Memory impairment and morphological changes in rats after continuous infusion of active fragment of anti-nerve growth factor-antibody.

We report here that the specific Fab' fragment of anti-nerve growth factor (NGF)-antibody (anti-NGF, 12, 120 and 400 micrograms/4 weeks, i.c.v.) impairs learning and memory. The goal latency of the control rats in water maze task was rapidly shortened by training compared to those of the anti-NGF-treated rats. The degree of reduction in movement counts of the anti-NGF-treated rats in habituation task was significantly smaller than that of the control rats. However, the step-through latency of the anti-NGF-treated rats was not significantly shorter than that of the control rats. With regard to the choline acetyltransferase activity, no effects were observed in any of the brain regions. Anti-NGF treatment altered nuclear morphology in the hippocampus and parietal cortex. As a result, it seems that the anti-NGF-induced amnesia could be due to an impairment of nuclear morphology.

T cell subsets (Tc, Th, Ts, Tsi) and IL2 receptor-bearing cells in peripheral blood of patients in the acute phase of alcoholic hepatitis.

The present study was designed to evaluate the proportion of T cell subsets and IL2 receptor positive cells in the peripheral blood of patients with acute alcoholic hepatitis (AAH) using monoclonal antibodies to T cell antigens and Tac in a double immunofluorescent technique. The results indicate that the percentage of the total T cells and the intensity of all the T cell antigens are significantly reduced and the percentage of T helper cells and the ratio between Th/Ts cells are significantly increased in AAH when compared to healthy controls. But, significant differences in values observed in this study were not found for the absolute number of these cells. These altered values returned to normal levels during convalescence.

The effect of hydroxyapatite coating on bone growth into porous titanium alloy implants.

In rabbits and goats, test implants with a porous surface of two layers of Tl-6A;-4V beads were examined at intervals for bond strength with bone. Half of the implants were coated with hydroxyapatite by plasma spray. The bonding strength with bone in the coated specimens was about four times greater than that of the uncoated specimens at two weeks, and twice as strong at six weeks. Twelve weeks after implantation, the strengths were similar. The hydroxyapatite coating of the beads provided earlier and stronger fixation.

[Cytotoxicity of mitomycin C in rabbit eyes].

The cytotoxic effects of mitomycin C (MMC) on intact rabbit eyes were studied using light microscopy and transmission electron microscopy. A volume of 0.1 ml MMC solution (0.4 mg/ml) was given to rabbits subconjunctivally once a day. The eyes were enucleated at 1, 2, 3, and 4 weeks after subconjunctival injection. MMC showed considerable cytotoxicity in most tissues of the rabbit eyes, such as the cornea, the iris, especially to the choroid and the ciliary body. However, MMC cytotoxicity in the sclera was slight. Based on the above results it is concluded that MMC should not be used when the cornea or sclera is invaded, for example after pterygial resection, and scleral necrosis due to MMC could occur due to the damage of alimentary vessels of the sclera.

[Circulating mature NK (Leu11+) cells positive for IL2 receptor and HLA-DR in patients with various liver diseases and asymptomatic HBsAg carriers].

IL2R+ Leu11+ cells (A) and HLADR+ Leu11+ cells (B) of peripheral blood in patients with various liver diseases and ASCs were measured using double colour immunofluorescence assay of MoAb with FACS flow cytometry. 1) The mean % of IL2R+ Leu11+ cells which was 0.5 +/- 0.2 in healthy controls, decreased significantly in HCC in comparison with CALD, ASC and healthy controls, and in ASC rather than in healthy controls. They (A) were less than 0.1% in eight of thirteen cases with HCC, in one of twenty cases with CALD and of eleven ASCs, respectively. 2) In the mean % of HLADR+ Leu11+ cells which was 2.9 +/- 1.8 in healthy controls there was not a significant difference among HCC, CALD, ASC, AH and healthy controls. They (B) were less than 0.1% only in one case with HCC. 3) The value of fluorescence intensity of IL2R on IL2R+ Leu11+ cells reduced in B-CALD, ASC, AH and HCC, and one of HLADR on HLADR+ Leu11+ cells increased in CALD. These results suggested that the decrease of IL2R+ Leu11+ cells was due to the existence of HCC.

Regulation of the temporal synthesis of proteins in bacteriophage BF23-infected cells.

Regulation of temporal synthesis of pre-early, early, and late proteins in bacteriophage BF23-infected cells has been studied by using five amber mutants defective in genes 1, 2, 10, 14, and 19. The synthesis of pre-early proteins is negatively regulated by the actions of gene 1, a pre-early gene. The switch from pre-early to early protein synthesis is mainly regulated by the second-step DNA transfer reaction, which is controlled by at least genes 1 and 2. Early proteins can be kinetically and genetically divided into two regulatory classes, designated Ea and Eb. The shutoff of Eb-early protein synthesis is associated with the turn-on of late protein synthesis. This step is controlled by genes 10, 14, and 19. Gene 10 also regulates negatively the synthesis of Ea-early proteins, indicating that this gene has a dual function in the regulation of early protein synthesis. The temporal synthesis of phage-encoded proteins is regulated mainly at the transcriptional level. Evidence is presented indicating that the host RNA polymerase is modified by the interaction with the gene products of genes 2, 10, and 14 (gp2, gp10, and gp14, respectively). gp2 interacts with the enzyme in the earlier stage of infection but is replaced by gp10 in the later stage. This exchange reaction depends on the presence of gp14 and gp19 and is related to the switch from Eb to late protein synthesis. Thus, the regulation of BF23 gene expression occurs in a coordinated manner throughout the development of this phage.

Computed tomography for preoperative evaluations of lung cancer.

Computed tomography was evaluated for its accuracy in diagnosing mediastinal node metastases, direct chest wall invasion, and direct mediastinal invasion by lung cancer among 61 patients who subsequently underwent surgery. Using 15-mm diameter or larger mediastinal lymph nodes as the criterion for metastasis, the sensitivity was 36% (8/22); the specificity was 92% (34/37). The accuracy for direct chest wall invasion was relatively high, with a sensitivity of 100% (7/7) and a specificity of 92% (22/24). Direct mediastinal invasion had a sensitivity of 67% (4/6) and a specificity of 91% (10/11). These results suggest that the ability of computed tomography to diagnose mediastinal lymph node metastasis when such nodal size is used as a criterion is limited.

[A case of Werner's syndrome with neurogenic bladder].

The present report is Werner's-syndrome associated with a neurogenic bladder. The patient visited our clinic with the chief complaint of retention of urine and urinary tract infections. The diagnosis of Werner's syndrome was made because the patient had such clinical features as bird-like or "masked" face, short stature, graying of the hair in youth, cataracts in youth, skin changes like scleroderma principally on limbs, early menopause. After trans-urethral-resection of bladder neck and administration of distigmine bromide, her bladder function was improved, she could urinate by herself and she was free from urinary tract infections.

[Allergic reaction to CDDP: report of 4 cases].

Four cases of CDDP allergic reaction (CDDP reaction) are reported. CDDP was given intravenously with vinka-alkaloid and bleomycin or cyclophosphamide (CDDP combination therapy). Case 1 was a 59-year-old man with left ureteral squamous cell carcinoma. The tumor was so large and infiltrated into the retroperitoneal space, that he had conservatively undergone only left nephrectomy and tumor biopsy. Post-operative CDDP combination therapy was performed. Soon after the beginning of the 6th CDDP drip-infusion, a reaction to CDDP occurred. Case 2 was a 22-year-old man diagnosed to have left testicular embryonal carcinoma with multiple pulmonary metastases. After radical orchiectomy and retroperitoneal lymphadenectomy, CDDP combination therapy was performed. At the beginning of the 8th CDDP drip-infusion, a reaction to CDDP occurred. Case 3 was a 49-year-old man diagnosed to have non-papillary bladder carcinoma. Although his bladder was tumor free by TUR-Bt, distant lymph node metastases occurred. CDDP combination therapy was performed. At the beginning of the 6th CDDP drip-infusion, a reaction to CDDP occurred. Case 4 was a 64 year-old-man diagnosed to have bladder carcinoma which was recurrent from right renal pelvic tumor. After total cystectomy and ileal conduit, CDDP combination therapy was performed for diffuse retroperitoneal lymph nodes metastases. At the beginning of the 8th CDDP drip-infusion, a reaction to CDDP occurred. Reaction to CDDP developed at the 6th to 8th course of combination chemotherapy with CDDP, i.e., 450 to 700 mg of CDDP in total doses.(ABSTRACT TRUNCATED AT 250 WORDS)
