[Percutaneous absorption of hematoporphyrin derivative--fluorescence- and electron microscopic studies].

Using surviving human skin, percutaneous absorption following topical application of hematoporphyrin derivative was investigated. Hematoporphyrin derivative gave strong red fluorescence, which was dependent from it's actual concentration, from the used kind of vehicle, from preparation time, and from time between preparation and application of hematoporphyrin derivative solution. Ultrastructural investigation verified cytoplasmatic membranes and mitochondria as the principal targets also after topical application followed by irradiation with visible light.

Different ultrastructural feature of keratinocytes in human malignant melanomas.

The ultrastructural feature of keratinocytes was investigated in human superficially spreading melanomas of type B. The tumor cells of these melanomas are characterized by spheroidal melanosomes and containing pheomelanins. Contrary to type A melanomas the keratinocytes show no striking transformation reaction neither in the nucleus nor in the cytoplasm although this part of the cell mostly contains abundant melanin granules surrounding the nucleus. Whether there exists a relation to the clinical course still has to be clarified.

[The ultrastructure of interdigitating cells in atopic eczema].

Electron microscopy was used in investigations of interdigitating reticulum cells of axillary lymph nodes sampled from patients with atopic eczema in different clinical stages. The ultrastructure of these cells was found to differ clearly from that of cells under largely normal conditions. For example, the lumina of the so-called tubulovesicular structures (TV) were clearly reduced. Considerable lobulation of peripheral cytoplasm was an additional finding recorded from the acute clinical stage, while in terms of cellular morphology (interdigitation) contact with T-lymphocytes was obviously reduced. Cytoplasmic processus were clearly decreased in the chronic stage.

Unusual mitochondrial reaction in psoriatic keratinocytes.

Electron microscopic investigation of skin biopsies revealed that keratinocytes in clinically uninvolved skin from psoriatic patients show the same mitochondrial ring phenomenon after the application of dithranol as has been observed in the cutaneous lesions proper. In various other skin diseases no mitochondrial reaction was evident after dithranol treatment, nor was it seen in chronic dermatitis when dithranol was not applied. It appears, therefore, that this morphological phenomenon is specific to psoriasis and may provide information concerning functional alterations in the pathogenesis of this disease.

Ultrastructural features of Langerhans cells in clinically uninvolved skin of psoriatics.

Epidermal Langerhans cells (LC) from skin of clinically normal appearance in psoriatics were studied electron microscopically and compared with LC from the skin of healthy volunteers and from those with other dermatoses. Results show that LC from psoriatics display partly distinct cytoplasmic alterations. Moreover, about half of LC investigated were partly or mainly surrounded by clearly widened intercellular spaces, such that an immediate cellular contact with neighbouring keratinocytes was correspondingly decreased. Whether such LC are functionally altered remains to be clarified.

[On the clinical aspects and ultrastructure of bullous pemphigoid (parapemphigus) of the oral mucous membrane (author's transl)].

Bullous changes in the oral mucosa are often the first signs of a general dermatological disease. Successful diagnosis of such diseases is largely dependent upon the use of complex laboratory methods. Clinical and ultrastructural changes are described and discussed with particular reference to a case of parapemphigus. Atypical intracellular vacuolation and marked acantholysis observed in electron micrographs is interpreted as a secondary phenomenon of direct blistering.

Aged

[Fabry disease: clinical, biochemical and electron microscopical studies (author's transl)].

Biochemical investigations on blood-plasma and serum as well as well as electron microscopic investigations on skin lesions biopsy specimens were carried out in one case of morbus Fabry 35 years of age distinctly showing cardio-renal symptoms. The diagnosis of Fabry disease was made when the patient was hospitalized for treatment of a chronic glomerulonephritis. The patients' serum alpha-galactosidase activity was reported to be 10 to 13% of normal controls, whereas the values of trihexosyl-ceramids for blood-plasma and serum were increased. Electron microscopic studies revealed that the endothelial cells and pericytes of the cutaneous vessels as well as the fibroblasts contain numerous liposome-like cytoplasmic inclusions of different size showing an internal lamellar structure with exact periodizity of 65 A. These inclusion are not surrounded by a membrane structure, however, it is possible that the arise from defect lysosomes which accumulate trihexosyl-ceramids owing to insufficient of alpha-galactosidase activity.

Ultrastructure and function of interdigitating cells in the guinea pig thymus.

Electron micrsocopic investigation of the thymus of normal guinea pigs of both sexes, approximately 3 months of age, have shown that the organ contains at least two types of non-lymphoid cells: epithelial reticulum cells and so-called interdigitating cells. The latter have been observed in the inner part of the cortex and are morphologically characterized by an irregularly shaped or kidney-shaped nucleus, many tubulovesicular cytoplasmic structures accumulated mostly within the cytocentre as well as by finger-like protrusions of the cytoplasm. These interdigitating cells are in close contant with neighbouring lymphocytes with partly destroyed pycnotic nucleus.

[Contribution to the epithelial tumors of the ciliary body (clinical, histopathological and electronmicroscopical observation) (author's transl)].

An adenoma of the non pigmented epithelium of the ciliary body was supposed to be an irisciliary body melanoma (32P-test positive) and was treated by irido-cyclectomy. Vision acuity remained 0,4 one year after operation. The tumor formed adenoid tubes lined with a cubical or cylindrical epithelium. The cytoplasm of all tumorcells was threaded with compactly arranged tonofilaments. The histopathological examination showed unequivocal the benignant character of this tumor. Nevertheless the tumorcells were infiltrating the deep scleral layer in one place. Also the lens was deformed by the tumor and developed a local opacity in the periphery. It is refered to the difficulty of a clear histopathological distinction between adenomas, adenocarcinomas and pseudo-adenomatous hyperplasia of the ciliary body (Fuchs adenoma).

[Electron microscopic representation of the pili structure of Neisseria gonorrhoeae (author's transl)].

The technique of negative staining and ultra-thin section has been used for investigations of 30 Neisseria gonorrhoeae strains in order to represent the structure of pili (fimbriae) electron microscopically. The staining of the gonococci was effected by phosphotungstic acid (0,5%). The pili ascertained were 30 to 60 A thick. In course of in vitro passages up to 10. subculture morphological changes of the pili have been observed. The application of trisbuffer or solution of Hylase (hyaluronidase) showed not any improved results in comparison with buffered NaCl-solution as suspension medium. The investigation of ultra-thin sections showed that the structure of the pili could be exhibited not clearly. Therefore, these technique seems to be not suitable for qualitative representative of the pili.
