Inducible nitric oxide synthase expression in the ischemic core and penumbra after transient focal cerebral ischemia in mice.

The present observations examined the hypothesis that the iNOS expression in the ischemic penumbra after a transient focal ischemic insult is involved in the recruitment of penumbra into infarction. The middle cerebral artery in mice was occluded for 2 h by an intraluminal filament and then recirculated. The measurement of iNOS activity, iNOS protein formation and NO concentration in the ischemic core and penumbra, and the determination of infarct volume were performed at 6, 12, 24 and 48 h after reperfusion. iNOS protein and iNOS enzymatic activity appeared at 6 h, peaked at 24 h, and declined at 48 h in the penumbra after reperfusion. iNOS protein was not detectable in contralateral area and in sham-operated brains. The time course of iNOS protein, enzymatic activity and NO concentration in the penumbra but not in the core matched the process of infarct maturation. Treatment with iNOS inhibitor aminoguanidine (100 mg.kg(-1), i.p.) at 6 and 12 h after reperfusion inhibited iNOS activity by 88.0 +/- 10.4% and reduced NO concentration by 48.5 +/- 8.3% in the penumbra, and lessened infarct size by 48.8 +/- 7.2%. The iNOS activity and NO level in the core were not affected by the administration of aminoguanidine. These results suggest that iNOS expression in the ischemic penumbra is involved in the recruitment of penumbra into infarction and thereby contributing to the enlargement of infarct.

Inhibition of telomerase activity and bcl-2 expression in berbamine-induced apoptosis in HL-60 cells.

The present study was aimed to investigate the effect of telomerase activity in berbamine-induced apoptosis and the regulation of B cell leukemia/lymphoma 2 ( bcl-2) gene expression in human leukemia HL-60 cells. Apoptosis of HL-60 cells was induced by berbamine (10 microM) for 3, 6, 12 and 24 h. Apoptosis and bcl-2 were determined by flow cytometry analysis. A polymerase chain reaction-based telomeric repeat amplification protocol assay was used to detect the telomerase activity. Berbamine induced growth arrest and apoptotic cell death in HL-60 cells. The telomerase activity was inhibited in a time-dependent manner during the berbamine-induced apoptosis of HL-60 cells, and the expression of bcl-2 was progressively down-regulated by berbamine. Inhibition of the telomerase activity of HL-60 cells was closely related to the berbamine-induced apoptosis. The present results indicate that inhibition of telomerase and reduced bcl-2 gene expression may play a role in the berbamine-induced apoptosis of HL-60 cells.

Daidzin protects PC12 cells from serum deprivation-induced apoptosis.

This article examines the effect of daidzin on PC12 cell apoptosis induced by serum-free medium. PC12 cell survival was measured by MTT assay. The DNA content and percentage of apoptosis were monitored by flow cytometry and DNA fragmentation was analyzed by agarose gel electrophoresis. The results showed that serum-free (12 h) medium induced apoptosis in PC12 cells. When the cells had been treated with daidzin (0.1, 1 microM) for 12 h, the percentage of PC12 cell apoptosis was significantly decreased to 12.21 and 4.24% from 91.94% in the group with serum deprivation, and DNA fragmentation was prevented. Daidzin (0.01-10 microM) attenuated the cytotoxic effect of sodium cyanide (20 mM), glutamate (0.5 mM) and sodium nitroprusside (0.5 mM) in a manner dependent on concentration. The results suggested that daidzin prevented PC12 cell from serum free-induced apoptosis.

Cyclosporin A enhanced protection of nimodipine against brain damage induced by hypoxia-ischemia in mice and rats.

AIM: To study whether P-glycoprotein (P-gp) inhibitor cyclosporin A (CsA) enhanced the protection of nimodipine (NMD) against brain damage. METHOD: (1) After mice were given ip NMD alone or co-administration of CsA, survival time of mice were recorded following decapitation and ip injection of NaNO2, respectively. (2) After rats were given ip NMD alone or co-administration of CsA, 20 min forebrain ischemia induced by the technique of two-carotid occlusion plus hypovolemic hypotension. Following reperfusion of 1 h, the content of malondialdehyde (MDA), lactic acid (LA) and activity of lactic dehydrogenase (LDH) in cortex tissue were measured. (3) NMD level in brain was also determined after ip injection of NMD 2 mg/kg alone and co-administration of CsA, respectively. RESULTS: NMD showed potent pharmacological activity in the three models. The survival time of mice by decapitation and ip NaNO2 were significantly (P <0.05) prolonged after co-administration of CsA. In rat forebrain ischemia/reperfusion, levels of MDA, LA, and LDH by co-administration of CsA were greatly modified, compared with those of NMD alone group. The level of NMD in rat brain was increased markedly after co-administration of CsA. CONCLUSION: P-gp inhibitor CsA may enhance the protection of NMD against brain damage.

Interaction of multidrug resistance reversal agents with P-glycoprotein ATPase activity on blood-brain barrier.

AIM: To gain further insights into the mechanism of the ATP-dependent interaction of P-glycoprotein (P-gp) with various multidrug resistance (MDR) reversal agents. METHODS: Bovine brain capillary endothelial cells (BCEC) were isolated from cerebral gray matter using modifications of the mechanical homogenization technique. Plasma membranes were prepared from BCEC. The P- gp adenosine triphosphatase (ATPase) activity of the isolated BCEC membranes was estimated by measuring inorganic phosphate liberation. RESULTS: The basal P-gp ATPase activity was increased by verapamil (Ver), vincristine (VCR), doxorubicin (Dox), tetrandrine (Tet), dauricine (DRC), berbamine (BBM), and daurisoline (DRS), with respective half-maximal activity concentrations Km of about 17, 5.9, 41, 2.3, 11, 23, and 22 micromol/L. Berberine (BBR) produced a relatively slight activation. dl-Tetrahydropalmatine (dl-THP) and l-tetrahydropalmatine (l-THP ) does not alter the basal P-gp ATPase activity. Cyclosporin A (CsA) inhibited both the basal and the drug-stimulated ATPase activity of P-gp with high affinity. Kinetic analysis indicated a competitive inhibition of Ver- or VCR-stimulated ATPase activity and a noncompetitive inhibition of Dox- or Tet-activated ATPase activity by CsA. Moreover, Dox inhibited Tet-activated P-gp ATPase activity in a noncompetitive manner. CONCLUSION: Various MDR reversal agents could interact with P-gp and alter its ATPase activity in different manners. This is the result of the b road molecular recognition specificity of P-gp. CsA, Ver, and VCR could bin d P-gp either on overlapping sites or distant but interacting sites, while CsA, Dox, and Tet could independently bind P-gp on separated sites on blood-brain barrier.

Effects of various principles from Chinese herbal medicine on rhodamine123 accumulation in brain capillary endothelial cells.

AIM: To search for novel effective P-glycoprotein (P-gp) reversal agents in the blood-brain barrier (BBB). METHODS: Using rhodamine123 (Rh123) to examine the functional activity of P-gp in cultured bovine brain capillary endothelial cells (BCEC) and screen various principles on P-gp modulation in BBB. RESULTS: All of tested compounds (1-10 micromol/L) increased the intracellular accumulation of Rh123 in a concentration-dependent manner. The rank order of these agents in increasing Rh123 accumulation in BCEC was: cyclosporin A (CsA) > tetrandrine (Tet) > vincrinstine (VCR) approximate, equals flunarizine (Flu) > dl-tetrahydropalmatine (dl-THP) > dauricine (DRC) > azithromycin (Azi) > verapamil (Ver) approximate, equals berbamine (BBM) > daurisoline (DRS) > berberine (BBR) approximate, equals doxorubicin (Dox) > l-tetrahydropalmatine (l-THP) > tetramethylpyrazine (TMP). These agents at concentration of 10 micromol/L increased Rh123 accumulation by 346 %, 203 %, 136 %, 129 %, 115 %, 103 %, 92 %, 87 %, 81 %, 75 %, 67 %, 67 %, 63 %, and 54 %, respectively. The effects of CsA, Tet, Ver, Flu, Azi, and dl-THP on cellular accumulation of Rh123 in BCEC were reversible. When CsA, Tet, Ver, Flu, Azi, or dl-THP-pretreated BCEC were examined at 48, 36, 24, 36, 36, or 12 h, respectively, after removing the agent, the amount of cellular Rh123 accumulation in BCEC returned to control levels (no drug treatment). CONCLUSION: The functional activity of P-gp on the blood-brain barrier could be modulated by various MDR-reversing agents and some principles with low toxicity extracted from medicinal herbs, such as some isoquinoline alkaloids without permanent modifying effects on the intrinsic level of P-gp function.

[Genetic analysis and molecular tagging of a gene for non-palea in rice].

The mutants involved in the development of floral organ are good material for understanding the molecular genetic mechanisms of floral development. A rice mutant, that lacks palea in its florets, was derived from a spontaneous mutation in an indica line, SAR III-93-369. Genetic analyses in three F2 populations from the mutant crossed with three rice varieties, Sheng 47, N625 and CDR22, respectively, showed that the mutant trait is controlled by a single recessive gene. In the F2 population from npa-1/Sheng47 the gene for the non-palea trait was mapped between two restriction fragment length polymorphism markers, C498 and RZ450, with distances of 7.5 cM and 2.4 cM, respectively. The tagged recessive non-palea gene is temporarily designated npa-1.

[Mapping of two fertility-restoring gene for WA cytoplasmic male sterility in minghui63 using SSR markers].

F2 population derived from Shanyou63, F1 hybrid developed from the cross Zhenshan97 A/Zhenshan97B, was used in this study. Fertile bulk was constructed by polling equal amount of 15 highly fertile lines. Sterile bulk was obtained by pooling equal amount of 15 highly sterile lines. Minghui63 and Zhenshan97A, parents of Shanyou63, were analyzed with 302 pairs of SSR primers. 244 pairs of primers gave amplification products, of which 58 pairs of primers on 12 different chromosomes showed polymorphism between the two parents with polymorphic frequency up to 23.77%. Gene bulks were further assayed with the 5 pairs of primers. RM1 on chromosome 1 and RM258, RM304 on chromosome 10 was found to be polymorphic between the two gene bulks. In theory, there should be no difference detected between the two gene bulks except for the target traits governed by fertility-restoring genes. RM1, RM258 and RM304 were probably related to the restorer genes. Ten highly fertile and ten highly sterile lines were selected from F2 population of Shanyou63 to screen the gene bulks. The results showed that specific bands of Minghui63 were detected in all ten highly fertile lines while not observed in all the sterile lines. It indicated that the three SSR markers might be linked to fertility-restoring genes. Dominant lines were not selected due to their inalbility to distinguish recombinant lines from non-recombinant lines. Pure recessive lines were chosen to conduct mapping analysis. A total of 53 highly sterile lines were selected from 900 lines of Shanyou63 F2 population to estimate the genetic distance between three SSR markers and fertility-restoring genes respectively. The results demonstrated that recombination occurred in 2, 3, lines with RM1 and RM258 while no one with RM304. Using MAPMAKER/EXP 3.0, the genetic distance between RM1, RM258, RM304 and the related restorer genes were calculated as 1.9, 2.9 and 0.0 cM, respectively. It is possible that the fertility restoring gene(s) on chromosome 10 for three different types of cytoplasmic male sterility(WA, BT and HL) are of the same, or belong to a gene family.
