Effectiveness of intra-arterial anesthesia for uterine fibroid embolization using dilute lidocaine.

A modified protocol of uterine fibroid embolization (UFE) is proposed for alleviating the postinterventional pain. This randomized and double-blinded clinical trial is to evaluate the effectiveness of intra-arterial infusion of dilute lidocaine for postinterventional pain relief in UFE. Forty-six patients who underwent UFE were randomly grouped equally. In the test group, after the poly(vinyl alcohol) embolization was complete, a dilute lidocaine solution with 40 mg in 6.0 ml, 3.0 ml for each side or 4.0 and 2.0 ml for two sides, was given through the catheter. In the control group, the patients received 6.0 ml of saline solution as a placebo. A simple pain degree classification method for patient self-evaluation was developed. A questionnaire was completed by each patient to record the degree of pain during five periods; these were during the procedure, the first 12 h, the second 12 h, between 24 and 48 h, and between 48 and 72 h. The numbers of patients with the same degree of pain in the five time segments from the two groups were statistically compared. Compared with the control group, the patients in the test group experienced less pain within 48 h after the procedure (p<0.01). The results suggest that this improved UFE protocol is a simple approach to prevent the acute postinterventional pain of UFE.

Chemical- and irradiation-induced mutants of indica rice IR64 for forward and reverse genetics.

IR64, the most widely grown indica rice in South and Southeast Asia, possesses many positive agronomic characteristics (e.g., wide adaptability, high yield potential, tolerance to multiple diseases and pests, and good eating quality,) that make it an ideal genotype for identifying mutational changes in traits of agronomic importance. We have produced a large collection of chemical and irradiation-induced IR64 mutants with different genetic lesions that are amenable to both forward and reverse genetics. About 60,000 IR64 mutants have been generated by mutagenesis using chemicals (diepoxybutane and ethylmethanesulfonate) and irradiation (fast neutron and gamma ray). More than 38,000 independent lines have been advanced to M4 generation enabling evaluation of quantitative traits by replicated trials. Morphological variations at vegetative and reproductive stages, including plant architecture, growth habit, pigmentation and various physiological characters, are commonly observed in the four mutagenized populations. Conditional mutants such as gain or loss of resistance to blast, bacterial blight, and tungro disease have been identified at frequencies ranging from 0.01% to 0.1%. Results from pilot experiments indicate that the mutant collections are suitable for reverse genetics through PCR-detection of deletions and TILLING. Furthermore, deletions can be detected using oligomer chips suggesting a general technique to pinpoint deletions when genome-wide oligomer chips are broadly available. M4 mutant seeds are available for users for screening of altered response to multiple stresses. So far, more than 15,000 mutant lines have been distributed. To facilitate broad usage of the mutants, a mutant database has been constructed in the International Rice Information System (IRIS; http: //www.iris.irri.org) to document the phenotypes and gene function discovered by users.

GmCOI1, a soybean F-box protein gene, shows ability to mediate jasmonate-regulated plant defense and fertility in Arabidopsis.

The F-box protein gene COI1 from Arabidopsis plays a fundamental role in response to jasmonates, which regulate plant root growth, pollen fertility, wounding and healing, and defense against pathogens and insects. Null mutations in COI1 were previously found to abolish all the jasmonate responses, and the Arabidopsis coil-1 mutant is male sterile and susceptible to pathogen infection. In this study, we isolated an F-box protein gene from soybean, which shares significant homology with the Arabidopsis COI1 and similarly contains an F-box motif and leucine rich repeats (LRR), here designated GmCOI1 (Glycine max L. (Merr.) COI1). To test whether the sequence homology and structural similarity are indicative of functional conservation, we expressed GmCOI1 in the Arabidopsis coil-1 mutant. The transgenic coil-1 plants with expression of the GmCOI1 gene were found to exhibit normal jasmonate responses, including jasmonate-regulated plant defense and fertility. In addition, the chimerical proteins with swapped domain of the F-box motif or LRR between GmCOI1 and COI1 were shown to functionally complement the coil-1 mutation. Furthermore, GmCOI1 was found to assemble into the Skpl-Cullin-F-box (SCF) complexes, similar to the formation of the Arabidopsis SCF(COO1). These data demonstrate the soybean F-box protein gene GmCOI1 is able to mediate jasmonate-regulated plant defense and fertility in Arabidopsis, which implies a generic jasmonate pathway with conserved signal components in different plant species.

[Bacterial flora composition and its dynamics in tidal-flat Sinonovacula constricta aquaculture area].

In this study, the bacteria from the mud in tidal-flat Sinonovacula constricta aquaculture area were isolated each month from March to December, 2002, and the temporal and spatial distribution of heterotrophic bacteria, ammonifying bacteria, denitrifying bacteria, and sulphate reducing bacteria were analyzed. The results showed that all the 515 isolated bacteria mainly belonged to 1 family and 13 genera. The bacterial flora in different layers of the mud was almost consistent, while the composition was different. The predominant genera were Clostridium, Bacillus, Corynebacterium, Photobacterium, and some Enterobacteriaceae. The number of heterotrophic bacteria in the surface layer and the bottom fluctuated in 7.6 x 10(3) cfu x g(-1) - 2.0 x 10(5) and 1.6 x 10(3) - 1.0 x 10(5) cfu x g(-1), ammonifying bacteria fluctuated in 1.5 x 10(6) - 9.0 x 10(7) and 9.0 x 10(5) - 1.0 x 10(7) cfu x g(-1), denitrifying bacteria fluctuated in 9.0 x 10(3) - 4.0 x 10(6) and 5.0 x 10(2) -1.9 x 10(6) cfu x g(-1), and sulphate reducing bacteria fluctuated in 5.0 x 10(4) - 5.0 x 10(6) and 1.9 x 10(4) - 2.0 x 10(6) cfu x g(-1), respectively. The detection rates of ammonifying bacteria, denitrifying bacteria and sulphate reducing bacteria in the mud were all 100%, and these bacteria increased significantly in the second half of the year, indicating that the environment of the Sinonovacula constricta aquaculture area was deteriorated due to the accumulation of NH3, nitrite and H2S, and it is important to regulate the breed capacity and redistribute the breeding environment.

Optimization of off-null ellipsometry in sensor applications.

The optimization of azimuth angle settings of a polarizer-compensator-sample-analyzer off-null ellipsometric sensor system to obtain maximum intensity changes with respect to changes in the properties of a sensing layer, with and without considering changes in s reflectance, is studied. Optimal conditions in the two cases are derived analytically under the assumption that linear relationships exist among the changes in the parameters of the sensing layer. The validity of these optimal conditions is verified by numerical examples. The advantage of using ellipsometry compared with reflectometry to readout sensing information for some sensing samples is also discussed.

Optimization of azimuth angle settings in polarizer-compensator-sample-analyzer off-null ellipsometry.

The dependence of the azimuth angle settings on the change in off-null intensity of a polarizer-compensator-sample-analyzer ellipsometer owing to changes in sample properties is studied. First, a closed-form expression for the relationship between azimuth angles that fulfill the null condition is presented. An approximation for the off-null light intensity near null that is valid for small changes of the p- and s-reflection coefficients of an isotropic sample is then derived. This approximation shows that the intensity change near the null can be described by changes in the ellipsometric parameters tan phi and A only. Expressions for finding the azimuth angle that gives the maximum possible intensity change for a given change in the sample parameters are also derived. The importance of optimization of azimuth angle settings for different samples is investigated and found to depend on tan psi. Numerical and experimental results chosen from the investigation of gas sensors based on porous silicon are included to verify the approximations as well as the optimization.

[Detection of heterotrophic bacteria and vibrios in shoal of Bullacta exarata].

The number of heterotrophic bacteria and vibrios in shoal for Bullacta exarata rearing fluctuated mainly within 1.25 x 10(4)-7.21 x 10(4) cfu.g-1 and 0-1.08 x 10(4) cfu.g-1, respectively. 173 strains of bacteria, mainly belong to 13 genera were isolated. The predominant genera were composed of Bacillus, Photobacterium, Pseudomonas, Vibrio and some genera of Enterobacteriaceae. The number of bacteria changed in line with the alteration of the temperature, and was especially affected by the organic matter content after the use of pesticide. The predominant genera such as Bacillus were closely related with the environmental deterioration of rearing shoal.

[Dynamic expression of tenascin in rat liver during liver fibrogenesis induced by CCl(4)].

OBJECTIVE: To study the expression of tenascin in normal and fibrotic rat liver. METHODS: Liver fibrosis induced in rat with CCl(4) were divided into three stages: the stage of hepatic injury (4 weeks), early stage of hepatic fibrosis (8 weeks) and later stage of hepatic fibrosis (12 weeks). Tenascin expression in liver tissue was observed by immunohistochemical method and in situ hybridization using digoxigenin-labelled DNA probe. RESULTS: In normal rat liver there was a weak staining for tenascin. In both liver injury stage and early stage of hepatic fibrosis, both mRNA signal and immunostaining for tenascin were significantly increased as compared to that in normal liver. In later stage of hepatic fibrosis, mRNA signal and immunostaining for tenascin were decreased compared with that in early stage of hepatic fibrosis. The cellular source of tenascin in liver mainly restricted in mesenchymal cells. CONCLUSIONS: Tenascin is a component of the extracellular matrix of liver tissue. Plays a role in early matrix organization during liver fibrogenesis.

[Influence of ursodeoxycholic acid on the therapeutic effects of low-calorie diet in obesity and hyperlipidemia rats with steatohepatitis].

OBJECTIVE: To explore the influence of ursodeoxycholic acid on the therapeutic effects of low-calorie diet in steatohepatitis with obesity and hyperlipidemia. METHODS: Thirty-five Sprague-Dawley rats fed with high-fat diet for 10 weeks were randomly allocated into 3 groups, and continued to experiment for 2 weeks. The animals in model group (n = 10) were still fed with high-fat diet; low-calorie diet group (n = 10) with common diet but only one third of the amount of normal demand; ursodeoxycholic acid group (n = 15) with low-calorie diet and ursodeoxycholic acid (15 mg/kg.d(-1)); and another 9 rats with common diet for 12 weeks as normal group. RESULTS: Compared with normal group, such indexes as body weight, liver weight, and the level of serum lipids and aminotransferase were all increased significantly in model group. Furthermore, all rats in model group developed steatohepatitis. On the other hand, such indexes as body weight and the degree of steatosis in rats of low-calorie diet group were decreased sharply compared with those in model group, but neither disorders of serum lipid nor the degree of hepatic inflammation and necrosis in low-calorie diet group were improved obviously. Disorders of serum lipid, aspartate aminotransferase, hepatic inflammation and necrosis in ursodeoxycholic acid group were ameliorated to some extent. CONCLUSIONS: Ursodeoxycholic acid might help to improve the therapeutic effects of low-calorie diet on steatohepatitis with obesity and hyperlipidemia.
