Luminographic detection of von Willebrand factor multimers in agarose gels and on nitrocellulose membranes.

Two methods for visualization of vWf multimers were compared with respect to sensitivity and detection of normal vWf and vWd variants IIA, IIB, IIC, IID, IIE, and IIF. Autoradiography and luminography after electrotransfer of vWf multimers onto nitrocellulose showed comparable sensitivity with vWf:Ag detectable after 1:500 dilution of normal plasma. The least sensitive method was luminography in agarose gels with vWf:Ag detectable after 1:300 dilution of normal plasma. No difference existed in the banding patterns of plasmas from patients with variant vWd.

Autoradiography↗

Luminography--an alternative assay for detection of von Willebrand factor multimers.

A luminescence assay was adapted for detection of von Willebrand factor multimers subsequent to SDS-agarose gel electrophoresis and electroblotting onto nitrocellulose. The method is as fast as chromogenic detection methods and appears to be as sensitive as autoradiography without the disadvantages of the latter.

Influence of factor substitution on the B-cell response in hemophiliacs.

Studies in hemophiliacs receiving factor concentrates demonstrated T-cell defects in vitro. Recently, B-cell dysfunctions were described in AIDS and pre-AIDS and in some hemophiliacs. To investigate the B-cell function in hemophiliacs in relation to factor substitution, we examined five patients with mild (substitution less than 20,000 U/year) and seven with severe (greater than 100,000 U/year) hemophilia A and compared the data with normal control individuals. The B-cell proliferative response (3H-thymidine uptake) to Staphylococcus aureus Cowan I and the differentiation response (Ig secretion into culture supernatants) to T-cell-dependent or -independent polyclonal B-cell activators (PBAs) were studied in vitro. In contrast to T-cell dysfunctions, which correlate with the amount of clotting factor concentrates, the B-cell proliferative response was not affected. Stimulation with PBAs however failed to increase elevated spontaneous IgG levels and showed a diminished increase in IgM levels in severe, but not in mild, hemophilia. Our data give evidence of a T-cell-independent B-cell dysfunction in asymptomatic hemophiliacs that correlates with factor substitution.

Abnormal B cell response to T cell-independent polyclonal B cell activators in haemophilia A.

In addition to T cell abnormalities, patients with haemophilia A show a separate B cell dysfunction. Characteristic are elevated spontaneous IgG (not IgM) levels in the patient's sera and in the culture supernatants of peripheral blood mononuclear cells. It is also observed that these cells fail to show a differentiation response to T cell-independent B cell activators. The B cell dysfunction correlates with the amount of factor VIII concentrates given prophylactically to patients with severe haemophilia. In contrast to the acquired immune deficiency syndrome (AIDS), the proliferation response to B cell mitogens is not affected.

Spatial dissociation of early and late colour evoked components.

Evoked potentials to the primary colours red, green, yellow and blue were recorded and compared with those evoked by white. The unpatterned 10 degrees X13 degrees stimuli were generated with the aid of a colour monitor. Activity was depicted with 5 electrodes, the central electrode 5 cm above the inion and two on each side 5 and 10 cm apart from the central electrode. With equally bright colour stimuli a previously described early negative colour-dominated component N87 was localized in all subjects at the central occipital electrode while the following positivity P100 was clearly more lateralized to the peripheral electrodes. With half-field stimulation N87 showed a similar--paradoxical--lateralization to the ipsilateral electrodes as has been demonstrated for pattern reversal. The existence and localization of N87 could be confirmed also for blue colours, its amplitude independent of the blue luminance, its latency decreasing for definite additional brightness increments and decrements. The N87 to blue was of the same latency as the N87 components to other colours. N87 is mainly generated foveally and parafoveally, its amplitude saturates with stimuli larger than 6-8 degrees in diameter.
