Mast cell degranulating peptide: a multi-functional neurotoxin.

This review discusses our present knowledge of the structure and activities of the mast cell degranulating peptide (MCDP). This peptide is a basic, 22 amino acid residue component of honey bee venom with striking immunological and pharmacological activities. MCDP is a potent anti-inflammatory agent, but at low concentrations it is a strong mediator of mast cell degranulation and histamine release. MCDP is also an epileptogenic neurotoxin, an avid blocker of the potassium channels and can cause a significant lowering of the blood pressure in rats. Some of the biological activities of MCDP appear to have distinct mechanisms and may represent a good illustration of the structure-function relationship.

Effect of partial discectomy on the stability of the lumbar spine. A study of kinematics.

The effect on the stability of the lumbar motion segment, as affected by partial discectomy, was investigated using three-dimensional motion measurement technique. Eight functional spinal units from fresh cadaver lumbar spines were potted and clinically relevant loads applied through a special loading frame attached rigidly to the top vertebra of the specimen. Partial discectomy was performed on the right side of the specimen. The three-dimensional motions of each lumbar motion segment, before and after partial discectomy, were recorded. The results showed that partial discectomy significantly affects the stability of lumbar spinal motion segments. The clinical relevance of this finding is discussed.

Congenital tuberculosis: a case report.

Congenital tuberculosis is a rare disease, difficult to be correctly diagnosed from clinical manifestations alone without maternal tuberculous history. In this article we report a neonate who exhibited abdominal distension and bloody diarrhea simulating neonatal sepsis and necrotizing enterocolitis. Exploratory laparatomy was performed for the markedly distended abdomen and deteriorated condition. Calcified lymph nodes over mesentery and spleen were found, and biopsy was done to show caseous tubercles. However, this neonate deteriorated rapidly and died one day after operation. Autopsy revealed generalized discrete miliary tubercles over the lungs, liver, spleen, pancreas, adrenal glands, thyroid and lymph nodes.

A rapid cold agglutinin test in Mycoplasma pneumoniae infection.

A definite diagnosis of Mycoplasma pneumoniae infection is currently based on cultural method or complement fixation test which is usually retrospective. A rapid cold agglutinin test was developed to determine its value on the early diagnosis of M. Pneumoniae infection. One hundred and thirty patients with pneumonia aged between 5 and 14 years were studied. Blood specimens from all the patients were collected for rapid cold agglutinin test, cold hemagglutination test, and complement fixation test. Thirty patients showed positive, rapid cold agglutinin test. All the patients with positive rapid cold agglutinin test had higher (greater than or equal to 1:32) cold agglutinin titers which were simultaneously performed. The rapid cold agglutinin test had 100% sensitivity and 97% specificity when a cut-off criterion was set at cold agglutinin titer greater than or equal to 1:64. Twenty-five of the 130 cases were serologically proven to have M. pneumoniae infection using complement fixation test or/and cold agglutinin titer. M. pneumoniae was a major cause (21/28) in cold agglutinin-positive pneumonic patients. The positive predictive value of the rapid cold agglutinin test is 70% (21/30). Only 28% (7/25) of the patients with M. pneumoniae infection were diagnosed at acute stage with serological method. We conclude that the rapid cold agglutinin test is of much value in the early detection of M. pneumoniae infection in office or hospital practice in children with pneumonia.

Deletions and insertions within an amino-terminal domain of pp60v-src inactivate transformation and modulate membrane stability.

We previously showed (V. W. Raymond and J. T. Parsons, Virology 160:400-410, 1987) that variants of the Prague A strain of Rous sarcoma virus containing large deletions impinging on a region of the src gene encoding amino acid residues 143 to 169 were defective for transformation of chicken cells in culture. Here we report that introduction of small (tri-and tetrapeptide) deletions into a region of pp60v-src containing amino acid residues 155 to 175 was found to inactivate transformation. In addition, insertion of four, but not one, amino acid residues at position 161 also inhibited transformation. Biochemical analysis of the src proteins encoded by individual transformation-defective variants revealed that the structural alterations introduced into this domain had only marginal effects upon src tyrosine-specific protein kinase activity. However, the src proteins encoded by defective variants exhibited a significantly shorter half-life within the cell, although these proteins efficiently and rapidly associated with cellular membranes. Our results suggest that the structural domain encompassing residues 155 to 177 may influence the stability of pp60src in the cellular membrane, possibly via the interaction of src with a cellular membrane component(s) or substrate(s).

Stable association of activated pp60src with two tyrosine-phosphorylated cellular proteins.

We have identified two phosphotyrosine-containing cellular proteins with relative molecular masses of 130,000 (pp130) and 110,000 (pp110) daltons in chicken embryo cells that coimmunoprecipitated with pp60v-src and activated forms of chicken pp60c-src (pp60(527)F). Most if not all of the tyrosine-phosphorylated forms of pp130 and pp110 could be immunoprecipitated from lysates with any of several src protein-specific monoclonal antibodies directed against at least three spatially distinct epitopes. Consequently, of the more than 15 prominent phosphoproteins detected on immunoblots with phosphotyrosine-specific antibodies, pp130 and pp110 were selectively removed by src protein-specific immunoprecipitation, and their presence in the immunoprecipitates appears to have been due to a direct interaction with activated src proteins. src protein variants that induce different morphological phenotypes were altered in their ability to form detergent-stable complexes with pp130 and pp110 or with pp110 alone. Mutant src proteins, defective for myristylation, showed increased tyrosine phosphorylation of and association with pp110. Expression of src variants with mutations in the A box (pp60dl92/527F) or B box (pp60dl155/527F) of the src homology region induced differences in phosphorylation of pp130 and pp110, as well as changes in their association with variant src proteins. Sequences within the B-box region appeared to be necessary for stable complex formation with pp130 and pp110 and may be involved in the interaction of activated src proteins with cellular substrates.

Characterization of a heat-resistant strain of Tilapia ovary cells.

Tilapia ovary cells (TO-2) cease to proliferate when moved from normal growth temperature of 31 degrees C to 37 degrees C, and arrest in G1 and G2 phases of the cell cycle. The ability of the arrested cells to re-enter the cell cycle when restored to 31 degrees C decreases inversely with time spent at 37 degrees C. A heat-resistant strain, TO-37c, cloned from the surviving fraction of TO-2 after heat treatment, has been found to re-enter the cell cycle with greater facility and to have a higher rate of survival. TO-37c cells have a smaller cell volume than TO-2 and show a distinct morphology at 37 degrees C. Most of the heat-shock proteins (hsps) induced on temperature change were similar, but in TO-37c the decline in the synthesis of a 27 X 10(3) Mr hsp was faster and a 37 degrees C-specific 60 X 10(3) Mr hsp was missing. Ultraviolet (u.v.) sensitivity was slightly affected if heat treatment was given after irradiation. However, when cells were preheated and then u.v. irradiated, the u.v. sensitivity increased sharply for TO-2 cells but not for TO-37c.

Production of transgenic mice carrying the human hepatitis B virus or the human papillomavirus DNA sequences in Taiwan: analysis of physical structure and hereditary mode.

In order to produce suitable animal model systems for the analysis of possible synergistic interaction between hepatitis B virus (HBV) or the human papillomavirus (HPV) with other viral or chemical co-factors, we have produced transgenic mice carrying HBV, HPV16 or HPV18. Transfer of 3343 micro-injected one-cell (C57/BL6 X CBA) F1 embryos into pseudo-pregnant ICR recipients produced 181 pups. Seventeen of 151 of the pups analysed were found to be transgenics. Southern blot and slot blot analysis showed that the copy number of the transgenome in the transgenic mice ranged from 1-50 copies per cell. All the transgenomes analysed appeared to be in tandem repeats without detectable sequence rearrangements. The transgenome sequences were also stable on transmission to the F3 generation. On breeding, six of the ten transgenic mice analysed showed the Mendelian mode of inheritance with integration at single or multiple loci. Of the remaining four mice, three were mosaics and one was sterile. Analysis of the gene expression of the transgenic mice is currently in progress.

Investigations of the partial reactions catalyzed by pyruvate phosphate dikinase.

The kinetic mechanism of pyruvate phosphate dikinase (PPDK) from Bacteroides symbiosus was investigated with several different kinetic diagnostics. Initial velocity patterns were intersecting for AMP/PPi and ATP/Pi substrate pairs and parallel for all other substrate pairs. PPDK was shown to catalyze [14C]pyruvate in equilibrium phosphoenolpyruvate (PEP) exchange in the absence of cosubstrates, [14C]AMP in equilibrium ATP exchange in the presence of Pi/PPi but not in their absence, and [32P]Pi in equilibrium PPi exchange in the presence of ATP/AMP but not in their absence. The enzyme was also shown, by using [alpha beta-18O, beta, beta-18O2]ATP and [beta gamma-18O, gamma, gamma, gamma-18O3]ATP and 31P NMR techniques, to catalyze exchange in ATP between the alpha beta-bridge oxygen and the alpha-P nonbridge oxygen and also between the beta gamma-bridge oxygen and the beta-P nonbridge oxygen. The exchanges were catalyzed by PPDK in the presence of Pi but not in its absence. These results were interpreted to support a bi(ATP,Pi) bi(AMP,PPi) uni(pyruvate) uni(PEP) mechanism. AMP and Pi binding order was examined by carrying out dead-end inhibition studies. The dead-end inhibitor adenosine 5'-monophosphorothioate (AMPS) was found to be competitive vs AMP, noncompetitive vs PPi, and uncompetitive vs PEP. The dead-end inhibitor imidodiphosphate (PNP) was found to be competitive vs PPi, uncompetitive vs AMP, and uncompetitive vs PEP. These results showed that AMP binds before PPi. The ATP and Pi binding order was studied by carrying out inhibition, positional isotope exchange, and alternate substrate studies.(ABSTRACT TRUNCATED AT 250 WORDS)

Feeding and drinking patterns of ovariectomized old female rats.

Circadian rhythms and patterns of feeding and drinking behavior of ovariectomized old female rats (23 to 34 months of age) were studied and the changes due to the aging process were analyzed. In order to assess sex difference in these patterns in old age, the data were also compared with the previous studies in old male rats. There was significant increase in daily feeding events but not in licking events in old female rats. Circadian rhythms disappeared in 2 out of 7 old female rats in feeding events and 1 out of 8 old female rats in licking events. The remaining old rats which still showed circadian rhythmicity in feeding events and licking events, had smaller meal number/day, larger mean intermeal interval, quicker feeding events/min/meal, larger draft number/day, shorter mean draft duration, longer mean interdraft interval and quicker licking events/min/draft than young female rats. Drinking/feeding ratio was higher in the dark period than the light period in young as well as old female rats. These changes resemble those in old male rats and in ventromedial hypothalamic lesioned rats in the obese stage, but to less extent.

The development of ACTH-like substance during tadpole metamorphosis.

The interrenals of tadpoles of Rana catesbeiana at various developmental TK stages were frozen, sectioned, and prepared histochemically for assessment of delta 5-3 beta-hydroxysteroid dehydrogenase (delta 5-3 beta-HSD) activity. The interrenals of hypophysectomized (HX) tadpoles from the same batch of eggs as control tadpoles were prepared similarly for delta 5-3 beta-HSD activity following a series of intraperitoneal injections of different concentrations of mammalian ACTH. The histochemically stained sections of control and HX tadpoles were scanned with a computerized microscope spectrophotometer for diformazan absorbance. Two regression equations were obtained: Y = 0.1460 + 0.0015X and Y' = 0.1494 + 0.0893X', where Y or Y' = delta 5-3 beta-HSD activity absorbance (for the control and HX tadpoles, respectively), X = developmental stages of control, and X' = exogenous ACTH dosage administered to the HX tadpoles. Taking absorbance of delta 5-3 beta-HSD activity as the common factor, a curve of concentrations of ACTH-like substance against TK stages was made. The developing concentration of ACTH-like substance was very low at first and then rose during metamorphic climax. The trend of the development of ACTH-like substance was discussed from the view point of thyroid and delta 5-3 beta-HSD activities.
