Relationship between HLA-D and in vitro and in vivo responsiveness to Candida allergen.

In vitro reaction to Candida allergen was studied in 100 normal healthy Japanese and related to HLA. A significant association was found between the low responder group (< 7,000 c.p.m.) and HLA-B15 and the high responder group (> 7,000 c.p.m.) and HLA-B7, as well as Dwl. Four HLA-D homozygote cells of types HLA-Dwl and DHO, were tested; these fitted in the high responder group. HLA-DYT and DEn fitted into the low responder group. Responsiveness to Candida allergen corresponded to skin test (r = 0.884, P < 0.01). From these results, in vitro lymphocyte reaction to Candida allergen and skin tests showed close correlation and an association with HLA antigens, HLA-D in particular.

Increase of IgA-bearing peripheral blood lymphocytes in families of patients with IgA nephropathy.

IgA-bearing peripheral blood lymphocytes, serum IgA, urinary sediments and HLA types of patients with IgA nephropathy and members of their families were examined to elucidate whether some familial factors might be related to the development of IgA nephropathy. Ten patients with IgA nephropathy, 31 family members and 36 age-matched healthy persons were examined. All families included certain members with increased amounts of IgA-bearing peripheral blood lymphocytes. The pattern of the emergence of family members with increased IgA-bearing lymphocytes was vertical. Some family members who had increased IgA-bearing lymphocytes showed microhematuria at the time of the study. There was no significant correlation between the amounts of IgA-bearing peripheral blood lymphocytes and levels of serum IgA. HLA types of the ten patients did not show significant deviation from those in the general population. It is suggested that the measurement of IgA-bearing peripheral blood lymphocytes among family members is useful for the screening of patients with IgA nephropathy.

Study on HLA system in IgA nephropathy.

Forty Japanese patients with IgA nephropathy were typed for HLA-A and HLA-B using local antisera. Thirty-seven of these patients were typed for HLA-D using homozygous cells "En", now known to be strongly related to DRw6. Forty-six patients with chronic hemodialysis were used as patient controls for renal disease and 115 healthy individuals were used as normal controls. The frequency of HLA-DEn was significantly increased in 17 out of 37 patients with IgA nephropathy, (46%) (normal controls 18.3%) (P less than 0.0007). Although the occurrence of HLA-B12 was high, it was not significant, i.e. 11 out of 40 (27.5%). These results suggest that some abnormality in the genes of patients with IgA nephropathy is more closely associated with the HLA-D region than the HLA-A or B regions.

Further study on HLA-A, B, C, D, DR and haplotype antigen frequencies in psoriasis vulgaris.

HLA-A, B, C, D, and DR antigens in 40 Japanese patients with psoriasis vulgaris were analysed using the antisera obtained from the Seventh International Histocompatibility Workshop. The most deviant HLA antigens in psoriasis were found to be HLA-Cw6 and DRw7. by observing the HLA-A, B, C and DR antigens en bloc in psoriasis, it is shown that the statistically significantly associated haplotype antigens were HLA-A1, B13, Cw6 and DRw7, and A1, B37, Cw6 and DRw6 respectively. No HLA-Dw11 (LD17) antigen was associated with the disease.

Stabilization of sulfisomidine tablets by use of film coating containing UV absorber: Protection of coloration and photolytic degradation from exaggerated light.

The effect of model polymer coating films of vinyl acetate, containing oxybenzone as a UV absorber, on the coloration and photolytic degradation of simple sulfisomidine tablets was examined to attempt stabilization of photosensitive solid dosage forms. Coloration of the tablet surface was followed by the tristimulus colorimetric method in the fading tester equipped with a mercury vapor lamp. Photolytic degradation in the UV region was investigated by a new method for measuring the absorption spectra of a crystal sample in the gas phase, i.e., the semi-integral attenuance spectra. Two parameters of a film, thickness and concentration of the UV absorber, were varied at every exposure. These physical and chemical changes are discussed in relation to light transmission properties of films.

In vitro synthesis of ornithine transcarbamylase.

An in vitro system for the synthesis of ornithine transcarbamylase (OTCase) was established using iS-30 extract from E. coli MDS6-2(lambda) and DNA of a lambda transducing phage carrying argI and argF genes. This in vitro synthesis was completely dependent on the additon of DNA, and was sensitive to chloramphenicol and rifampicin. Radioisotopic analysis confirmed that the synthesized enzyme catalyzes the carbamylation of ornithine to citrulline. In the in vitro system the repression and derepression of OTCase synthesis could be observed by mixing iS-30 extracts prepared from argR+ and argR- cells. A remarkable maturation effect could be observed for the FFF enzyme, but not for the III enzyme. This system is considered to reflect the in vivo situation, and should therefore be useful for investigations on the regulation of OTCase synthesis in vivo.

Possible associations between HLA antigens and the immune responsiveness to attenuated rubella vaccine.

We investigated the possible associations between HLA antigens and the antibody response patterns during a clinical trial with a live attenuated rubella vaccine in 172 Japanese schoolgirls. Those vaccine recipients were divided into three groups, 42 low responders, 102 intermediate responders and 28 high responders, according to convalescent-phase hemagglutination-inhibiting antibody titers. High frequencies of HLA--A11 and HLA--B15 in high responders and HLA--Aw24 and HLA--B5 in low responders were found. Among HLA--A antigens HLA--A11 had the highest geometric mean HI antibody titer and HLA--Aw24 had the lowest. Among HLA--B antigens HLA--B15 had the highest and HLA--B5 had the lowest. Strong linkage disequilibrium was found between HLA--A11 and HLA--B15 in high responders and between HLA--Aw24 and HLA--B5 in low responders. These results suggest that there may be a relationship between HLA antigens and specific immune response genes in man as well as in other vertebrates.

Synthesis and processing of an Escherichia coli alkaline phosphatase precursor in vitro.

Alkaline phosphatase [orthophosphoric-monoester phosphohydrolase (alkaline optimum), EC 3.1.3.1] of E. coli was synthesized in a cell-free system, and the size of the direct translation product was analyzed. The product has a higher molecular weight than the mature alkaline phosphatase found in the periplasm. The direct translation product can be processed to the mature size by an E. coli membrane fraction; the processing activity copurifies with the outer-membrane fraction. The presumed precursor can dimerize to form active enzyme without being processed, and the resultant enzyme appears to be more hydrophobic than the mature enzyme. These findings are discussed in connection with the "signal hypothesis" proposed for the excretion of proteins across membranes.

HLA antigens and Ia antibody in the gastric cancer patient.

The possible association of 18 HLA specificities in HLA-A locus and 23 specificities in HLA-B locus with susceptibility to gastric cancer was investigated in a total of 87 patients and 355 controls in Japan. Significantly decreased antigen, HLA-B5 (17.2%) was found in gastric cancer patients compared with 40.0% in controls. X2 for this antigen difference is 15.8 (corrected P less than 0.005). The most frequent HLA haplotype was A9-B5 in controls and A2-Blank B locus antigen in gastric cancer patients. No reaction was found of HLA antiserum, Bou-Pri with cells of gastric cancer patients, compared with 29.4% in control subjects. No linkage was observed between HLA-A and -B locus antigens and gastric cancer in family study with double the frequency of HLA haplotype A2-BW40. Ia-like B cell specific antibody was found 10% in 2 out of 20 gastric cancer patients in a preliminary study. There was no clear association between B cell specific antibody and clinical implications.

Studies on monoterpene glucosides and related natural products. XXXI. Gas chromatography and gas chromatography-mass spectrometry of iridoid and secoiridoid glucosides.

A total of 33 iridoid and secoiridoid glucosides were detected by gas chromatography on several columns such as OV-1 or OV-17. Representative glucosides were then subjected to gas chromatography-mass spectrometry, giving some characteristic peaks that permitted the discrimination of both types of glucosides from other compounds in most instances. The successful detection of both types of glucosides in several plant extracts showed the applicability of this combination of methods to small amounts of plant materials.
