[Immunomorphological and ultrastructural findings on the musculature in progressive scleroderma].

Muscle biopsies taken from 30 patients with generalized scleroderma were studied immuno-histologically as well as by light and electron microscopy. The immunological investigations have demonstrated that an increase of IgM and/or complement depositions in capillaries and in the walls of small arteries have been involved in most cases. Similar alterations were found in the subsarcolemmal zone. Vascular Ig-G precipitates were observed in the later stage of the disease only, whereas a positive reaction of fibrinogen have been proved in the most cases. By light and electron microscopy a variety of pathologic alterations have usually been found, but the most prominent changes involved were thickening of the capillary basement membrane and interstitial fibrosis. Scattered cellular infiltrates consisting of lymphocytes, histiocytes and plasmocytes were also seen either perivascularly or throughout the interstice. Around fibroblasts and capillaries, fine protocollagen fibrils and glycosaminoglycan filaments have been observed. They may partly represent fibrogenesis. Filamentous structures could be seen within the cytoplasm of numerous fibroblasts.

[Studies on the specificity of an antiserum against denaturated DNA from Sarcina maxima].

An antiserum against denatured DNA from Sarcina maxima (71% (A + T)) reacted in complement fixation tests with denatured DNA's of various sources. The serological activity of the antibodies is in correlation with the (adenine + thymine)-content of the DNA's used as antigens. Haptene inhibition tests demonstrate a preferable reaction of the antibodies with T2-sequences. The heterogeneity of DNA-antisera and the increasing specificity of such antisera, if DNA with high (A + T) or (G + C)-content served as immunogen, are discussed.

[Epidemiological and immunological study of human scabies].

The scabies epidemy being observed since 1965, had developed from persistent endemic centres and pursues continuous the cyclic course of the scabies humanis in periods of 15 to 20 years. The intracutaneous tests with antigens of Notoedres alepis show a cross antigenity with the antigens of Sarcoptes scabiei and a sensibility of patients suffering from scabies. These tests have a diagnostical value. The antigens of Notoedres alepis are free of antigens from human epidermis.--The results support the theory of immunity for the interpretation of the cyclic course of the scabies, whereas wars and social and hygienic nuisances only have a modifying influence, and promiscuity, Sarcoptotoxicity of environment and others have no deciding importance.

[The reactivity of lupus erythematosus serum with native and denatured DNA of various origins].

The serum of a female suffering from Lupus erythematosus visceralis was investigated by complement fixation for the reaction with native and denatured DNA's of various base composition. The reaction with native DNA is independent on the (G+C)-content of the DNA. It is apparent that the responsible antibodies react with determinants of the helical conformation, which are identical in the various DNA-molecules. Quantitative differences are found with denatured DNA's. The strongest complement fixation is observed with (G+C)-rich denatured DNA. The reaction with denatured DNA is only partially inhibited by DNA digest. These antibodies obviously react with sequential determinants containing bases. Therefore, they are induced by a different mechanism of sensitization.

[Studies on cross reactions of antisera against nucleotid-protein conjugates with denaturated desoxyribonucleic acids].

The reactivity of anti-nucleotide-sera (anti-UMP, anti-AMP, anti-GMP) with denatured DNA's of 10 different sources was investigated by means of quantitative complement fixation. The haptene specificity of the anti-nucleotide antibodies was shown by haptene inhibition of the reaction with DNA. Anti-UMP showed cross-reactions with other pyrimidine-derivatives, but to a lesser extent; anti-GMP cross-reacted with adenine structures. Quantitative differences were observed in the reaction of anti-UMP, anti-AMP and anti-GMP with DNA's of various (G + C)-contents. These differences could not be correlated in all cases with the overall base composition of the DNA.
