Structural investigation of the capsular polysaccharide of Escherichia coli O101 : K103 : H- using bacteriophage degradation and NMR spectroscopy.

NMR spectroscopy was performed on the depyruvated capsular antigen of E. coli K103 and on the oligosaccharide obtained by depolymerisation of the native polysaccharide with a viral-borne endoglycanase. This capsular polysaccharide is the only one to be co-expressed with O group 101 and joins a small group of unusual capsular polysaccharides which possess pyruvic acid as the only acidic function. Structural elucidation of the capsular polysaccharide produced by Escherichia coli O20 : K84 : H26.

The primary structure of the acidic capsular antigen produced by E. coli K84 was shown by glycose and methylation analysis, and by 1D and 2D 1H and 13C NMR studies of the polysaccharide and an oligosaccharide produced by lithium-ethylenediamine degradation of the polysaccharide, to be comprised of linear hexasaccharide repeating units of the following structure: -->4)alpha-D-GaI pA-(1-->3)-alpha-D-Man p-(1-->4)-beta-D-GIc p-(1-->3)-alpha- D-Glc pNAc-(1-->2)-beta-D-Man p-(1-->3)-beta-D-Man pNAc-(1-->.

Structure of the capsular antigen of Escherichia coli O8:K50:H-.

The primary structure of the acidic capsular antigen of Escherichia coli O8:K50:H- was shown by glycose analysis, methylation analysis, and one- and two-dimensional 1H and 13C NMR spectroscopy to be composed of repeating linear tetrasaccharide units having the structure: (formula: see text).

Escherichia coli K48 capsular polysaccharide: a glycan containing a novel diacetamido sugar.

The structure of the exocellular capsular polysaccharide expressed by Escherichia coli O8:K48:H9 has been investigated by hydrolysis and methylation analysis, and by NMR spectroscopic studies of the polysaccharide and of the oligosaccharides generated by solvolysis with anhydrous HF. The capsular polysaccharide was shown to have the repeating unit: [formula: see text] where beta-L-Sug p represents 2,3-diacetamido-2,3,6-trideoxy-beta-L- mannopyranose.

Escherichia coli serotype K45 capsular antigen: a glycan containing 3-acetamido-3,6-dideoxygalactopyranose.

The primary structure of the acidic capsular antigen of Escherichia coli O8:K45:H9 was shown by glycose analysis, methylation analysis, and one- and two-dimensional 1H and 13C NMR spectroscopy to be composed of repeating branched tetrasaccharide units having the structure: [formula: see text] The polysaccharide contains the uncommon amino sugar 3-acetamido-3,6-dideoxy-D- galactopyranose, which has not been previously identified in a capsular antigen.

Klebsiella K43 capsular polysaccharide: primary structure and depolymerisation by a viral-borne endoglycanase.

The capsular polysaccharide of Klebsiella K43 has been studied by glycose analysis, methylation analysis, and NMR spectroscopy, and by bacteriophage depolymerisation of the native polysaccharide. Additional evidence for the structure of the repeating unit came from base-catalysed degradation of the methylated polysaccharide, and from NMR spectroscopic analysis of the lithium-degraded polysaccharide and of the oligosaccharide-alditol derived from the repeating unit oligosaccharide obtained from a bacteriophage degradation. The structure of the capsular antigen of E. coli O8:K102:H- was investigated by methylation analysis, beta-elimination of the methylated polysaccharide, lithium-ethylenediamine mediated degradation, and by 1D and 2D 1H and 13C NMR spectroscopy of the lithium-degraded and native polysaccharides. The annexed structure of the capsular polysaccharide of Escherichia coli O9:K35:H- (A104a) has been determined by glycose and methylation analysis, base-catalysed degradation, and 1D and 2D NMR spectroscopy on the polysaccharide and the products obtained by bacteriophage-mediated depolymerisation. Determination of the structure of the capsular antigen of Escherichia coli O8:K46:H30, using FABMS and 2D-NMR spectroscopy.

The structure of the capsular antigen from Escherichia coli O8:K46:H30 was elucidated by methylation analysis and 1D and 2D 1H- and 13C-NMR spectroscopy, and by methylation analysis, 1D- and 2D-NMR spectroscopy, and FABMS of the oligosaccharide-alditol obtained after dephosphorylation of the polymer with aqueous hydrofluoric acid. The capsular polymer is of the teichoic acid type and has the following repeating unit. [Formula: see text]

Re-investigation of the structure of the capsular polysaccharide of Klebsiella K15 using bacteriophage degradation and inverse-detected NMR experiments.

The structure of the capsular polysaccharide from Klebsiella K15 has been re-investigated, principally by 1D and 2D 1H- and 13C-NMR spectroscopy of the oligosaccharide-alditol obtained by depolymerisation of the polysaccharide with a viral-borne endoglycanase followed by borohydride reduction of the isolated repeating oligosaccharide. The capsular polysaccharide was shown to have the repeating unit: [Formula: see text]

Structure of the glycocalyx polysaccharide of Pseudomonas fragi ATCC 4973.

The structure of the exocellular glycocalyx polysaccharide of Pseudomonas fragi ATCC 4973, a bacterium implicated in the spoilage of meat, has been determined using hydrolysis, methylation analysis and 1D- and 2D-n.m.r. spectroscopy. The polysaccharide, which aids in the adhesion of the cells to each other and to the meat tissue, has the regular repeating unit ----4)-3-O-[(R)-1-carboxyethyl]-alpha-D-Glcp-(1----3)-beta-D-ManpNAc+ ++- (1----4)-beta-D-Glcp-(1----. Random partial O-acetylation occurred in some preparations of the polysaccharide.
