Bronchial obstructive reactions after inhalation with endotoxin and precipitinogens of Haemophilus influenzae in patients with chronic non-specific lung disease.

Bronchial obstructive reactions occur after inhalation of Haemophilus influenzae in the absence of proteolytic activity in patients with CNLSD and a lowered histamine threshold in whom specific precipitins are demonstrated. It may be presumed, that the direct bronchial reaction is caused by its content of endotoxin, whereas the late bronchial reaction is the result of a toxic action and a type III allergic reaction. When proteolytic activity is present it lowers the histamine threshold and may influence the endotoxin effect in this way.

The role of the adrenergic system in allergy and bronchial hyperreactivity.

The adrenergic system may have an influence on the severity and activity of bronchial asthma. It has been suggested that there might be a relationship between the allergic process and the adrenergic system. Therefore this system was studied before and after an allergen challenge. Two parameters were measured: in vivo: propranolol threshold measurement to assess adrenergic involvement; in vitro: the beta-adrenergic response of peripheral lymphocytes as parameter for beta-receptor function. Seven allergic asthmatics who were not treated with beta-stimulants or corticosteroids and seven normals inhaled house dust mite extract. On the days before and after the challenge, propranolol threshold measurements were performed. Six patients had a dual reaction after a house dust mite inhalation; one patient had an early response. The propranolol threshold changed from 1.32% (SEM 0.16) on the day before challenge to 0.86% (SEM 0.19) on the day after (0.02 less than P less than 0.025). In the same patients the maximal cAMP response of lymphocytes in the presence of 10(-6) mM isoproterenol changed from 339% (SEM 18.9) above basal level before the allergen challenge to 194% (SEM 24.6) after the challenge (0.001 less than P less than 0.005). The pre-challenge beta-response of lymphocytes of the asthmatic patients was comparable with that of normal lymphocytes (298% SEM 59.8). The results suggest that an allergen-induced asthmatic attack may lead to an impaired beta-adrenergic function.

Determination of N tau-methylhistamine in plasma and urine by isotope dilution mass fragmentography.

The determination in N tau-methylhistamine by gas chromatography-mass spectrometry is described using bis-heptafluorobutyryl derivatives and deuterium labelled N tau-methylhistamine as internal standard. A description of the synthesis of the internal standard, N tau-trideuteromethylhistamine, is given. Normal concentrations for plasma were 1.4 +/- 0.4 microgram/l (mean +/- 1 S.D., n = 10). The normal values for urine ranged from 60 to 280 microgram/24 h (n = 20). Five asthmatic patients showed above-normal plasma concentrations.

Effect of ligands on cytochrome d from Azotobacter vinelandii.

Spectra of oxidized and reduced cytochrome d in particles of A. vinelandii were studied in the presence of the ligands CO, azide, and NH2OH under oxidizing, reducing, and turnover conditions. Under oxidizing conditions, spectral changes were observed on oxidized cytochrome d (absorption maximum at 648 nm) in the presence of CO and NH2OH showing a shift of the maximum to shorter wavelengths (639 and 645 nm, respectively) and a broadening of the half-band width. Under reducing conditions, spectral changes were observed on reduced cytochrome d (absorption maximum at 631 nm) in the presence of CO (absorption maximum at 636 nm), NO, NO2-, and NH2OH (absorption maximum at 642 nm in the presence of dithionite). The spectral changes of cytochrome d in the presence of NH2OH or with dithionite and NO2- were ascribed to the formation of the NO-cytochrome d compound. Under turnover conditions CO, NH2OH, and azide cause a spectral shift of the absorption maximum of cytochrome d from 648 nm to 636, 645, and 655 nm, respectively. With NH2OH and azide a broadening of the half-band width of 7 and 6 nm, respectively, was also observed. The spectral changes caused by CO and NH2OH were interpreted as a binding of the ligands to cytochrome d changing its conformation from the oxidized state absorbing at 648 nm into a more stable liganded form. Since azide does not affect the spectral bands of oxidized and reduced cytochrome d, the spectral change during turnover in the presence of azide were ascribed to a preferential binding of azide to enzymically active conformation of cytochrome d (cytochrome dx).

Antibodies against Aspergillus fumigatus. I. Standardization of the antigenic composition.

Three different stages were found during growth of Aspergillus fumigatus as characterized by the changes in pH of the medium: (1) An acidic phase (phase I); (2) a second phase (phase II) during which the pH rises till values around pH = 8.5; (3) a third phase (phase III) during which the pH remains constant or drops slightly. During phase I a fast appearance of certain antigenic components was found that is ascribed to an active process of excretion. Additional antigenic components appeared in the culture medium after lysis of the microorganisms (phases II and III). Lysis of the microorganisms and appearance of antigenic components are dependent on the glucose concentration of the medium.

Purification of specific precipitinogen and extraction of endotoxin from Haemophilus influenzae.

After purifying a Haemophilus influenzae precipitinogen from endotoxic activity by means of ultracentrifugation, column chromatography (Sepharose 6B) and ion exchange chromatography (DEAE Sephadex A25) a fraction was obtained which still contained a specific precipitinogen that was virtually free of endotoxin. Furthermore, during the chromatographic procedures fractions with a high and a low molecular weight endotoxic activity were found. The limulus lysate test was more sensitive in the high molecular weight fractions and the LD50 in mice in the low molecular weight fractions with endotoxic activity.
