Reaction of human smooth muscle autoantibody with foetal glial cells and their precursors.

Tissue culture monolayers of dissociated human and rat embryonic brains were examined by indirect immunoflurescence with human serum containing smooth muscle antibody. Flat, epithelioid cells showed staining of a fine, filamentous network in the cell body while multipolar cells showed diffuse cytoplasmic fluorescence of the cell body and processes. Both cell types were identified as glial cells by silver stains. Specificity of the immunofluorescent staining reaction was established by failure to obtain staining with normal serum, with smooth muscle antibody serum neutralized by homogenates of smooth muscle or by extracts contining actin-like protein derived from smooth muscle. Reacitivity of of embryonic glial cells with smooth muscle antibody indicates the presence of actin-like proteins which are probably located in cytoplasmic microfilaments.

Smooth muscle-associated contractile protein in human and experimental acute leukaemias.

Cells from ten human myeloblastic leukaemias and from spontaneous rat myeloblastic and lymphoblastic leukaemias were examined by indirect immunofluorescence with human serum containing smooth muscle antibody. Strong positive cell outline staining of blast cells was seen in cryostat sections of rat leukaemic lymph nodes, spleen and liver. In cell smears of human and rat leukaemias, cell outline fluorescence was restricted to cells in close contact with each other. Membrane immunofluorescence tests of suspensions of viable leukaemic cells were negative. Specificity of the immunofluorescent staining reaction was established by failure to obtain staining with normal serum, or with smooth muscle antibody serum neutralized by homogenates of smooth muscle or extracts containing actin derived from smooth muscle. These observations suggest that smooth muscle-associated microfilaments are present in leukaemic blast cells and that expression of the antigen is dependent on cell-cell contact. The presence of this antigen may facilitate leukaemic infiltration of tissues.

Antiserum specific for reticulin of the bursa of fabricius.

Antiserum produced by immunizing rabbits with embryonic chicken spleen cells showed, after appropriate absorption with cell suspensions, specificity for fetal spleen cells in membrane immunofluorescence tests. However, when the original antiserum was absorbed with tissue homogenates and tested on cryostat tissue sections, it reacted specifically with the reticular framework which separates the cortical and medullary lymphocytes in the follicles of the adult bursa of Fabricius. It is suggested that the reticulin may influence bursal lymphoid differentiation.

Correlation of regional lymph node in vitro antitumor immunoreactivity histology with colorectal carcinoma.

Lymph nodes from resected specimens of human colorectal carcinoma were investigated for in vitro lymphocyte cytotoxicity against primary cultures of autologous tumor cells. Regional lymph node lymphocytes were cytotoxic in 32 of 142 cases (23%). Altogether 200 nodes were examined and the cytotoxicity correlated directly with sinus histiocytosis, seen in 43 nodes from 35 cases, and with hyperplasia of B- and T-lymphocyte areas combined, seen in 92 nodes from 65 cases. Lymph nodes with combined B- and T-cell hyperplasia were significantly more common in cases of good tumor differentiation. The findings suggest that sinus histiocytosis and hyperplasia of both major lymphocyte populations are morphological expressions of in vitro antitumor immunoreactivity in the regional lymph node.

Lymphoreticular response to a syngeneic rat tumour: gravimetric and histological studies.

Gravimetric and histological studies of lymphoreticular tissues during growth of a syngeneic squamous cell carcinoma in Wistar rats show that the regional lymph node anergy reported previously in this system is associated with replacement of paracortical small lymphocytes by large blastoid cells. The regional node continued to gain weight throughout the period of anergy and showed no atrophy and minimal necrosis. Over the same period the spleen increased in both weight and cytotoxicity.

Smooth muscle-associated antigen in experimental cutaneous squamous cell carcinoma, keratoacanthoma, and papilloma.

Cryostat sections of 29 squamous cell carcinomas, 13 keratoacanthomas, and 12 papillomas, induced by 7,12-dimethylbenz(a)anthracene in the skin of rabbits and rats were examined by indirect immunofluorescence with human serum containing antibody to smooth muscle. Linear or granular staining of the cell outlines of the basal squamous cell layers was seen most extensively in the carcinomas, less in keratoacanthomas, and least in papillomas. In addition, squamous cell carcinomas showed this pattern of staining at advancing tumor margins and in invasive cords and tumor cell nests in the dermis. Four keratoacanthomas also showed prominent staining of the basement membrane area. The specificity of the staining reaction was established by its prevention on neutralization absorptions of the serum with extracts or homogenates of smooth muscle. The epidermal cells of normal rabbit and rat skin gave negative staining reactions. The presence of smooth muscle-associated antigen probably corresponds to cellular microfilaments.

Specific immune response in human skin carcinoma.

Eight out of nine patients with squamous cell carcinoma of skin have shown immunological reactivity against their own tumour cells by one or more tests with their sera or peripheral blood lymphocytes. The tests included membrane and cytoplasmic immunofluorescence, and, with cultured tumour, complement-dependent serum cytotoxicity and lymphocyte attack. One case examined in depth had an unusually conspicuous lymphocyte and plasma cell reaction on histological examination, and was positive by all four tests; a time-lapse cinephoto-micrographic record over seven days was obtained of the attack on the carcinoma cells in culture by the patient's lymphocytes.
