Inhibitory effects of various progestins and deoxycorticosterone on the rapid onset of maternal behavior induced by ovariectomy-hysterectomy during late pregnancy in rats.

The inhibitory effects of progesterone (P), 5 alpha-dihydroprogesterone (5 alpha-DHP), 17 alpha-hydroxyprogesterone (17-HP), and deoxycorticosterone (DOC) upon the rapid onset of maternal behavior induced during late pregnancy in primigravid rats by ovariectomy-hysterectomy (OH) were examined. Progesterone administration at a dosage of 5.0 mg oil vehicle daily (beginning on Day 17 and ending when the subject responded maternally to foster pups) significantly delayed by about 1.5 to 2.0 days the onset of maternal behavior. In contrast, P at dosages of either 1.0 or 2.5 mg daily, 5 alpha-DHP at 5.0 mg administered daily in either an oil or Tween-80 vehicle, 17-HP at 5.0 mg in oil daily, and DOC at dosages of either 5.0 or 10.0 mg in oil daily failed to effect the rapid onset of maternal behavior induced by ovariectomy-hysterectomy on Day 17 of pregnancy. These data suggest that during pregnancy the onset of maternal behavior is inhibited in a rather specific manner by progesterone.

Facilitation of lordosis behavior by clonidine in female guinea pigs.

Lordosis behavior was induced in previously unreceptive, ovariectomized estrogen-primed female guinea pigs by administration of the noradrenergic agonist clonidine. Clonidine also enhanced lordosis responding in females that were weakly receptive after estrogen priming. Unlike progesterone, the lordosis-facilitating effect of clonidine was not accompanied by a subsequent refractory period. Clonidine had a weak lordosis facilitatory effect when administered during the decline in receptivity to estrogen plus progesterone-primed animals and failed to induce lordosis when administered during the refractory period.

Role of catechol estrogens in activation of lordosis in female rats and guinea pigs.

In a variety of experiments, we tested the effectiveness of the 2-hydroxylated estrogen in facilitating sexual receptivity. A single injection of 2-hydroxy-estradiol-17beta (2-OHE2) to ovariectomized rats or 2-hydroxy-estrone (2-OHE1) to ovariectomized guinea pigs was ineffective in priming animals for facilitation of sexual receptivity even when a subsequent injection of progesterone was administered. The only facilitatory effect of catechol estrogens on lordosis that was demonstrated in this study occurred when 2-OHE2 was injected in combination with E2 and a subsequent injection of progesterone was given to rats. These results suggest a cooperatively between catechol estrogen and E2, but they also indicate that catechol estrogens, by themselves, do not play a crucial role in mediating sexual receptivity in rodents.

Relation of uptake and metabolism of (1,2,6,7-3H)testosterone to individual differences in sexual behavior in male guinea pigs.

Male guinea pigs were given 3 tests for sexual behavior. Animals that never ejaculated were classified as low activity (LA), animals that ejaculated on one test were classified as medium activity (MA), and animals that ejaculated on two or more tests were classified as high activity (HA). Subsequently, animals from each group were castrated and given an s.c. injection of 43 muCi of [1,2,6,7-3H]testosterone and were killed 0.5, 1, or 4 h after injection. There were no significant differences in uptake or metabolism of radioactive testosterone among LA, MA, and HA males in homogenates of anterior and posterior hypothalamus, cerebral cortex, midbrain, or seminal vesicle. Thus, differences in sexual behavior could not be attributed to differences in testosterone uptake in tissue homogenates. At the 1 h time interval (time of peak plasma radioactivity), radioactivity in the seminal vesicles of all males was primarily in the form of steroids with the chromatographic mobility of dihydrotestosterone. In all males, anterior and posterior hypothalamus contained a higher proportion of steroids with the mobility of testosterone than did midbrain, and midbrain contained more testosterone zone radioactivity than cerebral cortex at 1 h. The highest proportion of dihydrotestosterone zone radioactivity in neural tissues was found in anterior hypothalamus. These results are discussed in terms of androgenic mediation of sex behavior by the anterior hypothalamus in guinea pigs.
