[Contact eczema caused by pesticides in East Germany].

19 allergic, occupational contact eczemas and 1 irrative contact dermatitis caused by pesticides from 1985-86 in the GDR are described. The 20 casuistic reports are supplemented with data concerning age, sex, exposure time, patch-test results, occupation site and dermatological details. The following pesticides were determined: maneb, zineb, mancozeb, propineb, thiram, demephion, dimethoate, propachlor, metham-Na, benomyl, chloralhydrate with chloralmethylhalbacetal, carbaryl, dinocap captan, phenyl-Hg-acetate and trichlodinitrobenzene. Each individual case adhered to the recommended test concentration for skin tests. Contact eczemas brought about by the pesticides demephion and trichlordinitrobenzene have not yet to be described. Those working in gardening seem to be particularly prone.

[Recommendations for the therapy of atypical immunization courses and complications of BCG vaccination (Position: April, 1988)].

After introduction of the "direct" BCG-vaccination of the 16 years old youngsters in 1981 a small increase of atypical reactions and complications of the vaccination was probable. References and experiences about different therapy methods of these atypical reactions and complications gave rise to recommend uniform guide-lines for definition, diagnostic and therapy.

[Three-dimensional structure determination of antibodies. Primary structure of crystallized monoclonal immunoglobulin IgG1 KOL, I].

The crystallizable myeloma immunoglobulin IgG1 KOL [allotype Gm(-1,4), (gamma 1, gamma)2] which is well characterized in its three-dimensional structure by X-ray diffraction analysis of high resolution has been proved to be homogenous by polyacrylamide gel electrophoresis. The H- and L-chains were separated by gel filtration after complete reduction and carboxymethylation and were characterized by amino acid analysis, end group determination and polyacrylamide gel electrophoresis, respectively. The intact IgG1 KOL was cleaved by cyanogen bromide and all CNBr-fragments were isolated and characterized. The reduced and carboxymethylated H-chain was digested by trypsin and the tryptic hydrolysate was separated by ion-exchange chromatography. Using different procedures of rechromatography 35 out of 37 tryptic H-chain peptides could be isolated in sufficient amounts, the missing 2 peptides were produced by tryptic digestion of 2 CNBr-fragments. The amino acid sequences of all tryptic peptides were determined using a modified Edman degradation method after separation of the enzymatic cleavage products by high-performance liquid chromatography (HPLC). The complete primary structure of the VH-part of the H-chain was established by isolation and partial sequence determination of overlapping peptides obtained from cleavage of the intact H-chain by Staphylococcus aureus proteinase. The gamma 1-H-chain KOL comprises 455 amino acid residues and belongs to subgroup III. The switch from the variable to the constant part occurs at position 126/127, thus making VH-KOL one of the longest variable parts among the yet known immunoglobulin H-chains. This is due to the hypervariable region Hhv4 which is made up by 17 amino acid residues (4-9 residues more compared with other VH-parts). Within this region a so far not described additional intrapeptidal disulfide bridge could be localized (Cys 105-Cys 110) that creates a short loop with antiparallel running peptide strains in beta-pleated sheet conformation. Its role in the three-dimensional structure of the antigen-binding site of the IgG1 KOL molecule is discussed using the data obtained from X-ray diffraction analysis.

[Epidemiology of malignant melanoma in East Germany and West Germany].

Data are given on the incidence of the malignant melanoma in the German Democratic Republic. The latter steadily increased from 1955 to 1975, i.e., from 1.55/100,000 (male) and 1.80/100,000 (female) to 3.1/100,000 (male) and 4.3/100,000 (female). A genuine increase of the incidence can be proven by age standardization from 1968 to 1975. Between 1955 and 1975 the incidence in men and women doubled in the German Democratic Republic. The death rate from malignant melanomas is about stationary. The survival rate in men lies about 50%, that in women is higher. In the Federal Republic of Germany about 1,674 incidences in 1969, about 1,825 in 1974, and more than 2,000 incidences of malignant melanomas in 1975 were to be expected. UV radiation and chemical carcinogens influence the incidence of the malignant melanoma in our area.

[The current state of cutaneous tuberculosis].

Thirty-three years after the introduction of the chemotherapeutics and as result of massive prophylactic and preventive measures the tuberculosis of the skin shows a clear epidemiologic and clinical change. Nowadays the clinical picture of the tuberculosis of the skin restricts essentially to the classical forms of the lupous, verrucous and colliquative tubervulosis of the skin, the specific etiology and pathogenesis of which are ascertained. The so-called tuberculids should at present only be treated in a qualified sense in the chapter of tuberculosis of the skin. After a short description of the diagnostic possibilities and after a historical survey on former measures of treatment the present GDR standard in therapy is explained. The qualification for carrying on the isonicotinic acid hydrazide monotherapy in the tuberculosis cutis luposa and verrucosa is proved on the basis of bacteriological, pathologo-anatomical and clinical peculiarities of these forms of tuberculosis of the skin.
