Effects of iron and desferrioxamine on Rhizopus infection.

To investigate the association among iron, desferrioxamine, and a Rhizopus infection, the influence of iron and/or desferrioxamine on experimental mucormycosis in mice was examined. All mice pretreated with iron, desferrioxamine, or a combination of iron and desferrioxamine died within 5 days after the inoculation of R. oryzae. In the mice fungal lesions were observed in the brain which resembled human cerebral mucormycosis. By contrast, the mortality in the control mice with R. oryzae was 20% through the 3-week experimental period. Therefore, it was demonstrated that iron as well as desferrioxamine administration markedly promotes the growth of R. oryzae. The increased susceptibility to R. oryzae was considered to be due to increased serum iron in the animals pretreated with iron only; however, pretreatment with desferrioxamine did not affect the amount of serum ion. Thus, the data suggest that desferrioxamine acts as a siderophore to R. oryzae and exerts an adverse effect on mucormycosis. This study has shown that the presence of iron and desferrioxamine enhances the virulence and pathogenicity of R. oryzae by serving as a growth factor.

A tubular bioreactor for high density cultivation of microorganisms.

By simulating the functions of the animal intestine, the authors have developed a novel tubular bioreactor (TBR) which is capable of containing both the reaction and separation of products in a single system. This reactor consisted of inorganic ultra filtration membrane modules in the primary part of the system, a heat exchanger and a recycling pump. The operation characteristics of the TBR were studied by cultivating Lactobacillus casei at a laboratory scale. The cell density obtained was up to 10 times higher than the density obtained by using the conventional jar fermentor. Furthermore, 40 g l-1 of cell mass was obtained in only 14 hours with 20 l of fresh medium when the dilution rate was increased according to the cellular growth. Afterwards, the cultivation time and the volume of fresh medium were reduced to 44% and 74%, respectively, of the values in the cultivation operation at constant dilution rate.

[Cytotoxicity of lymphocytes sensitized by autologous tumor cells in vitro, against autologous lung cancer cell lines].

In order to obtain killer lymphocytes that would have a strong cytotoxicity against autologous tumor cells, we conducted a cytotoxic assay of effector cells cultured from human peripheral blood lymphocytes (PBL) using 10 cultured lung cancer cell lines as target cells. We present this result. (1) Lymphocytes obtained by 17 days mixed lymphocyte tumor cell culture (MLTC: Fresh PBL from lung cancer patients was mixed with irradiated autologous tumor cells in a medium, and cultured for 3 days. Irradiated allogeneic lymphocytes were then added, and cultured for 14 days) were cytotoxic against 4 of the 10 autologous lung cancer cell lines. (2) Lymphocytes obtained by 14 days MLTC and 3 more days culture in a medium containing interleukin 2 were cytotoxic against all 10 of the autologous lung cancer cell lines. These lymphocytes proliferated to 4.13 times of the original cell number, and their surface marker was OKT3+. These killer lymphocytes are expected to be effective in adoptive immunotherapy as effector cells.

Augmentation of superoxide generation in phagocytosing murine peritoneal macrophages by dietary restriction.

The effects of dietary restriction on the generation of superoxide anion (O2-) in phagocytosing mouse peritoneal macrophages (MPs) were investigated. MPs obtained from diet-restricted mice generated larger amount of O2- during phagocytosis of opsonized zymosan than those from unrestricted mice. This augmentation of the O2- generation was not inhibited by the protein kinase C (PKC) inhibitor 1-(5-isoquinolinylsulfonyl)-2-methylpiperazine (H-7). However, the augmentation was clearly disappeared when the calmodulin (CaM) antagonist N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide (W-7) was added. These results strongly suggest that the augmentation of O2- generation in phagocytosing MPs by dietary restriction is due to the increased activity of CaM in the MPs, and that PKC is not involved in the augmentation. It is thought that one of the major factors for the reduced incidence of tumor and infection in diet-restricted animals is the augmentation of O2- generation in MPs.

Effects of intraperitoneally administered dietary fibers on superoxide generation from peritoneal exudate macrophages in mice.

To clarify whether edible polysaccharides (dietary fibers) have a host defense stimulating activity, the effects of intraperitoneally administered dietary fiber on the generation of microbicidal superoxide anion (O2-) in mouse peritoneal exudate macrophages (MP) were investigated. MP obtained from mice injected cellulose or pectin generated larger amount of O2- when stimulated by 12-o-tetradecanoylphorbol-13-acetate (TPA) or during phagocytosis of opsonized zymosan than those from mice injected proteose peptone, a widely used elicitor for MP. Especially, generated O2- from cellulose-elicited MP was as strikingly large as that from MP elicited by lentinan, a strong antitumor polysaccharide, when stimulated by TPA. Yeast mannan had no effect on O2(-)-generation. These results suggest the possibility that cellulose and pectin may act as host defense stimulators. The augmentation by cellulose or pectin of O2(-)-generation during phagocytosis was not inhibited by the calmodulin (CaM) antagonist N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide (W-7). However, the augmentation was completely disappeared when the protein kinase C (PKC) inhibitor 1-(5-isoquinolinylsulfonyl)-2-methylpiperazine (H-7) was added. These results strongly suggest that the augmentation of O2(-)-generation in phagocytosing MP by cellulose or pectin might be due to the increased activity of PKC in the MP, and that CaM may not be involved in the augmentation.

[A useful combination chemotherapy of cisplatin and etoposide in mediastinal yolk sac tumor].

A 28-year-old male was referred to our hospital because of chest pain and dry cough. Chest x-ray film revealed a tumor mass in the anterior mediastinum indicating invasion into the chest wall and upper lobe of right lung. No tumor was found in the testis. Serum level of alpha-fetoprotein (AFP) was 6,400 ng/ml. Percutaneous biopsy of the tumor suggested yolk sac tumor. The patient was treated with a combination chemotherapy schedule including 30 mg of cisplatin (CDDP) i.v. on Days 1, 2, 3, 4, and 5 and 150 mg of etoposide on Days 1, 2, and 3 as one course therapy. Two courses were employed. Partial response was achieved followed by successful resection of the tumor. This case indicated that a combination chemotherapy CDDP and etoposide combined with surgery could play an active role increasing long-term survival rates in mediastinal yolk sac tumors.

[A case of thoracic neurilemoma with neurilemomas of the upper limb and sacrum].

A 64-year-old male was admitted in February 24, 1988 because of an abnormal shadow on chest X-ray. The tumor in the thoracic wall was removed at the thoracotomy and histology showed neurilemoma. Aspiration biopsy of tumors on the left upper limb and sacrum revealed the typical histopathologic appearance of neurilemomas. The possibility of malignant transformation and sequential appearance of tumors in patients with a neurilemoma indicates that complete resection and thorough follow-up examination are important.

[A case of mediastinal yolk sac tumor successfully treated with chemotherapy and surgery].

A 28-year-old man was admitted due to increasing respiratory symptoms. X-ray examination of the chest showed a tumor mass in the anterior mediastinum with possible invasion into the chest wall and upper lobe of right lung. No tumor was found in the testis. Serum alpha-fetoprotein (AFP) concentration was 6400 ng/ml. Serum levels of CEA and HCG were within normal limits. Percutaneous biopsy of the tumor strongly suggested yolk sac tumor with an evidence of AFP by an immunoenzyme labelling technique. The serum AFP rapidly decreased after two courses of combination chemotherapy. En bloc resection of the tumor was successfully performed and third chemotherapy was added. Mediastinal yolk sac tumors should be treated with combination chemotherapy and surgical resection.

Generation of O2- and tyrosine cation-mediated chemiluminescence during the fertilization of sea urchin eggs.

Sea urchin eggs emit light in the visible region during their fertilization. Judging from the chemiluminescence spectra, one of the excited species generated is considered to have originated from a tyrosine cation radical-mediated reaction. Chemiluminescence probes such as luminol or a cypridina luciferin analog (See text) are useful for detecting the ovoperoxidase +H2O2 reaction associated with membrane hardening and O2- generation, respectively, during fertilization of sea urchin eggs.

Detection of chemiluminescence in lipid peroxidation of biological systems and its application to HPLC.

A combined system of chemiluminescence detection and high performance liquid chromatography (CL-HPLC) was developed to determine primary peroxidation products in biological tissues, such as phosphatidylcholine hydroperoxide (PCOOH). The CL-HPLC assay consists of separation of lipid classes with HPLC and detection of hydroperoxide-specific chemiluminescence. Hydroperoxides react with heme compounds to produce oxidants as suggested by our early studies on tissue low-level chemiluminescence in which singlet molecular oxygen is generated as one of the excited species in several biological systems involving free radical events. In the CL-HPLC method, a cytochrome c-luminol mixture was used as a hydroperoxide-specific luminescent reagent, and the quantification of hydroperoxide was performed by detecting chemiluminescence due to the luminol oxidation caused by the oxidant produced during the lipid hydroperoxides with heme. The detection limit of PCOOH was 10 pmole hydroperoxide-O2. PCOOH in normal human blood was found to be 10-500 pmol/ml plasma and significantly higher levels of PCOOH were observed in some hospitalized patients.

Mild hemophilia A resulting from Arg-to-Leu substitution in exon 26 of the factor VIII gene.

Polymerase chain reaction amplification and nucleotide sequencing were used to identify the molecular defect in a Japanese patient with mild hemophilia A and an alteration of a TaqI site in exon 26 of the factor VIII gene. The mutation was a G-to-T transversion in codon 2326 of the factor VIII gene resulting in an Arg-to-Leu substitution at amino acid 2307 of the protein. The mutation, which is not of the common CG-to-TG type, is at the same codon in which both nonsense and a different missense (Arg to Gln) have previously been observed.

Weak ergodicity of population evolution processes.

The weak ergodic theorems of mathematical demography state that the age distribution of a closed population is asymptotically independent of the initial distribution. In this paper, we provide a new proof of the weak ergodic theorem of the multistate population model with continuous time. The main tool to attain this purpose is a theory of multiplicative processes, which was mainly developed by Garrett Birkhoff, who showed that ergodic properties generally hold for an appropriate class of multiplicative processes. First, we construct a general theory of multiplicative processes on a Banach lattice. Next, we formulate a dynamical model of a multistate population and show that its evolution operator forms a multiplicative process on the state space of the population. Subsequently, we investigate a sufficient condition that guarantees the weak ergodicity of the multiplicative process. Finally, we prove the weak and strong ergodic theorems for the multistate population and resolve the consistency problem.

Direct characterization of factor VIII in plasma: detection of a mutation altering a thrombin cleavage site (arginine-372----histidine).

An immunoadsorbent method has been developed for the direct analysis of normal and variant plasma factor VIII. Using this method, the molecular defect responsible for mild hemophilia A has been identified for a patient whose plasma factor VIII activity is 0.05 unit/ml, even though the factor VIII antigen content is 3.25 units/ml. Although the variant factor VIII has an apparently normal molecular mass and chain composition, the 92-kDa heavy chain accumulates when the variant protein is incubated with thrombin and the 44-kDa heavy chain fragment cannot be detected. In contrast, thrombin cleavage of the 80-kDa light chain to the 72-kDa fragment is normal. As these data indicate a loss of factor VIII cleavage by thrombin at arginine-372, the genetic defect was determined by polymerase-chain-reaction amplification of exon 8 of the factor VIII gene and direct sequencing of the amplified product. A single-base substitution (guanine----adenine) was identified that produces an arginine to histidine substitution at amino acid residue 372. These data identify the molecular basis of an abnormal factor VIII, "factor VIII-Kumamoto," that lacks procoagulant function because of impaired thrombin activation.

Culture and changes in dynamics of proliferation of pulmonary small cell carcinoma cells.

We had developed a method for the selective cultivation of cancer cells in short-term which was simple and required minimal technical skills. There have been so far 23 cases of pulmonary small cell carcinoma in which long-term passage of more than one year and establishment of a cell line were successful by the culture method. In particular, from November 1982 to March 1987, there were 29 patients who were diagnosed as having pulmonary small cell carcinoma on preoperative cytodiagnosis and whose cells were primary cultured. In 24 (83%) of these, isolation could be carried out in a short period and stable passage was possible. Long-term passage of more than one year and establishment of a cell line were successful in 21 (72%) of the 29 patients. As a result of primary culture of small cell carcinoma cells, a pattern of a suspended colony was observed in 22 (88%) of 25 cases. As passage advanced, the tendency toward attachment to the basal surface increased. The changes in dynamics of proliferation after long-term passage of more than one year were divided into three types: epithelial like cell type was observed in 8 (32%), the neurite like cell type was observed in 4 (16%) and the suspended colony type was observed in 13 (52%) of the 25 cases. Variant clones composed of relatively large cells appeared as a result of long-term passage or the use of carcinostatics in some pulmonary small cell carcinoma cells.

Intracellular compartmentalization of fura-2 dye demonstrated by laser-excitation fluorescence microscopy: a problem in measuring cytosolic free calcium concentration using fura-2 fluorescence in vascular smooth muscle cells.

Recently, we have developed a novel laser-excitation fluorescence microscope system to study intracellular calcium (Ca2+) in individual cultured vascular smooth muscle (VSM) cells using fluorescent indicators for (Ca2+). In the course of our study, it was shown that the subcellular fluorescence distribution of fura-2 was not homogeneous in VSM cells incubated with the acetoxy-methyl ester form of fura-2, fura-2/AM. The fluorescence appeared spotty or filamentous and resembled in shapes the intracellular organelles, suggesting that there was fura-2 dye compartmentalization in the organelles. To clarify the nature of the subcellular fluorescence, the soluble fraction of cells loaded with the dye was analyzed through high performance liquid chromatography (HPLC). We also examined the excitation spectra of fluorescence in the soluble fraction, which was compared with that in the cell suspension. Using HPLC, it has been shown that no other than fura-2 was found in the soluble fraction, whereas analyses of excitation spectra have indicated that the membrane fraction contained fura-2/AM or its lipophilic metabolite. On the other hand, indo-1 dye fluorescence showed a diffuse intracellular distribution, but the nuclear region had higher or sometimes lower fluorescence levels than the cytoplasm. The present results suggest that it may be necessary to assess subcellular fura-2 compartmentalization and possible interference by fura-2 AM or its lipophilic metabolite for the accurate measurement of intracellular Ca2+ concentration in VSM cells. It is also suggested that indo-1 may be more suitable for estimating Ca2+ concentration than fura-2 in individual VSM cells.

Dietary restriction augments superoxide generation in mouse peritoneal macrophages: an investigation using a chemiluminescence probe specific for superoxide anion.

It has not been clarified whether dietary restriction alters macrophage functions, although the augmentation of T cell functions by dietary restriction is well known. Forty percent dietary restriction on 9-week-old male C3H/He mice caused a decrease of body weight. However, one of the major macrophage functions, the generation of superoxide anion (O2), was augmented in proteose peptone-elicited peritoneal macrophages (MPs) from diet-restricted mice. This increase was more striking when the cells were stimulated by 12-o-tetradecanoylphorbol-13-acetate (TPA), which directly activated protein kinase C, than by opsonized zymosan which binded to receptors on the cells. These results strongly suggest that the augmentation of O2- generation in MPs by dietary restriction is due to the increased activity of protein kinase C which phosphorylate and activate O2-generating enzyme system NADPH oxidase. It is thought that one of the major factors for the reduced incidence of tumor and infection in diet-restricted animals is the augmentation of O2-generation in MPs.
