Comparative study on renal accumulation of aminoglycoside antibiotics.

Antibiotic activities in the subcellular fractions of rat kidney were determined after a single intraperitoneal injection of streptomycin, gentamicin and amikacin in the doses of 100 mg/kg of body weight. The concentration in the sediment fraction, which consisted mainly of lysosome and mitochondria, is a valuable index for renal accumulation and subsequent toxicity. Half-lives of the aminoglycosides in this fraction were in the order of amikacin (2.34 h), streptomycin (5.86 h), and gentamicin (9.26 h).

The localization of antibiotics by continuous sucrose gradient density.

Rat kidney was homogenized and centrifuged by continuous sucrose gradient density. Aminoglycosides and beta-lactam antibiotics were localized in the lysosomes, whereas the localization of tetracyclines was in the mitochondria. This study demonstrated a technique for the localization of antibiotics in the subcellular fractions without the use of a radioactive tracer.

Reversibility of antibiotic incorporation in large organelles of kidney cell.

Large organelles, namely mitochondria and lysosomes, were fractionated from rat kidney. Antibiotics were added to the fraction and recombinated with no antibiotic-containing supernatant fraction. Tetracycline was quickly dissolved from the large organelle fraction. Cephaloridine had more affinity to organelles than tetracycline. Binding of amikacin was strongest and partly irreversible when added in vivo.

A pharmacokinetic study of cefoperazone.

A pharmacokinetic study of cefoperazone on rats showed long half-life in serum and kidney. The most remarkable finding was that the drug was not detected in the fraction of mitochondria and lysosomes in all the experiment animals. The finding suggests the low nephrotoxic characteristics of cefoperazone.

[Cefotaxime concentration in the prostatic tissue].

Concentrations of cefotaxime (CTX) in serum and prostatic tissue were studied in human and rat. CTX kept high level of 40 to 100 micrograms/g in prostatic tissue at least as long as 2 hours following intraperitoneal injection in a dosis of 100 mg/kg of body weight in rat. Patients intravenously injected of 1 g of CTX showed the activity of 6.8 +/- 4.1 micrograms/g in prostatic tissue.

[Cefotaxime concentrations in the blood and kidney].

Serum and renal levels of cefotaxime (CTX) were studied. Biological half-life time values in serum of rat following 100 mg/kg intraperitoneal injection were 27.58 minutes by bioassay and 25.02 minutes by high performance liquid chromatography, and that in kidney was 27.79 minutes by bioassay. CTX attained high levels in kidney of both human and rat. The concentration was higher in the cortex than that in the medulla. The activity could not be detected in the fraction of brush border, mitochondria and lysosome. This finding was thought to be a favorable characteristic in conjunction with the gradual decrease in the kidney tissue parallel to serum level.

Hydronephrosis in ACI/N rats.

Of 45 ACI/N rats 9 were found with renal agenesis and 2 with hydronephrosis. The hydronephrosis may have been due to faulty development of the mesonephric duct.

Klinefelter's syndrome and bladder cancer.

A patient with Klinefelter's syndrome is described who also had transitional cell carcinoma of the bladder. His mother and maternal grandfather died of neoplasms. It is suggested that neoplasm and aneuploidy in the same family could have been caused by an inherited chromosomal instability rather than coincidence.
