The topic of this review is first to analyze in normal conditions the signal transduction pathways induced by members of the HER-ErbB receptor family and their ligands, and second, to decipher some deregulations occurring in various cancer types. As a result, new therapeutic opportunities will be mentioned.
The proteasome degradation system is one of the most efficient proteolytic process in living cells. We will focus here on a new compound, called PS341, which is the first proteasome inhibitor entering clinical phases. First results about its use in multiple myeloma or leukemic cells will be discussed.
The vascular endothelial growth factors (VEGF) plays major roles during tumor development. VEGF expression is regulated by the transcription factor HIF (hypoxia inducible factor) which is very sensitive to oxygene variations. We will show here that HIF1alpha is today a good prognostic and diagnostic marker for an increasing number of cancers from various origins. Furthermore, the reduction of the expression or the activity of HIF1alpha impairs tumour angiogenesis and subsequently growth. Altogether, these results suggest that HIF1alpha could be an interesting target in cancer therapy.
