Generation of cytotoxic T lymphocytes and cytostatic acting cells in T. annulata-immune cattle.

Cattle immunized against Theileria annulata with schizont containing autologous cell lines are immune to challenge with a homologous parasite strain. Two cell types have been detected in the peripheral blood of the immunized animals: cytotoxic T lymphocytes (CTL) and cytostatic acting cells (CAC). Killing the target cells by CTL is infection associated and is MHC class I restricted. Hence, no cytotoxicity was observed against target cells that were treated with the theilericidal drug buparvaquone or autologous Con A-blasts. The growth inhibition of CAC is MHC unrestricted, and not mediated by cytokine interferon gamma (IFN-gamma).

Determination of trovafloxacin in human body fluids by high-performance liquid chromatography.

For the quantitative determination of trovafloxacin (a new naphthyridinone antibacterial agent) in serum and urine a simple isocratic HPLC method with fluorimetric detection is described. Serum was deproteinised with a mixture of acetonitrile and perchloric acid. The protein-free extract was separated on a reversed-phase column (Nucleosil 100-5 C18) and quantified by means of fluorescence (excitation 275 nm, emission 405 nm). The mobile phase consisted of a mixture of 250 ml acetonitrile and 750 ml distilled water containing 10 mmol/l tetrabutylammonium phosphate. Urine was diluted with 0.25 mol/l phosphoric acid 1:20 (v/v) which was adjusted to pH 3.6 with sodium hydroxide solution. Diluted urine samples were separated on a cation-exchange column (Nucleosil 100-5 SA) and also detected by means of fluorescence. Trovafloxacin was sufficiently separated from endogenous compounds. Results of validation are given. The method was applied successfully to a study of healthy volunteers.

The Rotational Spectrum of the p-Fluorotoluene- Argon van der Waals Complex.

The microwave rotational spectrum of the p-fluorotoluene-argon van der Waals complex was analyzed with a molecular beam Fourier transform microwave spectrometer. In the frequency splitting of molecular transitions caused by the internal rotation of the methyl group with respect to the aromatic ring is contained the structure and barrier information. Elucidation is reduced by the analysis of internal rotors direction with respect to the principal axis system of the complex the number of possible solutions in the structure. An rs and r0 structure of the complex was calculated from these data. The argon is located 3.541(1) Å above the aromatic ring. By forming the complex, the barrier height of internal rotation is changed. An additional V3 term in the potential function occurs in the complex because the molecular symmetry of the p-fluorotoluene, containing a V6 but no V3 term, is reduced by the argon. For assumed V6 = 144.79(19) GHz or 57.777(76) J/mol, V3 is found to equal 552.0(10) GHz or 220.31(40) J/mol. Copyright 1999 Academic Press.

HPLC determination of clofazimine in tissues and serum of mice after intravenous administration of nanocrystalline or liposomal formulations.

A simple HPLC method is described for the determination of clofazimine in mouse tissues and in serum. The main application of the method was the determination of the drug in mouse tissues after i.v. administration of nanocrystalline suspensions or liposomal encapsulated clofazimine. Tissues were extracted with a 10-fold (w/v) volume of an extraction solution consisting of methanol/glacial acetic acid 9:1 (v/v). Serum proteins were precipitated with a 2-fold volume of acetonitrile. Isocratic chromatography was performed using an anion exchange column (Nucleosil 100-5 SA, Macherey & Nagel) for separation. The mobile phase was a mixture of acetonitrile and 0.1 mol/l aqueous phosphoric acid (75:25, v/v), adjusted to pH 2.9 with sodium hydroxide solution. Absorption of the eluate was monitored at 495 nm. The assay was precise, simple to perform and fast. Recovery from tissues was > or = 98%, from nanoparticles > or = 98%, and from liposomes > or = 96%. No interference was observed in extracts from mouse liver, spleen, lungs and human serum.

The Microwave Spectrum of the Methanol Dimer for K = 0 and 1 States

The rotational spectrum of (CH3OH)2 has been observed in the 8 to 24 GHz region with a pulsed-beam Fabry-Perot cavity Fourier-transform microwave spectrometer. Previously we demonstrated that each transition of the a-type R(J), Ka = 0 is split into 15 states of the 16 theoretically expected states by tunneling motions. Here we show that the K = 1 states are split into the 16 expected states through the assignment of the Ka = 1 a-type transitions and DeltaKa = 1 b-type transitions. The internal-rotation analysis of the two inequivalent methyl groups presented here was guided by the previous experimental observations and theory for multidimensional tunneling, which predicts 16 tunneling components for each R(J) transition from 25 distinct tunneling motions. The effective barrier to internal rotation for the donor methyl group of (CH3OH)2 is V3 = 183.0 cm-1, and is one-half of the value for the methanol monomer (370 cm-1), while the barrier to internal rotation of the acceptor methyl group is 120 cm-1, one-third of the methanol monomer. The structure of the methanol dimer complex is similar to that of water dimer with a hydrogen bond distance of 1.96(2) A and tilt of the acceptor methanol of 77(2)degrees from the O-H-O axis (one standard deviation uncertainty). This structure shows good agreement with the angular orientation of the methyl groups derived in the internal-rotation analysis. Copyright 1997 Academic Press. Copyright 1997Academic Press

[Alcohol and traffic safety--study of epidemiology (II. 1990/1991 status].

With the co-operation of a further 13 institutes and as a continuation of our own epidemiological-statistical survey, we recorded the total and relative distribution (frequency profile) of the blood alcohol concentration of car drivers for the 3rd quarter 1990 and the 1st quarter 1991. The participating institutes recorded almost 27,000 blood samples and compared them with results of the first part of the study (1989). The survey dealt separately with sex-, age- and time of day-distribution of drivers involved and not involved in road accidents. One of our repeatedly presented field research into the problem of "alcohol and road safety", once again, provided to be a useful method in the run-up to the unprejudiced alcohol test which we are striving for. We noticed characteristic changes in the frequency profiles of the old and the new counties (Bundesländer) in Germany in the 1st (1989) and the 2nd (1990/91) report of the study. The results of Police supervisory operations in the administrative district of Cologne have basically given useful indications as to their epidemiological-statistical value as evidence.

Effect of cyclosporin A on the proliferation of bovine lymphocytes to concanavalin A and on the growth of Theileria annulata-infected bovine cells.

After being inoculated by Hyalomma ticks, the sporozoites of Theileria annulata invade bovine lymphocytes, where they subsequently differentiate to schizonts. The infected cells are induced to a continuous proliferation which can be enhanced by human recombinant interleukin 2 (hrIL-2). In the present study, we examined the influence of cyclosporin A (CsA) on the growth of schizont-containing cells and compared with its effect on bovine peripheral blood lymphocytes (PBL) responding to Concanavalin A (ConA). In both cell types, the proliferation was inhibited in a dose dependent manner, which was not restorable in T. annulata-infected cells even after addition of hrIL-2. In contrast, ConA-blasts were able to undergo a proliferative response provided they were treated with high doses of CsA. Both, T. annulata-infected cells and bovine ConA-blasts express IL-2 receptors (IL-2R). The binding of radiolabelled hrIL-2 to ConA-blasts and T. annulata-infected cells was only partially inhibited after treatment with CsA. CsA was not toxic for the parasites, since the treated cells still contained schizonts which did not show any morphological abnormality.

[Operation indications of blow-out and midface fractures].

89 patients with fractures of the orbital floor - both in isolation (blow-out fractures) and combined with a complex fracture of the midregion of the face - were followed up for an average of 5 years. Particular attention was paid to the quantitative evaluation of binocular fusion, diplopia and enophthalmos. Comparison of operated and conservatively treated patients with blow-out fractures led to the following conclusions with regard to indications for surgery: 1. It is not justified to regard either surgery or its postponement pending developments as obligatory in all cases. 2. Surgery is indicated by clinical, orthoptic and roentgenological findings as follows: a) In case of enophthalmos or roentgenological demonstration of prolapse, surgery is indicated independent of diplopia. b) Diplopia outside the main field of view does not, in general, necessitate surgery. c) If there is diplopia within the main field of view, surgery is indicated. It is generally accepted that orbital-floor fractures associated with complex mid-face fractures require early surgical treatment. The present investigation confirms this view.

Drugs in pregnancy - a prospective study.

A prospective study starting with 1182 women in the early stage of pregnancy investigated prescribed drug therapy; self-medication; consumption of coffee, cigarettes, and alcohol; and a series of social and sociologic factors. The main results are as follows: compared with international reports, drug consumption during gravidity was low. During the first weeks of gravidity, 11% of the women took drugs. Drug consumption increased during gravidity, reaching a maximum of 26% of the pregnant women taking drugs. Drugs most frequently prescribed were antiemetics, analgesics, sedatives, iron preparations, and vitamins. Self-medication (1 or 2%) was extremely low; in most cases analgesics were used. Coffee consumption declined, particularly during the last trimester; consumption of cigarettes showed a distinct decrease from 27% to 12%.

[Retraction syndrome (author's transl)].

A report on 70 cases of retraction syndrome (Stilling-Türk-Duane). The patients presented with typical symptoms: severe limitation of abduction, slight limitation of adduction and convergence, which is always accompanied by retraction of the globe, narrowing of the palpebral fissure, vertical deviation and elevated intraocular pressure, and a remarkable motility of the globe in the orbital axis. All symptoms are explicable by fibrosis or coinnervation of the lateral rectus muscle. Usually, single binocular vision is present with or without a forced attitude of the head. This forced attitude is such that the affected eye utilizes the reduced abduction fully and does not avoid the abduction (as in typical abducens-paresis), but retraction. In cases of single binocular vision, surgery is indicated only when the forced attitude of the head is intolerable. We restrict surgery to recession of horizontal rectus muscles according to the forced attitude.

[The pharmacocinetics of azlocillin (author's transl)].

Azlocillin concentrations in the rabbit eye were studied after intravenous and subconjunctival injections. The following results were found: therapeutic levels of Azlocillin were achieved in the aqueous humor, using either way of administration. Concentrations of the antibiotic were much higher after s. c. injection than after i. v. administration. The pharmacocinetics of Azlocillin proved to be comparable to those of other penicillins. No antibiotic could be detected in the vitreous body, irrespectively of the administration. With regard to the better antibacterial activity of Azlocillin in comparison with other known penicillins it should be preferred in eye infections caused by gramnegative rods, especially by Pseudomonas aeruginosa.

[Pharmacokinetics of mezlocillin in the rabbit eye].

Following intravenous injection of 50 mg/kg Mezlocillin into rabbit eyes the concentrations measured in the aqueous humor were of low-grade therapeutic efficacy. In the vitreous, this dose and form of application had no therapeutic effect. The transfer concentration in the aqueous humor after subconjunctival application of 20 mg Mezlocillin was comparably low. High therapeutic levels in the aqueous humor and even, in isolated cases, traces of Mezlocillin in the vitreous were found after subconjunctival injection of 100 mg.
