Influence of K-ras activation on the survival responses of Caco-2 cells to the chemopreventive agents sulindac and difluoromethylornithine.

The nonsteroidal anti-inflammatory drug sulindac and the ornithine decarboxylase inhibitor difluoromethylornithine (DFMO) are both potent inhibitors of colon carcinogenesis in experimental models of this disease. The combination of these two agents is undergoing evaluation as a strategy for colon cancer chemoprevention in humans with resected colon polyps. We evaluated the effects of the major sulfide and sulfone metabolites of sulindac and DFMO alone, or in combinations, on the growth and survival of Caco-2 colon cancer-derived cells and in clones of these cells transfected with an activated K-ras oncogene. Both the sulfide and sulfone metabolites of sulindac reduced cell viability, measured by colony-forming assays, primarily by inducing apoptosis. Expression of an activated K-ras oncogene caused cells treated with either sulindac sulfide or sulfone to undergo apoptosis earlier than nontransfected controls. However, clonogenic survival, measured 2 weeks after drug treatment, was the same in both Caco-2 and ras-transfected Caco-2 cells treated with sulindac metabolites. A 24-h treatment with DFMO caused a dose-dependent decrease in the colony-forming ability of cells expressing an activated K-ras but had no effect on the viability of the parental Caco-2 cells. The DFMO-dependent decrease in colony formation in K-ras-activated cells occurred in the absence of apoptosis. Assessment of cell survival by colony-forming assays indicated that these two agents acted in an additive manner when combined. These data indicate that K-ras can influence the kinetics of apoptosis induction by sulindac metabolites and cell survival in response to DFMO. However, cytotoxicity induced by these agents occurs via unique mechanisms. These studies suggest that the combination of DFMO and sulindac may be useful in human cancer prevention strategies.

The prevalence of lactase deficiency and lactose intolerance in Chinese children of different ages.

OBJECTIVE: To determine lactose metabolism and lactase activity in Chinese children of different ages, prevalence of lactase deficiency (LD), and lactose intolerance (LI). METHODS: All 1168 healthy subjects between 3 and 13 years were recruited from schools in four large cities in China. They were screened by a 25 g lactose tolerance test. Some subjects were challenged with 50 g milk powder on different days. Both indicators, the expiratory H2 concentration and intolerance symptoms, were analyzed. RESULTS: LD occurred in 38.5% of children in the 3-5 year age group, and 87% of the 7-8 year and 11-13 year old groups. The age of occurrence for LD may be at 7-8 years among Chinese children. The prevalence of LI among Chinese children was 12.2% at age 3-5 years, 33.1% at age 7-8 years, and 30.5% at age 11-13 years, respectively. CONCLUSION: The results demonstrate that LD is very common in Chinese children from these four cities. LD and LI have a dose dependent response: lactose absorption and symptoms are based on lactase activity. The relationship between breast feeding history (or the history of cow milk intake) and lactase activity among Chinese children has not been established.

[Analysis of multi-wavelength overlapping chromatographic data by orthogonal projection resolution].

Orthogonal projection resolution(OPR) has been proved to be a high performance chromatographic data processing technique. In this paper, it is applied to the resolution of multi-wavelength chromatographic overlapping peaks. When the highest number of degree of overlapping in chromatogram is less than or equal to the number of wavelengths used, a real resolved chromatogram can be completely obtained from multi-wavelength chromatographic overlapping peaks by this method. When based on dual wavelength chromatographic data analysis, a new method, namely dual wavelength characteristic information analysis (DWCI), used for the base line correction, determination of number of components and region of pure components signal. Analysis of overlapping cases of every components in the overlapping chromatogram is also proposed. DWCI has been successfully used for the analysis of overlapping chromatogram with dual peaks of three components and with single peak of two components. But based on single wavelength chromatographic data, the analysis of these overlapping peaks is a very difficult and complicated problem.

[Effect of exogenous lactase on the absorption of lactose and its intolerance symptoms].

The objective of this study was to evaluate the effects of lactase on lactose malabsorption and its intolerance symptoms, as well as the available way to improve lactose absorption. Healthy adults with a history of lactose intolerance were screened by 25 g lactose tolerance test. The individuals with higher H2 expired and/or lactose intolerance symptoms were selected as the subjects. Subjects were challenged twice with "400 ml low fat milk" and "400 ml low fat milk + 9000Fcc lactase" separately in 3 days interval. The breath H2 concentration and intolerance symptoms were tested in 4 hours after the challenge. The results showed that exogenous lactase can significantly decrease the incidence of lactose malabsorption (the abnormal expiration of H2 decreased from 100% to 48.9%) and milk intolerance symptoms(from 51.1% to 13.3%). The results from this study demonstrate that lactose malabsorption and intolerance symptoms are resulted from the reduced enzyme activities of individuals, and the exogenous lactase can improve lactose absorption and intolerance symptoms. Lactose supplementation may be an available way to increase the dairy consumption and promote health of people.

Determination of polyphenols by high-performance liquid chromatography with inhibited chemiluminescence detection.

A chemiluminescence reaction detector was developed for the detection of polyphenols separated by HPLC based on the inhibition of chemiluminescence from the luminol-potassium hexacyanoferrate(III) reaction by polyphenols. The separation was carried out on a RP-C18 column at 37 degrees C by using stepwise gradient elutions. The detection limits are in the range of 6.8 x 10(-7)-2.0 x 10(-9) g/ml for catechol, protocatechuic acid, chlorogenic acid, rutin, resorcinol, hydroquinone and p-tert.butylpyrocatechol. The method is sensitive, selective, fast and simple. It has been successfully applied to the determination of chlorogenic acid and rutin in real tobacco samples.

Mosaic allelic insulin-like growth factor 2 expression patterns reveal a link between Wilms' tumorigenesis and epigenetic heterogeneity.

Numerous observations link the loss of imprinting of insulin-like growth factor 2 (IGF2) and an overdosage of this growth factor gene with cancer, in general, and with Wilms' tumorigenesis, in particular. It is not known, however, if loss of imprinting correlates with specific stages of neoplasia or if allelic expression patterns vary within the tumor. By applying an allele-specific in situ hybridization technique to formalin-fixed thin sections, we show that the parental IGF2 alleles can be differentially expressed, not only in Wilms' tumors, but also in nephrogenic rests (which represent premalignant lesions) of Wilms' tumor patients. Moreover, a subpopulation of mesenchymal cells, which surrounds tumor nodules, expresses IGF2 biallelically irrespective of the imprinted state of IGF2 within the tumor. These data show that Wilms' tumorigenesis involves epigenetic heterogeneity as visualized by variable allelic IGF2 expression patterns.

[Study on the incidence of lactose intolerance of children in China].

1168 healthy children of 3-13 years old were selected as subjects from Beijing, Shanghai, Guangzhou and Haerbin. Total subjects were challenged by 25 gram lactose test. The breath hydrogen was determined, the symptoms of lactose intolerance were recorded. The results showed that the incidences of lactase deficiency of children of 3-5, 7-8 and 11-13 years old were 38.5%, 87.6% and 87.8% respectively. The incidences of lactose intolerance were 12.3%, 33.2% and 30.5% respectively. Lactase activity decreasing of Chinese children occurred at the age of 7-8 years old, and the symptom of lactose intolerance depended on lactose dose. But, there still are 39%-41.7% children with lactose maldigestion and 14%-16% children with intolerance symptom, after taking 50 gram milk powder (12 g lactose). The relationship between lactase activity and milk intake was not found.

Random monoallelic expression of the imprinted IGF2 and H19 genes in the absence of discriminative parental marks.

The IGF2 and H19 genes are genomically imprinted and expressed preferentially from the paternal and maternal alleles, respectively, during human prenatal development. The exact role of the parental imprint(s), however, is not known. To explore this issue in some detail, we have examined human androgenetic cells which by definition should be incapable of allelic discrimination given the paternal origin of both genomes. Allele-specific in situ hybridisation analysis of dispermic complete hydatidiform moles shows that IGF2 and H19 can be found to be transcriptionally active in a variegated manner, which results in the generation of random monoallelic expression patterns. This data shows that imprinted genes can be expressed monoallelically in the absence of discriminating parental marks and raises the question whether or not mechanisms underlying monoallelic expression preceded the acquisition of parental imprints during evolution.

Effects of calcium channel blockers on calcium release-activated calcium currents in rat hepatocytes.

AIM: To study the influences of calcium channel blockers on calcium release-activated calcium currents (ICRAC) in rat hepatocytes. METHODS: Whole-cell patch-clamp technique was used. RESULTS: The peak amplitude of ICRAC was -0.41 nA +/- 0.09 nA (n = 15), its reversal potential was about 0 mV. Verapamil (Ver), diltiazem (Dil), and nifedipine (Nif) decreased ICRAC strikingly, without affecting its reversal potential. The inhibitory rate of Ver 5 mumol.L-1 was 40% +/- 12% (n = 3), Ver 50 mumol.L-1 reduced the peak amplitude of ICRAC from -0.49 nA +/- 0.12 nA to -0.20 nA +/- 0.09 nA (P < 0.01 vs control, n = 5). The inhibitory rate was 57% +/- 15%. Dil 50 mumol.L-1 and Nif reduced ICRAC from -0.43 nA +/- 0.10 nA to -0.29 nA +/- 0.07 nA (P < 0.01 vs control, n = 5), from -0.32 nA +/- 0.08 nA to -0.27 nA +/- 0.08 nA (P < 0.01 vs control, n = 5). The inhibitory rate was 31% +/- 11%, 19% +/- 7%, respectively. The amplitude of ICRAC was dependent on extracellular Ca2+ concentration. The peak amplitude of ICRAC was -0.21 nA +/- 0.08 nA (n = 3) in Tyrode's solution with Ca2+ 1.8 mmol.L-1 (P < 0.01 vs the peak amplitude of ICRAC in external solution with Ca2+ 10 mmol.L-1). CONCLUSION: The three calcium antagonists inhibited ICRAC effectively and protected hepatocytes from calcium overload via the inhibition of ICRAC.

[The growth effect of granulocyte colony-stimulating factors on ovarian carcinoma cell lines].

OBJECTIVE: To evaluate the growth effects of granulocyte colony-stimulating factors (G-CSF) on ovarian carcinoma cell lines. METHODS: We investigated the possible growth effects of G-CSF on three ovarian carcinoma cell lines (SKOV3, 3AO, and CAOV3). Cell proliferation and viability was measured by XTT colorimetric assay. Flow cytometer (FCM) and G-CSF receptor mRNA were measured for response cell lines, the response cell lines were xenotransplanted into athymic mice, to investigated in vivo growth under systemic subcutaneous infusion of G-CSF. RESULTS: G-CSF showed no growth-stimulating effects in 3AO and CAOV3 cell lines. In SKOV3, an increase in growth (< 5%) was seen by XTT assay at 100 ng/ml and 1000 ng/ml after 24 hours treatment, but no significant proliferation was observed by FCM. No G-CSF receptor mRNA was detected by RT-polymerase chain reaction, and no significant alteration of the tumor growth was by G-CSF. CONCLUSION: Although, the above results suggest that G-CSF may act as growth factors in a few ovarian carcinoma cell lines, yet the worry about tumor growth stimulation by G-CSF as potential hazards for their clinical application in ovarian carcinoma patients as an adjuvant with cytotoxic chemotherapy is not necessary.

[Effect of food composition of mixed food on glycemic index].

UNLABELLED: In order to study the effect of protein, fat and dietary fiber on glycemic index(GI) of mixed food, the response of blood glucose and plasma insulin to nine diets with different components of food were tested by using glucose oxidase and radioimmunoassay. Glycemic index of 9 mixed foods show as follows, rice: 83.2 +/- 3.1, rice + stir fry pork: 72.0 +/- 14.0, rice + stir fry pork and celery: 57.1 +/- 11.2, rice + stir fry garlic sprout: 57.9 +/- 7.8, rice + stir fry garlic sprout and eggs: 62.8 +/- 16.7, steamed bread: 80.1 +/- 22.5, steamed bread + butter: 68.0 +/- 16.3, and steamed bread + beef: 49.4 +/- 22.8. Protein(P, beta 1 = -0.696, P < 0.01) and dietary fiber(Fi, beta 2 = -7.364, P < 0.01) can reduce the blood glucose response and were significantly related to GI. Fat also can inhibit the increment of blood glucose, but there is no significant relation with GI. When the co-ingestion of protein with carbohydrate, the serum insulin response increased greatly and the glycemic response reduced. The addition of fat can reduce the glycemic response without change in serum insulin. Dietary fiber can reduce the serum insulin response and inhibit the glycemic response. CONCLUSION: Protein and dietary fiber of mixed food could markedly affect the glycemic index of foods and reduced the blood glucose response.

[Effect of tian ma cu zhi chongji medicinal granules (TMCZCJ) on blood rheology in hypostasis rats due to stagnation of liver-qi and learning-memory disorder in cerebral ischemia-reperfusion mice].

OBJECTIVE: To investigate the effect of TMCZCJ on the blood rheology in hypostasis rats due to stagnation of the liver-qi and on the learning-memory disorder in cerebral ischemia-reperfusion mice. METHOD: The models of hypostasis due to stagnation of the liver-qi were made by combining irritation with hypodemic injected adrenalin and ice-bath in rats, and the models of learning-memory disorder were made by repeatedly ligating-reperfusing the bilateral carotid in mice. The hypostasis rats and learning-memory disorder mice were perorally administrated TMCZCJ(7.7, 15.4 g.kg-1) once a day, for fourteen consecutive days. RESULT: TMCZCJ can significantly reduce plasma viscosity, blood viscosity and reduced viscosity in hypostasis rats. It can also improve the latent learning-memory capability and memory gain-repetition ability in water-maze for cerebral ischemia-reperfusion mice. CONCLUSION: TMCZCJ can improve remarkably blood rheology in hypostasis rats and learning-memory disorder in cerebral ischemia-reperfusion mice.

[Flow injection analysis of 3,4-dihydroxybenzoic acid with inhibited chemiluminescent detection].

It was found that 3,4-dihydroxybenzoic acid (protocatechuric acid) inhibited chemiluminescence of luminol-H2O2 reaction catalyzed by Co2+. On this basis, a flow injection method has been developed for the determination of protocatechuric acid. The method is simple, convenient and sensitive with detection limit of 2.7 x 10(-7) mol/L and is effective to determine protocatechuric acid in the range of 1 x 10(-6)-1 x 10(-5) mol/L. The variation coefficient of eleven determinations for 8 x 10(-6) mol/L protocatechuric acid is 2.3%.

Inhibition of glycogen synthase kinase 3beta activity regulates proliferation of cultured cerebellar granule cells.

Insulin-like growth factor I (IGF-I) is mitogenic for several types of neuronal progenitors including cerebellar granule neuron progenitors. The present study confirms that IGF-I can function as a mitogen in purified cultures of cerebellar granule cells and identifies intracellular signal transduction molecules that mediate this mitogenesis. In cultured granule cells, IGF-I inhibits GSK-3 activity and leads to phosphorylation of serine9 an inhibitory site on GSK-3beta. Phosphoinositide 3-kinase (PI3-K) activation by IGF-I can lead to phosphorylation and inactivation of GSK-3. A PI3-K inhibitor, LY294002, completely inhibited IGF-I-induced proliferation with half-maximal inhibition occurring at a concentration (1.5 micrograms) close to its reported IC50 value for inhibition of PI3-K. Lithium chloride (LiCl), a direct inhibitor of GSK-3beta, can alone stimulate granule cell proliferation and enhance proliferation induced by IGF-I. LiCl can reverse the inhibitory effect of LY294002 on granule cell proliferation suggesting that GSK-3 inhibition may be downstream of PI3-K activation in IGF-I's mitogenic pathway. Experiments further show that the expression of a dominant active form of GSK-3beta antagonizes IGF-I-induced mitogenesis. These studies support a role for inhibition of GSK-3beta activity in the signal transduction pathway by which IGF-I regulates granule neuron progenitor proliferation.

[Effect of selenium status on the morbidity of interstitial pneumonia in rats infected with mycoplasma].

To investigate the effect of selenium(Se) status on morbidity and process of interstitial pneumonia, we used the Wistar rats as the animal model infected with Mycoplasma Pneumonia (MP). The rats were maintained on the based diets with different doses of Se, infected with MP and exposed to sodium selenite(1.5-2.0 ml/day, containing Se 2 micrograms/ml) or 0.9% NaCl (1.5-2.0 ml/d) by gavage for different time. The experimental rats were divided into five groups, group A, feeding with normal control diet and without infected MP, group B with adequate Se diet and infected MP, group C with Se deficiency diet infected MP and added Se at the same time, group D with Se deficiency diet and added Se after suffered with MP, and group E with Se deficiency diet and infected MP. The results showed that the MP morbidity of the rats fed with high Se diet was significantly lower than that of the rats fed with low Se diet. The pathological change was significant in the low Se group and relatively mild in the high Se group. After infected with MP, the supplementation of sodium selenite to the low Se rats might decrease the MP morbidity and shorten the disease course compared with the group without supplementing Se. The incidence of myocarditis in the infected group with normal Se level diet or in the infected group with Se supplement was lower than that in the group without Se supplementation. The concentrations of Se in plasma and glutathione peroxidase in plasma and white blood cells decreased to a certain level after infection. The present study indicated a positive influence of Se supplement on the morbidity, disease course, and state of interstitial pneumonia in rats.

Expression of the POU transcription factor Brn-5 is an early event in the terminal differentiation of CNS neurons.

The POU domain transcription factors are a subgroup of homeodomain proteins which have two independent domains that function cooperatively in sequence-specific DNA binding. Recent experiments suggest that POU proteins may be critical regulators of central nervous system (CNS) neuronal differentiation. The present study evaluated the temporal and spatial expression pattern of the POU protein Brn-5 in the developing and mature mouse CNS. Immunocytochemistry using a Brn-5-specific antiserum revealed that Brn-5 expression occurs in many CNS neuron populations. In the developing CNS its expression occurs in postmitotic neurons but not in proliferating neuronal progenitors. Experiments in the developing retina suggest that expression of Brn-5 occurs 24-48 hours after progenitors exit the cell cycle. Thus, the expression of Brn-5 appears to be an early event in the process of terminal neuronal differentiation. Brn-5 may function as a transcriptional regulator involved in specifying the mature phenotype of CNS neurons.
