Triploid origin of the gibel carp as revealed by 5S rDNA localization and chromosome painting.

5S ribosomal DNA (rDNA) was isolated and sequenced from the gibel carp Carassius auratus gibelio with 162 chromosomes and crucian carp Carassius auratus with 100 chromosomes, and fluorescent probes for chromosome localization were prepared to ascertain the ploidy origin and evolutionary relationship between the two species. Using fluorescence in-situ hybridization (FISH), major 5S rDNA signals were localized to the short arms of three subtelocentric chromosomes in the gibel carp and to the short arms of two subtelocentrics in the crucian carp. In addition, some minor signals were detected on other chromosomes of both species. Simultaneously, six chromosomes were microdissected from the gibel carp metaphase spreads using glass needles, and the isolated chromosomes were amplified in vitro by degenerate oligonucleotide primed-polymerase chain reaction (DOP-PCR). Significantly, when the DOP-PCR-generated probes prepared from each single chromosome were hybridized, three same-sized chromosomes were painted in each gibel carp metaphase, whereas only two painted chromosomes were observed in each crucian carp metaphase spread. The data indicate that gibel carp is of triploid origin in comparison with diploid crucian carp.

Averaging method of granular materials.

This paper presents an averaging method to link discrete to continuum variables of granular materials. Compared to the other methods proposed in the literature, it has advantages of being applicable to all flow regimes, and to granular flows with or without the effect of physical boundaries. Its application is demonstrated in the determination of the macroscopic properties such as mass density, velocity, stress, and couple stress distributions of a hopper flow, where the discrete results are generated by means of discrete particle simulation. While highlighting the need for considering properly the effect of physical boundaries, the results indicate that the proposed method is an effective way to determine the flow properties of granular materials.

[Studies on the susceptibility of Anopheles anthropophagus to experimental infection with Wuchereria bancrofti].

This paper reports the susceptibility of Anopheles anthropophagus (Xu et Feng, 1975) to experimental infection with Wuchereria bancrofti (Cobbold, 1877), comparing with that of An. sinensis Wiedemann, 1828, Culex pipiens quinque fasciatus Say, 1823, and Aedes togoi (Theobald, 1907). Of 188 An. anthropophagus, 202 An. sinensis, 280 Cx. pipiens quinque fasciatus and 129 Ae. togoi infected by the blood of a bancroftian microfilaremia with mff density of 190 mff/20cmm, the infective rates were 35.64%, 9.41%, 30.00% and 65.89%, respectively; while of 188 An. lesteri anthropophagus, 134 An. sinensis, 289 Cx, pipiens quinque fasciatus and 176 Ae. togoi infected by the blood of another bancroftian microfilaremia with mff density of 83 mff/20 cmm, the infective rates were 18.09%, 3.73%, 13.84% and 39.77%, respectively. It is concluded that the susceptibility of An. anthropophagus to experimental infection with W. bancrofti is significantly higher than that of An. sinensis, significantly lower than that of Ae. togoi and at the same level with that of Cx. pipiens quinquefasciatus (Fig. 1-7).
