Use of flagellin-enriched antigens in a rapid, simple and specific quantitative enzyme immunoassay for Lyme disease antibodies in human serum samples.

An enzyme immunoassay (EIA, ELISA) using microwells coated with a flagellin-enriched fraction of B. burgdorferi and absorbent-containing sample diluent for the quantitative determination of Lyme disease (LD) IgG and IgM antibodies in human serum samples was described. This LD EIA required three 15-minute incubations at room temperature, followed by a 1-step normalization of photometer readings to EIA units (EU/ml). Compared with tests using the whole bacterial extract as antigens and a sample diluent containing 6% BSA, this new LD EIA revealed lower values for 20 syphilis (SS) and 21 normal serum samples (NS) but about the same for 21 Lyme disease (LD) samples, allowing lower cut-off points which would place almost all these SS and NS samples below while almost all LD samples above the positive cut-off point. The LD EIA results of larger numbers (67 to 291) of mixed samples correlated with results of four reference EIA. However, the LD EIA gave lower (2 to 4 fold) reactivities (index values) with SS and NS samples but higher values with positive serum samples than reference EIA. Thus, this LD EIA showed improvements in both specificity and sensitivity over other tests compared.

[Color perception disorders in therapeutic glycoside serum concentrations].

Vision deficiencies are dominant symptoms from digitalis intoxication. Twenty-nine and twenty-eight patients have been examined by the Farnsworth-Munsell-100-Hue-test under common maintenance therapy with digitoxin respectively digoxin or beta-acetyldigoxin. This was done to prove how glycosides may create colour vision disturbances even in "therapeutic" glycoside serum concentrations, and in which degree the lipophile digitoxin differs in its effect from the less lipophile digoxin. To exclude any congenital colour vision deficiencies the study was restricted to females. No definite difference in the total error score has been found among patients treated with digitoxin and digoxin. Our results and further new investigations have pointed out that colour vision in digitalized patients is influenced even under common maintenance therapy. It can also be stated that digitoxin does not influence colour vision other than digoxin.

Membrane attach complex of complement (MAC): three-dimensional analysis of MAC-phospholipid vesicle recombinants.

The three-dimensional structure of recombinants of the isolated membrane attack complex (MAC) of complement with single bilayer dioleoyllecithin (DOL) vesicles and with dimyristoyllecithin (DML) vesicles was determined. A total of four MAC-vesicle complexes were analyzed by imaging negatively stained specimens at various defined tilting angles under minimal dose conditions in the electron microscope and by computer-aided three-dimensional reconstruction. The information on electron micrographs obtained at 6 degrees angular increments from +60 degrees to -60 degrees was digitized by densitometric scanning, Fourier-transformed, corrected for imaging errors, cross-correlated, and synthesized to the three-dimensional image. All four MAC-vesicle recombinants showed stain penetration into the interior of the vesicle, indicating increased permeability of the bilayer to negative stain. The MAC appeared as a hollow structure of 16-nm height, 2.0-nm wall thickness, and a 3.0-nm torus at the free end with an outer and inner diameter of 20.0 nm and 10.0 nm. In MAC-DOL vesicles the hollow core of the MAC terminated at the membrane-binding site, and only small pores of up to 2.0-nm in diameter penetrated the bilayer. In one MAC-DML vesicle lipid discontinuities on the outer circumference of the MAC binding site mediated stain penetration. The second MAC-DML vesicle showed a channel of approximately 4.0 nm connecting the hollow core of the MAC across the bilayer with the vesicle interior. The results suggest the MAC may mediate increased membrane permeability by protein channel formation in addition to lipid reorientation.

[Tolerance and contrast density in intravenous cholangiography and cholecystography using a new contrast medium (author's transl)].

The tolerance and diagnostic usefulness of the new intravenous bile contrast medium Endomirabil were subjected to clinical radiological testing. Endomirabil is well tolerated. It is our impression that it has a lower side effect quota than the other contrast media used so far. Diagnostic results in injection cholangiography and cholecystography with 30 ml Endomirabil were good. In more than 80% of the cases the bile ducts were well demonstrated. The diagnostic evaluation was fully satisfactory. In most cases both the bile ducts and the gallbladder showed up very well within 60 minutes after completion of the injection. This means both film material and time are saved.

[Asymptomatic excretors of rotavirus].

Asymptomatic Carriers of Rotavirus. Samples of faeces from healthy, mostly adult persons, which were sent to us according to legal requirements to permit food handling, were additionally tested for the presence of rotaviruses. 1.6% of this group proved to be positive at the time of sample collection. Contrary to the typical winter peak of diseased patients, the virus carriership was evenly distributed throughout the year in healthy persons. The results are discussed especially with regard to epidemiological points of view.
