Purification and properties of human cytosolic folylpoly-gamma-glutamate synthetase and organization, localization, and differential splicing of its gene.

Human cytosolic folylpolyglutamate synthetase (FPGS) was expressed in Escherichia coli and purified to homogeneity. Tetrahydrofolate and dihydrofolate were the most effective substrates, while 5-substituted folates were poor substrates. Most pteroyldiglutamates were better substrates than monoglutamates. The human FPGS gene spans 12 kilobases and contains 15 exons and 14 introns. A single FPGS gene was located to chromosome region 9q34.1. Four exon 1 variants were identified, each of which was spliced to exon 2. The exon 1 variant corresponding to the isolated cDNA contains two ATG codons and multiple transcription start sites in this region generates mitochondrial and cytosolic FPGS (Freemantle, S. J., Taylor, S. M., Krystal, G., and Moran, R. G. (1995) J. Biol. Chem. 270, 9579-9584). Exons 1B and 1C, generated by alternate splicing in intron 1, and exon 1A, which is 5' to exon 1 and may encode an additional mitochondrial isoform, are preceded by a number of potential promoter sites. Chinese hamster ovary cell transfectants expressing FPGS activity in the mitochondria contained normal mitochondrial and low cytosolic folylpolyglutamate pools. Mitochondrial FPGS activity is required for mitochondrial folate accumulation, while cytosolic FPGS activity is needed for establishment of normal cytosolic folate pools. The reconstructed FPGS gene restored normal cytosolic and mitochondrial folate metabolism in hamster cells.

Comparative study of skeletal response to estrogen depletion at red and yellow marrow sites in rats.

BACKGROUND: Skeletal sites with red and yellow marrow may respond differently to estrogen depletion. To investigate this possibility, bone histomorphometric changes were compared at skeletal sites with different marrow composition in ovariectomized (OVX) rats, an animal model for postmenopausal bone loss. METHODS: Female rats were OVX or sham-operated at 3 months of age and sacrificed at 1 month or 1 year postsurgery. Eight rats were sacrificed at the beginning of the study as baseline controls. The proximal tibial metaphysis (PTM) and first lumbar vertebra (LV), bone sites with red marrow, as well as the distal tibial metaphysis (DTM) and fifth caudal vertebra (CV), bone sites with yellow marrow, were processed undecalcified for quantitative bone histomorphometry. RESULTS: In control rats, an age-related decrease in bone turnover was observed at all bone sites. Indices of bone turnover, such as osteoclast surface, mineral apposition rate, and bone formation rate, were lower in the DTM and CV compared to the PTM and LV. At 1 month postovariectomy, most bone resorption and formation indices at all bone sites were increased in OVX rats relative to control rats, but decreased cancellous bone volume was found only in the PTM. At 1 year postovariectomy, OVX rats exhibited severe cancellous osteopenia in the PTM and moderate cancellous osteopenia in the LV. In contrast, cancellous osteopenia did not develop in the DTM and CV even at 1 year postovariectomy, despite the fact that OVX rats still exhibited a trend for increased bone turnover at these yellow marrow sites. CONCLUSIONS: The results indicate that unlike skeletal sites with red marrow, estrogen depletion does not induce cancellous osteopenia at skeletal sites with yellow marrow in rats. Since increased bone turnover occurred at the various skeletal sites in OVX rats regardless of marrow composition, this skeletal alteration does not by itself induce cancellous bone loss. An imbalance between bone resorption and formation appears to be the major factor that determines the magnitude of bone loss at a given skeletal site in OVX rats. Differences in the anatomical characteristics of red and yellow marrow that may form a basis for these findings are discussed.

Trisomies 8 and 20 in desmoid tumors.

A nonrandom occurrence of trisomy 8 and of trisomy 20 in desmoid tumors has been recently reported. The finding of trisomy 8 in nondividing desmoid tumor cells by in situ hybridization prompted us to evaluate, in a similar way, the occurrence of trisomy 20 and the possible occurrence of both trisomies together because their co-existence was cytogenetically observed in a few cases. Double fluorescence in situ hybridization (FISH) with centromeric probes for chromosomes 8 and 20 was performed on 16 single cell suspensions of desmoid tumors. FISH confirmed the occurrence of trisomy 8 or 20 in a single cell suspension of desmoid tumors. Both individual trisomies, and even more their association in the same cells, are rare to extremely rare in solid tumors in general and in mesenchymal tumors in particular, and are only known to occur in infantile fibrosarcoma.

Effect of the deletion of the sigma 32-dependent promoter (P1) of the Escherichia coli topoisomerase I gene on thermotolerance.

Topoisomerase I and DNA gyrase are the major topoisomerase activities responsible for the regulation of DNA supercoiling in the bacterium Escherichia coli. The P1 promoter of topA has previously been shown to be a delta 32-dependent heat-shock promoter. A mutant strain with a deletion of P1 was constructed. This mutant is > 10-fold more sensitive to heat treatment (52 degrees C) than the wild type. After brief treatment at 42 degrees C, wild-type Escherichia coli acquires an enhanced resistance to the effects of a subsequent 52 degrees C treatment. This is not the case for the P1 deletion mutant, which, and under these conditions, is about 100-fold less thermotolerant than the wild type. The presence of a plasmid expressing topoisomerase I restored the heat-survival level of the mutant to that of the wild type. During heat shock, the superhelical density of a plasmid with the heat-inducible rpoD promoter is increased in the P1 deletion mutant. We also note that the pulse-labelling pattern of proteins at 42 C (displayed on SDS-polyacrylamide gels) is different in the mutant, and, most notably, the amounts of DnaK and of GroEL protein are reduced. A model is proposed in order to unify these observations.

[Effect of inhaled nitric oxide on endotoxin induced acute lung injury in rabbit].

OBJECTIVE: In order to observe the effects of inhaling nitric oxide (NO) on acute lung injury (ALI). METHODS: 24 rabbits divided into 4 groups. Six rabbits injured with intravenous E. Coli endotoxin, then followed by treatment of inhaling 80 ppm NO in inspired gas. Before and after the infusion of endotoxin, the mean pulmonary arterial pressure (mPAP), mean systemic arterial pressure (mPSA) and the PaO2 were examined. The venous methemoglobin (MHb) was measured by using spectrophometer colorimitry. The extravasculur lung water was evaluated with rate of dried to wet lung weight at the end of study. RESULTS: The rabbits injured with endotoxin inhaling 80 ppm NO could rapidly reduce the mPAP, increase the PaO2 and without inducing significant change of mPSA, MHb and extravasculur lung water. CONCLUSIONS: Inhalation of 80 ppm NO can selectively cause pulmonary artery dilatation, reduce mPAP, improve pulmonary gas exchange, without producing system vasodilation and toxic effects to the rabbits.

Calcium channel blockade and anti-free-radical actions of panaxatriol saponins in cultured myocardiocytes.

AIM: To identify the calcium channel blockade and anti-free-radical actions of panaxatriol saponins Re, Rf, Rg1, Rg2, Rh1, and oleanolic acid saponin Ro. METHODS: On ventricular myocytes of Wistar rats, single channel activities of B, L, and T type calcium channels were recorded with the cell-attached configuration of patch-clamp technic, and free radical contents were measured with electron spin resenance method. RESULTS: Re, Rg1, Rg2, and Rh1 shortened the open times, prolonged the close times, and reduced the open-state probabilities of B, L, and T type calcium channels; Rf shortened the open time, prolonged the close time and reduced the open-state probability of L type calcium channel; Ro did not influence the activity of calcium channels (60 mumol.L-1). Re, Rg1, Rg2, and Rh1 antagonized the increase of free radical content induced by xanthine 0.42 mmol.L-1-xanthine oxidase 5.3 nmol.L-1; Ro and Rf had no effect (30 mumol.L-1). CONCLUSION: Re, Rg1, Rg2, and Rh1 had both the calcium channel blockade and antifree-radical actions. Rf had blockade action on L type calcium channel.

A comparison of the anabolic effects of parathyroid hormone at skeletal sites with moderate and severe osteopenia in aged ovariectomized rats.

Previous studies have shown that parathyroid hormone (PTH) stimulates bone formation and completely restores lost cancellous bone at skeletal sites with moderate osteopenia in relatively young ovariectomized (OVX) rats. The current study was designed to determine whether PTH has similar bone anabolic effects in aged OVX rats and to compare the bone restorative response to PTH at skeletal sites with moderate and severe osteopenia. Female Sprague-Dawley rats were subjected to sham surgery or bilateral ovariectomy at 3 months of age and maintained untreated for the first year after surgery to allow for the development of moderate vertebral osteopenia and severe tibial osteopenia in OVX rats. Groups of baseline control and OVX rats were sacrificed at the end of this pretreatment period. The remaining OVX rats were then treated for 10 weeks with vehicle, antiresorptive agents alone (estrogen, the bisphosphonate risedronate, or calcitonin) or PTH alone. Other groups of OVX rats were treated concurrently with PTH and each of the antiresorptive agents. As expected, the proximal tibia of baseline OVX rats exhibited severe cancellous osteopenia, whereas the first lumbar vertebral body was moderately osteopenic. Treatment of OVX rats with antiresorptive agents alone failed to restore cancellous bone at both skeletal sites, whereas treatment with PTH alone markedly stimulated bone formation and completely restored lost cancellous bone in the lumbar vertebra. PTH also stimulated bone formation and in the severely osteopenic proximal tibia of OVX rats but only marginally restored lost cancellous bone, possibly due to an inadequate number of bone spicules to serve as a foundation for new bone formation.(ABSTRACT TRUNCATED AT 250 WORDS)

Effects of K+, pH and glutamate on 45Ca kinetics in hippocampal brain slices.

Altered calcium homeostasis is likely to play a pathogenetic role in cerebral ischemia. In order to further understand which factors associated with ischemia contribute to disturbances of calcium metabolism, the influence of 3 isolated insults, 8 mM K+, pH 6.1 and 1 mM glutamate, on total tissue calcium were studied by analysis of steady-state kinetics of 45Ca in 500 microns hippocampal brain slices. 45Ca kinetics were analyzed with 2 bi-exponential models by non-linear least-squares analysis. Tissue wet weight/protein was measured simultaneously. Each experimental condition produced a unique tissue response. Raising K+ had no effect on tissue water but increased the rate of uptake of Ca2+ into the larger, rapidly equilibrating tissue Ca2+ space. Acidosis reduced tissue water and the amount of Ca2+ in the slowly equilibrating compartment due to enhanced efflux from that space. Glutamate increased tissue water in a time-dependent manner and increased the influx and amount of Ca2+ in the slowly equilibrating space. Combined insults revealed minimal interaction between K+ and acidosis or glutamate, but glutamate with acidosis worsened tissue injury. We discuss the relationship of this technique to other methods for studying tissue calcium and the significance of the observations regarding ischemia.

Calcium influx induces neurite growth through a Src-Ras signaling cassette.

We find that calcium influx through voltage-dependent calcium channels causes extensive neurite outgrowth in PC12 cells. The calcium signal transduction pathway promoting neurite outgrowth causes the rapid activation of protein tyrosine kinases, which include Src. Protein tyrosine phosphorylation results in the formation of an Shc/Grb2 complex, leading to Ras activation, MAP kinase activation, and the subsequent induction of the immediate early gene NGFI-A. Protein tyrosine phosphorylation, gene induction, and neurite outgrowth are inhibited by the expression of dominant negative forms of both Src and Ras, indicating a requirement for both proto-oncoproteins in calcium signaling. Our results suggest that a signaling cassette which includes Src and Ras is likely to underlie a broad range of calcium of actions in the nervous system.

Droloxifene inhibits cortical bone turnover associated with estrogen deficiency in rats.

Droloxifene (DRO), an estrogen antagonist/agonist, has been shown to possess estrogen-like effects in inhibiting bone turnover leading to cancellous bone loss in ovariectomized (OVX) rats. The purpose of this study was to determine the effects of DRO on cortical bone turnover in OVX rats. Sprague-Dawley female rats at 5 months of age were sham-operated (sham, n = 8) and orally treated with vehicle, or OVX (n = 56) and orally treated with either vehicle, DRO at 0.1, 1, 5, or 10 mg/kg/day, or 17 alpha-ethynyl estradiol (EE) at 3 or 30 micrograms/kg/day for 4 weeks. Static and dynamic cortical bone histomorphometry was performed on double fluorescent labeled, undecalcified cross sections of tibial diaphyses (proximal to the tibiofibular junction). There were no significant differences in tibial diaphyseal cross sectional area, marrow cavity area, and cortical bone area between groups after 4 weeks of administration. Periosteal mineralizing surface, mineral apposition rate, and bone formation rate-surface referent and endocortical eroded surface increased significantly, while endocortical mineral apposition rate and bone formation rate-surface referent increased nonsignificantly in OVX controls compared to sham controls. Treatment with DRO at doses of 0.1 to 10 mg/kg/day dose-dependently attenuated the OVX-induced higher bone formation indices in both the periosteal and endocortical surfaces and higher bone resorption index in the endocortical surface. At the highest dose (10 mg/kg/day), DRO completely inhibited the increases in bone formation and resorption indices in OVX rats.(ABSTRACT TRUNCATED AT 250 WORDS)

Effects of droloxifene on prevention of cancellous bone loss and bone turnover in the axial skeleton of aged, ovariectomized rats.

The purpose of this study was to determine the efficacy of droloxifene (DRO), an estrogen antagonist/agonist, in preventing ovariectomy (OVX)-induced lumbar vertebral cancellous bone loss and bone turnover in aged female rats. Fifty-three Sprague-Dawley female rats were OVX or sham-operated at 19 months of age, and divided into 6 groups: (I) sham-operated controls; (II) OVX vehicle controls; (III) OVX rats treated with E2 at 30 micrograms/kg/day; (IV)-(VI) OVX rats treated with DRO at either 2.5, 5, or 10 mg/kg p.o. daily. The treatment period was 8 weeks. Static and dynamic cancellous bone histomorphometric parameters were determined on 4 and 10 microns thick, undecalcified, double-fluorescent labeled sections of the fourth lumbar vertebral body. Changes in body weight, uterine weight, and total serum cholesterol were also determined. OVX for 8 weeks in 19-month-old female rats resulted in reduced trabecular bone volume (-18%) and trabecular width (-10%) and increased labeling perimeter (+52%), bone formation rate/bone surface referent (+60%), bone formation rate/bone volume referent (+77%), osteoclast number (+41%), and osteoclast perimeter (+41%). E2 treatment at 30 micrograms/kg/day for 8 weeks prevented OVX-induced cancellous bone loss and decreased bone resorption, bone formation, and bone turnover to the values of sham controls. DRO at 2.5-10 mg/kg/day completely prevented bone loss and bone turnover associated with estrogen deficiency. Osteoclast number and perimeter were significantly decreased in DRO-treated-OVX rats compared to both sham and OVX controls. Trabecular bone volume, trabecular width, labeling perimeter, bone formation rate/bone surface referent, and bone formation rate/bone volume referent showed no differences in DRO-treated OVX rats compared to those of E2-treated OVX rats and sham controls. These histomorphometric results indicated that DRO is an estrogen agonist on cancellous bone of lumbar vertebral bodies of aged, OVX rats. Further, E2 treatment prevented the OVX-induced increase in body weight gain and nonsignificantly reduced total serum cholesterol compared to OVX controls. Body weight gain and total serum cholesterol did not differ between OVX rats treated with E2 and sham controls. In OVX rats treated with DRO, body weight decreased significantly in a dose-response manner, and total serum cholesterol was significantly reduced by 65% to 70% compared to both sham and OVX controls. In addition, treatment with E2 increased uterine weight to the value of sham controls in OVX rats. However, DRO had no effect on uterine weight at either 2.5 or 10 mg/kg/day, while it only slightly but significantly increased uterine weight over OVX controls at 5 mg/kg/day. We conclude that DRO was efficacious in the prevention of lumbar vertebral cancellous bone loss and in the decline of total serum cholesterol but had no effect on uterine weight in the aged, OVX female rats. Our data suggest that DRO is a potentially useful agent for the prevention of vertebral bone loss leading to spinal fractures in postmenopausal women.

[Calcium channel blockade and anti-free-radical actions of panaxadiol saponins Rb1, Rb2, Rb3, Rc, and Rd].

AIM: To identify the calcium channel blockade and anti-free-radical actions of panaxadiol saponins Rb1, Rb2, Rb3, Rc, and Rd. METHODS: On ventricular myocardiocytes of Wistar rats, single channel activities of L, T, and B type Ca2+ channels were recorded with the cell-attached configuration of patch-clamp technic, and free radical contents were measured with electron spin resenance method. RESULTS: Rb1, Rb2, Rb3, and Rc 200 mumol.L-1 shortened the open times, prolonged the close times, and reduced the openstate probabilities of calcium channels and 30 mumol.L-1 antagonized the increase of free radical content induced by xanthine 0.42 mmol.L-1-xanthine oxidase 5.3 nmol.L-1, but Rd in the same dose behaved none of the effects. CONCLUSION: Rb1, Rb2, Rb3, and Rc had both the calcium channel blockade and anti-free-radical actions.

[Study on incidence of acute flaccid paralysis in Hebei].

A study on the incidence of Acute Flaccid Paralysis (AFP) among 0-14 year-old children in 1990-1992 was carried out in all over 12 prefectures and cities level hospitals in Hebei. The county level hospitals were investigated in 3 of 12 prefectures and cities. A total of 214 hospitals were investigated. The result showed that the annual average incidence of AFP and non-polio AFP among 0-14 year-old children in 1990-1992 were 1.498/100000 and 0.988/100000, respectively. The annual average incidence of them among < 5 year-old children were 2.917/100000, 1.591/100000, respectively. The incidence of AFP and non-polio AFP were different in each prefecture and city. GBS accounts for 54.06% of all AFP cases.
