SLS and (SBE)(7M)-beta-CD on the solubilization of NSC-639829.

Complexation and micellization are two effective ways of solubilizing drugs. In this study, the combined effect of surfactant and complexant on the solubilization of a poorly water soluble compound (NSC-639829) is investigated. With increasing concentration of sodium lauryl sulphate (SLS) in solutions of fixed concentration of (SBE)(7M)-beta-CD, the total solubility of the drug decreases linearly, reaches a minimum and then increases linearly. At each minimum, the molar ratio of SLS to (SBE)(7M)-beta-CD is close to unity. The above observation is attributed to the fact that the surfactant molecule competes with the drug to "fit" in the non-polar cyclodextrin cavity. The surfactant depletes cyclodextrin to form a 1:1 complex. Once the concentration of free SLS reaches the CMC, it starts forming micelles and hence, solubilizes the drug. A slight decrease of the solubilizing power is noticed in the presence of SLS/(SBE)(7M)-beta-CD complex. The combined use of two solubilizing agents, a surfactant and a complexant, results in a much lower solubility than when either one is used alone at the same concentration. The surfactant molecule acts as a competitive inhibitor in the solubilization of the drug by the complexant. Similarly the complexant "pulls" the surfactant out of solution, making it unavailable for solubilizing the drug.

Development of a decision support model for scheduling clinical studies and assigning medical personnel.

Clinical studies for the development of new drugs in the pharmaceutical industry consist of a number of individual tasks which have to be carried out in a pre-defined chronological order. Each task requires certain types of medical personnel. This paper investigates the scheduling of clinical studies to be performed during a short-term planning horizon, the allocation of workforce between the studies, and the assignment of individual employees to tasks. Instead of developing a complex monolithic decision model, a hierarchical modelling approach is suggested. In the first stage, a compact integer optimization model is solved in order to determine the start-off times of the studies and the required staffing while taking the limited availability of personnel into account. The objective is to minimize total staffing costs. The assignment of individual employees to tasks is then made in the second stage of the procedure using a binary optimization model.
