Use of intravaginal progesterone-releasing inserts in a synchronization protocol before timed AI and for synchronizing return to estrus in Holstein heifers.

Holstein heifers (n = 189) were submitted to a 42-d artificial insemination (AI) period in which they underwent AI after once-daily evaluation of rubbed tail chalk. At the onset of the AI period (d 0), heifers were assigned randomly to receive synchronization of ovulation and timed AI (TAI; d 0: 100 mug of GnRH; d 6: 25 mg of PGF(2alpha); d 8: 100 mug of GnRH + TAI) either without (GPG; n = 95), or with inclusion of a CIDR insert (CIDR; n = 94) from d 0 to 6. No CIDR heifers received AI before d 8 compared with 24% of GPG heifers, and pregnancy rate per AI (PR/AI) at 30 d after TAI did not differ between treatments. To synchronize return to estrus for heifers failing to conceive after TAI, heifers (n = 166) receiving TAI to first service were randomly assigned to receive no further treatment (control; n = 85) or receive a new CIDR insert between 14 and 20 d after TAI (Resynch; n = 81). No Resynch heifers received AI during CIDR treatment compared with 35% of control heifers, and the proportion of heifers receiving AI within 72 h after the day of CIDR removal was 78 vs. 50% for Resynch vs. control heifers, respectively. No treatment x inseminator interaction was detected at first or second AI; however, overall PR/AI was modest for heifers throughout the experiment due to poor performance of 2 of the 3 herd inseminators (14, 6, and 58% PR/AI, respectively). Inclusion of CIDR inserts suppressed estrus during the TAI protocol with no reduction in PR/ AI. Resynchronization of estrus using CIDR inserts resulted in tighter synchrony of estrus among nonpregnant heifers compared with untreated controls.

Relationship between level of milk production and multiple ovulations in lactating dairy cows.

Our objective was to evaluate factors associated with spontaneous multiple ovulations in lactating dairy cows. Ovaries of cows [n = 267; >50 days in milk (DIM)] were evaluated weekly using ultrasound to determine spontaneous (i.e., no hormonal treatment) ovulation rate starting at 50 DIM and continuing until pregnancy diagnosis. Cows were fitted with a transmitter to record standing activity during estrus, and serum progesterone concentration was assessed weekly starting at wk 1 postpartum for all cows. Overall, 76 (28.5%) cows were anovular and 191 (71.5%) were ovular by 71 DIM. Incidence of anovulation was not associated with level of milk production but was associated with lower body condition. For anovular cows (n = 41) that spontaneously recovered, the multiple ovulation rate at first ovulation was 46.3%. For second and subsequent ovulations (n = 463), the level of milk production for 14 d preceding estrus was associated with increased ovulation rate. To illustrate, incidence of multiple ovulations was 1.6% (2/128), 16.9% (32/189), and 47.9% (70/146) for ovulations when cows were producing <35, 35 to <45, and >or=45 kg/d, respectively. Among cows for which estrous behavior was recorded, those with multiple ovulations (n = 48) had shorter duration of estrus (4.3 +/- 0.7 vs. 9.9 +/- 0.5 h) and higher production (47.2 +/- 0.9 vs. 38.1 +/- 0.5 kg/d) than cows with single ovulations (n = 237). Circulating concentrations of estradiol were lower (5.5 +/- 0.3; n = 15 vs. 7.8 +/- 0.4 pg/mL; n = 71) during periods of estrus with multiple ovulations despite a greater preovulatory follicular volume (4136 +/- 123 vs. 3085 +/- 110 mm(3)). Similarly, serum progesterone concentration 7 d after estrus was lower for cows with multiple than single ovulations (2.5 +/- 0.3 vs. 3.2 +/- 0.1 ng/mL) despite a greater luteal volume (8291 +/- 516 vs. 6405 +/- 158 mm(3)). In summary, the first spontaneous ovulation in anovular cows and a higher level of milk production for 14 d preceding estrus were associated with increased multiple ovulation rate. Additionally, cows with multiple ovulations had lower estradiol at estrus, a shorter duration of estrus, and lower progesterone at 7 d after estrus than cows with single ovulations.

Effect of dietary phosphorus concentration on estrous behavior of lactating dairy cows.

The objective of this study was to determine the effect of dietary phosphorus (P) concentrations of 0.38 (adequate) or 0.48% (excess) of the total mixed ration (TMR) (dry matter basis) on estrous behavior of lactating cows as measured by a radiotelemetric system (HeatWatch; De Forest, WI, USA). At calving, 42 Holstein cows (n=21 per treatment) were randomly assigned to one of two dietary P treatments. Cows were milked twice daily and milk weights were recorded. Cows were housed in a free-stall barn and were fitted with a radiotelemetric transmitter 40 days postpartum to record estrous mounting activity. The total number of estruses recorded for the 42 cows were 72 (37 and 35 for cows in the adequate and excess P groups, respectively). The mean duration of estrous cycles was 22 +/- 0.6 days and 21 +/- 0.4 days for cows fed the adequate and excess P diets, respectively (P=0.14). The mean duration of estrus was 8.9 +/- 1.1 h and 8.6 +/- 1.2 h (P=0.86), the average number of mounts during estrus was 7.0 +/- 1.2 and 8.2 +/- 1.7 (P=0.57), and the total mounting time was 27.1 +/- 4.3 s and 30.8 +/- 6.5 s (P=0.64) for cows fed the adequate and excess P diets, respectively. Phosphorus treatment had no significant effect on intensity or duration of estrus.

Relationship between level of milk production and estrous behavior of lactating dairy cows.

The objective of this study was to determine if there is an association between level of milk production and duration of estrus as determined by standing activity recorded by a radiotelemetry system. Holstein cows (n = 267; 50 DIM) were fitted with a transmitter that allowed continuous recording of standing activity. Cows were housed in a free-stall barn and milked twice daily. Ovulation was confirmed for all estruses (n = 380). Average milk production for the 10 days before the day of estrus was used to classify cows as lower (< 39.5 kg/day) or higher (>/= 39.5 kg/day) producers at the time of estrous expression. Follicle size and serum estradiol (E(2)) concentrations were determined in a subset of cows (n = 71) on the day of estrus. Duration (6.2 +/- 0.5 h versus 10.9 +/- 0.7 h; P < 0.0001), standing events (6.3 +/- 0.4 versus 8.8 +/- 0.6; P = 0.001), and standing time (21.7 +/- 1.3 s versus 28.2 +/- 1.9 s; P = 0.007) were shorter for estruses from higher (46.4 +/- 0.4 kg/day; n = 146) than lower producers (33.5 +/- 0.3 kg/day; n = 177). Milk production was correlated with the duration of estrus (r = -0.51; P < 0.0001; n = 323). Higher producers had lower E(2) concentrations than lower producers (6.8 +/- 0.5; n = 31 versus 8.6 +/- 0.5 pg/ml; n = 40; P = 0.01) in spite of larger pre-ovulatory follicle diameter (18.6 +/- 0.3; n = 31 versus 17.4 +/- 0.2 mm; n = 40; P 0.004). Interestingly, E(2) concentrations were not correlated with diameter of the pre-ovulatory follicle (r = -0.17; P = 0.15) but milk production was correlated with both E(2) concentrations (r = -0.57; P < 0.0001) and diameter of the pre-ovulatory follicle (r = 0.45; P < 0.0001). Thus, high milk production decreases duration of estrus probably due to decreased circulating concentrations of E(2).

Fertility of holstein dairy heifers after synchronization of ovulation and timed AI or AI after removed tail chalk.

Nonlactating Holstein dairy heifers (n=352) 13 mo of age were managed using a 42-d artificial insemination (AI) breeding period in which they received AI after removed tail chalk evaluated once daily. At AI breeding period onset (d 0), heifers were randomly assigned to receive synchronization of ovulation (100 microg of GnRH, d 0; 25 mg of PGF2alpha, d 6; 100 microg of GnRH, d 8) and timed AI (TAI; d 8) and AI after removed tail chalk for the entire AI breeding period (GPG; n=175), or AI after removed tail chalk for the entire AI breeding period (TC; n=177). As expected, 17.7% (31/175) of GPG heifers received AI after removed tail chalk before scheduled TAI. Pregnancy rate per artificial insemination (PR/AI) at approximately 30 d after first AI tended to be greater for TC (46.5%) than for GPG (38.3%) heifers. No treatment x inseminator interaction was detected; however, overall PR/AI was low for heifers in both treatments due to variation among the 3 inseminators (24.8, 30.0, and 58.0%). Pregnancy loss from approximately 30 to approximately 75 d after first AI was 10% and did not differ between treatments. Based on survival analysis, days to first AI was greater for TC than for GPG heifers, whereas days to pregnancy across the 42-d AI breeding period did not differ between treatments. Overall, 81.2% of GPG heifers receiving TAI synchronized luteal regression and ovulated within 48 h after the second GnRH injection. We conclude that this synchronization protocol can yield acceptable fertility in dairy heifers if AI to estrus is conducted between treatment with GnRH and PGF2alpha and AI efficiency is optimized.

Effect of dietary phosphorus on performance of lactating dairy cows: milk production and cow health.

The objective of this study was to measure cow response to feeding of two dietary concentrations of P, one of which was close to recent National Research Council requirements, and the other of which was well in excess of the requirement. Diets containing 0.37 or 0.57% P (dry basis) were fed to Holstein cows for the first 165 d of lactation, and occasionally longer until cows were confirmed pregnant approximately 60 d after insemination. At calving, cows were randomly assigned to experimental diets. The number of cows completing a minimum of 165 d of lactation was 123 for the 0.37 and 124 for the 0.57% P groups. Cows were housed in a stanchion barn and fed one of two transition diets, each formulated to contain one of the P treatments for the first 3 wk of lactation, and then cows were moved to a free-stall barn where the experimental diets were group fed. Milk production, milk fat, and milk protein averaged 35.1 kg/d, 3.92%, and 2.90% for the 0.37% P diet, and 34.9 kg/d, 3.98%, and 2.91% for the 0.57% P diet. None of these measures were different between treatments. Blood serum P concentrations on d 50 and 100 of lactation averaged 6.1 and 6.2 mg/dL for the 0.37% P diet, and 6.8 and 6.9 mg/dL for the 0.57% P diet. No treatment differences were detected in milk production, cow health, or body condition score.

Reproductive performance of dairy cows fed two concentrations of phosphorus.

The objective of this study was to determine the effect of dietary P concentrations of 0.37 (recommended) or 0.57% (excess; dry matter basis) on reproductive performance. At calving, Holstein cows were randomly assigned to 1 of 2 dietary treatments (n = 134 for 0.37% P and n = 133 for 0.57% P). Cows were fitted with a radiotelemetric transmitter (50 d in milk [DIM]) to record mounting activity during estrus and bred to natural estrus from 50 to 100 DIM, then to synchronized estrus (Ovsynch protocol) after 100 DIM. Weekly ultrasonography was performed from 50 DIM until pregnancy was diagnosed (-30 d after artificial insemination). Pregnancy was confirmed approximately 60 d after artificial insemination (artificial insemination). Weekly blood samples were analyzed for progesterone concentrations. Days to first increase (>1 ng/ml) in progesterone, days to first estrus detected by radiotelemetry, days to first service detected by herd personnel, and conception rate at first service did not differ between the recommended and excess P groups, respectively. Similarly, conception rate at 30 d, days open, pregnancies lost from 30 to 60 d, multiple ovulation rate, and the incidence of anovulatory condition at 71 DIM did not differ between these groups. The mean duration of estrus was 8.7 +/- 0.5 and 8.7 +/- 0.7 h, and the average number of mounts per estrus was 7.4 +/- 0.5 and 7.8 +/- 0.5 for a total mounting time during estrus of 25.8 +/- 1.8 and 24.5 +/- 1.6 s for cows fed the recommended and excess P diet, respectively. Phosphorus treatment had no detectable effect on reproductive performance.

Estrogen concentrations in milk at estrus and ovulation in dairy cows.

The objective of this study was to determine the efficacy of using milk estradiol-17beta (E(2)) sampled during regular milking periods as a predictor of estrus in dairy cows. Twenty-three primiparous Holstein cows received radiotelemetric transmitters on day 16 of the estrous cycle (day 0 = estrus) for continuous monitoring of behavioral estrus. Milk and blood samples were collected every 12h at each milking, from day 18 of the estrous cycle until the fourth milking after the onset of estrus, for radioimmunoassay of E(2). Onset of estrus was indicated by the first standing event identified by radiotelemetry. Ultrasound examination of ovaries was conducted daily in a subset of cows (n = 17) from day 18 until ovulation was confirmed. Statistical analysis involved utilization of Pearson correlation to observe any association of mean plasma and milk E(2) concentrations. Intervals from highest measured plasma and highest measured milk E(2) until the first expression of standing behavior and intervals from highest measured plasma and highest measured milk E(2) until ovulation were compared using Student's t-test. Repeated measures were utilized to evaluate the effect of day and time on milk E(2) concentration. Chi-square procedures were utilized to detect differences in actual time of onset of estrus compared to the expectation of random time of onset of estrus throughout the 24 h day. Mean duration from highest measured milk E(2) until onset of standing behavior was 21+/-3.7h and until ovulation was 46.7+/-5.3h. Mean duration from onset of standing behavior until ovulation was 26.4+/-4.2h. Mean milk E(2) concentrations increased (P<0.01) from the fourth milking period before estrus to the milking period immediately before the first expression of standing behavior, followed by a reduction (P<0.01) in mean concentrations at the milking immediately after the onset of estrus.

Comparative in vitro activity of gemifloxacin against gram-positive and gram-negative clinical isolates in Argentina.

The in vitro activity of gemifloxacin against 1,000 clinical isolates of 147 Streptococcus pneumoniae (115, penicilin susceptible; 26, intermediate penicillin-resistant and 6, penicillin-resistant), 127 Hemophilus influenzae (109, beta lactamasa non-producer; 18, beta lactamase producers), 95 Streptococcus pyogenes (6, azytromycin-resistant), 84 Moraxella catarrhalis (79, beta lactamase producers), 110 Staphilococcus aureus (89, methicillin-susceptible; 21, methicilin-resistant), 98 Eenterococcus faecalis and 339 Enterobacteriacea, (recovered from patients with respiratory tract infection; skin and soft tissue infection and urinary tract infection), was compared with the activities of four fluorquinolones and five other antimicrobial agents. Of the quinolones tested, gemifloxacin was the most potent against Streptococcus pneumoniae, including penicillin intermediate and resistant strains. Mic(90) values obtained for gemifloxacin, ciprofloxacin, ofloxacin, levofloxacin and trvafloxacin were 0.03, 2, 2, 1 and 0.25 mg/L respectively. Gemifloxacin was 16 fold more potent than ciprofloxacin against methicillin-susceptible Staphylococcus aureus and 32 fold more potent than ciprofloxacin against Streptococcus pyogenes. When tested against Hemophilus influenzae, Moraxella catarrhalis and Enterobacteriaceae, all the quinolones showed similar activity. Our results demonstrate that gemifloxacin has similar activity than the other quinolones tested against Gram-negative organisms and is considerably more potent against Gram-positive organisms.

Multicentre study of the in vitro evaluation of moxifloxacin and other quinolones against community acquired respiratory pathogens.

The in vitro activity of moxifloxacin was compared with that of ciprofloxacin, levofloxacin, ofloxacin and trovafloxacin against 710 strains (180 Streptococcus pneumoniae, 180 Haemophilus influenzae, 160 Moraxella catarrhalis and 190 Streptococcus pyogenes) isolated from patients with community-acquired respiratory tract infections. MIC values for moxifloxacin, trovafloxacin were 0.25/0.25, 0.03/0.03, 0.06/0.03 and 0.125/0.0125 mg/l for S. pneumoniae, H. influenzae, M. catharralis and S. pyogenes. Based upon the MIC(90) values and the MIC distributions, moxifloxacin and trovafloxacin were the most active of the quinolones tested. They showed enhanced activity against Gram-positive organisms including penicillin non susceptible S. pneumoniae strains. Moxifloxacin was also highly active against ciprofloxacin-resistant S. pneumoniae strains.

Activity of gatifloxacin compared to those of seven agents against bacteria recovered from outpatients with respiratory tract infection.

The in vitro activity of gatifloxacin and levofloxacin, ciprofloxacin, penicillin, ampicillin, ampicillin-sulbactam, ceftriaxone and clarithromycin was evaluated against 173 S. pneumoniae strains (128, penicillin-susceptible strains; 32, intermediate penicillin- resistant strains and 13, penicillin-resistant strains), 163 H. influenzae strains (128, beta-lactamase non-producer; 35, beta-lactamase producers), 111 M. catarrhalis (9, beta-lactamase non-producer; 102, beta-lactamase producers), 95 Streptococcus pyogenes and 116 S. aureus strains (96, methicillin-susceptible; 20, methicillin-resistant) recovered from outpatients with respiratory tract infection. Based upon the MICs at which 50% and 90% of the isolates were inhibited we concluded that gatifloxacin proved to be the most active antibiotic against respiratory pathogens, including all the penicillin-resistant pneumococci and H. influenzae or M. catarrhalis producing beta-lactamase. Furthermore, their MICs against S. pneumoniae and methicillin-resistant S. aureus were lower than those of levofloxacin and ciprofloxacin.Therefore, this new fluoroquinolone displayed in vitro features that make it suitable for treating community-acquired respiratory tract infections.

Application of chemometric techniques in obtaining macerates with phenolic compound content similar to that of wines from the Jerez-Shery region subjected to oxidative aging.

This paper studies and quantifies the final concentration of phenolic aldehydes and acids (determined by HPLC) in a series of sobretablas wine macerates prepared with American oak shavings subjected to an accelerated aging system developed by our research group and based on thermal processes traditionally used in cooperage. This experiment aims to considerably reduce and control the oxidative aging period of oloroso wines from the Jerez-Sherry region as occurs in the dynamic system of soleras and criaderas, with the consequent economic benefits. To standardize the process by controlling the production technique of the macerates, the results were subjected to surface response methodology as a means of optimizing the experiment. The proposed model was found to be suitable after evaluation of the factors affecting the final concentration. Of the factors studied, it was found to be essential to control the charring time and/or the interactions between temperature and charring time for 10 of the 11 phenolic compounds studied.

Ultrasound interactions with free silicone in a tissue-mimicking phantom.

This study attempts to explain the physical basis for the sonographic appearance of different stages of free silicone in breast tissue by laboratory simulations. A tissue-mimicking breast phantom was constructed, into which silicone inclusions were introduced, to simulate various forms of silicone leakage within the body. These simulations suggest that silicone leakage into surrounding body tissues can cause three primary physical interactions with an ultrasound beam: (1) distortions due to changes in speed of sound; (2) refraction, causing a "lens" effect; and (3) multiple scattering, producing the "snowstorm" effect described previously as a signature pattern for detection of silicone migration in breast tissue.

Characterization of renal ischemia-reperfusion injury in rats.

The purpose of this study was to characterize the time course of renal ischemia-reperfusion injury in the rat. Male Sprague-Dawley rats were subjected to bilateral renal clamping for 45 min. At reestablishment of blood flow, the rats were divided into nine groups (representing 0, 0.5, 1, 2, 4, 6, 9, and 24 h, and 1 week post-ischemia). At each time point, blood samples were taken for analysis of blood urea nitrogen (BUN) and creatinine, and both kidneys were harvested for histopathology and myeloperoxidase activity (MPO) assays. An intracellular adhesion molecular (ICAM-1) monoclonal antibody (IMAb) was tested in a separate group of animals (1 mg/rat) to confirm that it may provide renal protection previously reported by Kelly et al. (1994). Following renal ischemia, significant increases in serum BUN and creatinine were observed compared to levels in normal animals. Serum BUN and creatinine increased 2, 4, and 6 h post-ischemia leading to peak elevations 24 h post-ischemia. Values returned to normal at the 1 week time point. MPO activity was slightly increased 2 and 4 h following ischemia, with peak elevations occurring at the 6-h and 9-h time points. Histopathologic examination of kidneys revealed that the most severe damage occurred at the 24-h time point, which correlated with the peak elevations in serum BUN and creatinine. Evidence of renal injury was still evident histologically 1 week following ischemia, although renal function tests (BUN and creatinine) had returned to normal. In summary, renal injury following ischemia may be demonstrated as early as 4 h post-ischemia as judged by changes in renal function, MPO levels, and renal histopathology. However, based upon renal function tests and histology, peak injury is observed approximately 24 h following ischemia. The ICAM-1 monoclonal antibody, ICAM-Ab, provided some renal protection against ischemia-reperfusion injury in this study as measured by serum creatinine, BUN and renal histopathology. However, in contrast to the results reported by Kelly et al., the magnitude of the protective effects was not as dramatic in the present study, and furthermore, no reductions in renal MPO activity were observed.

Effect of compressive straining on corrosion resistance of a shape memory Ni-Ti alloy in Ringer's solution.

The effect of various degrees of deformation was investigated at specific locations in the stress-strain curve under compression on the corrosion resistance of a wrought Ni-Ti alloy with a martensite to austenite transformation peak of 110 degrees C. Two metallurgical conditions were evaluated: 30% cold drawn and annealed at 900 degrees C for 1 h. The cold drawn material was tested for corrosion resistance after 5.8, 7.4, 12.2, and 24.5% applied strain. Similarly, the corrosion resistance of the annealed material condition was examined after deformation in compression to 11.9, 22.3, and 24.4% strain. Tafel extrapolation and cyclic polarization tests were used for corrosion characterization of each alloy condition. It was found that the corrosion current density undergoes a significant reduction while the breakdown potential improves at increasing strains. In particular, the alloy in the annealed condition exhibited breakdown potentials above 1000 mV with current densities lower than 10 microA cm-2 when it was strained to 24.4%.

Effects of protein deficiency and diet consistency on the parotid gland and parotid saliva of rats.

Protein deficiency results in an increased susceptibility to dental caries, suggesting that oral host-defense properties are compromised. An important component of oral host defense is salivary gland function, which is affected by both protein deficiency and diet consistency. This study describes the effects on rat parotid gland growth and secretory function induced by feeding rats diets of normal (20%) or moderately low (7%) protein content, provided in either a powdered or solid form. In addition, since protein deficiency may result in a secondary zinc deficiency which, in turn, may affect salivary gland function, the effects of these diets on liver zinc concentration were also measured. From 22 to 47 days of age, rats (18/group) were fed the following diets: normal protein, powdered; normal protein, solid; low protein, powdered; and low protein, solid. With each diet consistency, liver zinc was higher for the normal protein group. Within each protein level, liver zinc was higher for the solid diet. This latter observation suggests that food mastication and the resultant stimulation of salivary gland function may also play a role in zinc metabolism. With the normal-protein diet, parotid gland weight was higher for the solid diet; with the low-protein diet, parotid gland weight was similar for both consistencies and did not differ from that of the group fed the "normal protein, powdered" diet. For both consistencies, parotid saliva protein concentration was greater for malnourished rats.(ABSTRACT TRUNCATED AT 250 WORDS)
