[Relationships between course of anesthesia, depth of anesthesia and acetylcholine content of the brain after urethane and pentobarbital administration in the rat].

Urethane (1.25 g/kg) and pentobarbital (0.06 g/kg) 30 min after i.v. application increase the content of acetylcholine in telencephalic areas but not in the brain stem. These acetylcholine levels are normalized again 120 min p.i.; but urethane treated animals show an elevated acetylcholine content in the brain stem at this time. A different course of anesthesia by pentobarbital and urethane respectively correlated with this result: Only in the presence of urethane a persistent depression of spontaneous motility was seen, whereas the effectivity of both anesthetics on pinna and corneal reflexes and in respect of pentobarbital also on active movements occurrence declined in the end of the test period.

Acetylcholine↗

Interaction between cellular metabolism and acetylcholine turnover of rat brain cortex slices in the presence of arecoline.

Centrally acting cholinomimetic drugs like arecoline stimulate active ion transport processes in the synaptic region. Regarding the connection between cellular metabolism and active Na"-K+-transport the effect of arecoline on the cerebral metabolic status was proved. Arecoline decreased the level of high energic phosphates and glycogen (energy charge diminished from 0,57 to 0,48) and increased the glucose consumption and lactate production. Thus, the increased rate of CoA acetylation via oxidative breakdown of pyruvate seems to be prerequisite for the cholinergically stimulated ACh synthesis.

Acetylcholine↗

Correlation between drug induced changes of acetylcholine release and acetylcholine fractions in rat brain.

After administration of eserine, a new acetylcholine (ACh) subfraction called f+ is formed in rat brain tissue. References and methods are given for the calculation of this subfraction, which can be isolated and determined only together with the so-called "free" ACh fraction. Alterations of the f+-ACh subfraction caused by barbital, urethane, pentetrazol, arecoline and scopolamine in telencephalon, cortex and striatum of rat brain are connected with changes of ACh concentrations determined in comparable releasing tests.
