[Analysis of HLA-DQWa antigen ethnic polymorphism among a Japanese and Caucasian population and an association with Harada's disease].

A novel HLA-DQ antigen, DQWa was analysed with its serological, immunochemical, and molecular biological polymorphism. Furthermore, A disease association with DQWa was analysed by investigating HLA antigen frequencies of 24 patients with Harada's disease. DQWa was in linkage disequilibrium mainly with DR4-Dw15 in Japanese, and with DRw8 -Dw8 in Caucasians. The former haplotype was observed only in Japanese. A significant increase of DQWa was observed in the patients with Harada's disease (73.9% compared with 35.9% of the control, chi 2 = 9.77, P less than 0.005, Pc less than 0.05, Relative risk = 5.17). DQWa antigen molecules were different from each other between Japanese and in Caucasians, when they were analysed immunochemically by two dimensional electrophoresis, or molecular biologically by restriction fragment length polymorphism analysis. These results suggest that HLA-DQWa antigen may play a role in the pathogenesis of Harada's disease, which is seen only in Japanese.

Inhibition of autologous mixed lymphocyte reaction by monoclonal antibodies specific for the beta chain of HLA-DR antigens.

Recent studies using rabbit antisera to the separated HLA-DR alpha and beta subunits have suggested that alpha chain-specific, but not beta chain-specific, antisera inhibit T cell proliferative responses in primary and secondary human autologous mixed lymphocyte reaction (AMLR). In the present study, with the aid of sequential co-precipitation assays and Western blotting methods, a monoclonal rat alloantibody 1E4, specific for the beta chain of rat class II molecules carrying an Ia determinant Ba-2.7, was characterized to recognize a monomorphic determinant located on the beta chain of DR antigens. This antibody and a murine monoclonal antibody HU-4, also specific for the beta chain of DR antigens, strongly inhibited both primary and secondary AMLR through a mechanism distinct from an antibody-dependent cell-mediated cytotoxicity reaction. These results indicate that the inhibition of AMLR is not a unique feature of DR alpha-specific antibodies.

Myocardial metastasis from primary lung cancer: myocardial infarction-like ECG changes and pathologic findings.

Myocardial metastasis from neoplastic disease is often clinically unapparent, and very difficult to diagnose. Of 151 consecutive autopsies of lung cancer patients, cardiac metastases were found in 67 patients (44.4%). Myocardial metastasis was found in only 8 patients (11.9%). ECG of patients with myocardial metastasis revealed ST-T wave changes and various types of arrhythmia. ST-T wave changes were observed in 4 with myocardial metastasis, and in 6 without myocardial metastasis (pericardial metastasis alone). ST-T wave changes is not a specific finding of myocardial metastasis. Two very rare cases with myocardial metastasis showing progressive ST segment elevation with a QS pattern are presented. The appearance of ST segment elevation with a QS pattern in clinically stable lung cancer patients without cardiac symptoms suggestive of myocardial injury indicates the possibility of myocardial metastasis. Myocardial metastasis is often elusive, thus careful observation of ECG changes is of primary importance for the antemortem diagnosis.

Effect of taxol on vinblastine sulfate-induced crystallization of tubulin.

In human lung adenocarcinoma cells exposed to vinblastine (VLB) and stained with fast green, large tubulin crystals were seen in the cytoplasm. In contrast cells exposed to taxol had prominent bundles of stabilized microtubules. Pre- or co-incubation of cells with taxol before exposure to VLB prevented formation of the crystals. However, VLB did not prevent formation of stabilized microtubules by taxol if added at the same time. Under a fluorescent microscope, VLB induced crystals stained with secondary antibodies against anti-alpha- and anti-beta- tubulin antibodies. Neither of the drugs altered binding of the antibodies to these crystals.
