Molecular parameters of three GABA-ergic agonists.

A comparison of the molecular parameters of gamma-aminobutyric acid (GABA), the hydroxamic acid of GABA (HAGABA), and muscimol has been made using structural data and partial charges from the AM1 semiempirical quantum mechanical method. It was concluded that HAGABA was more closely related to GABA than muscimol.

The actions of the hydroxamic acid of gamma amino butyric acid on the isolated rabbit atria and jejunum.

The hydroxamic acid of gamma amino butyric acid (HAGABA) has been prepared and was studied in the isolated rabbit atria and jejunum. HAGABA did not interfere with the spontaneous contractile rhythm and frequency of the isolated atria except in large mM concentrations in the bath. HAGABA displayed a biphasic effect in the isolated jejunum, which manifested itself first as stimulation followed by inhibition with an increasing concentration in the bath. Bicuculline and HAGABA did not antagonize each other in the enteric nervous system, but rather an augmentation was observed.

Structure-activity studies of hydroxamic acids as direct vasodilators.

A series of beta-hydroxylamine-phenylpropyl hydroxamic acids were prepared. The ability of these compounds to act as hypotensive agents was studied pharmacodynamically. Key structural features for the hypotensive action were identified and correlated to the degree of responses observed. It was ascertained that these compounds do not act through the adrenergic, cholinergic and histaminergic receptor system, but can be classified as direct acting vasodilators.
