Genetic diversity and divergence in Chinese yak (Bos grunniens) populations inferred from blood protein electrophoresis.

In 6 Chinese yak (Bos. grunniens) populations including 177 yaks, 34 blood protein loci were studied by horizontal starch gel electrophoresis, four of these loci (AKP, ALB, LDH-1, TF) were found to be polymorphic. The percentage of polymorphic loci(P) is 0.118, the mean individual heterozygosity(H) is 0.015, which means a low level of genetic diversity in the whole Chinese yak population. The coefficient of gene differentiation (Gst) is 0.0625, which indicated an almost-indistinguishable divergence among different populations at the level of blood protein electrophoresis.

Evidence that GCN1 and GCN20, translational regulators of GCN4, function on elongating ribosomes in activation of eIF2alpha kinase GCN2.

In the yeast Saccharomyces cerevisiae, phosphorylation of translation initiation factor eIF2 by protein kinase GCN2 leads to increased translation of the transcriptional activator GCN4 in amino acid-starved cells. The GCN1 and GCN20 proteins are components of a protein complex required for the stimulation of GCN2 kinase activity under starvation conditions. GCN20 is a member of the ATP-binding cassette (ABC) family, most of the members of which function as membrane-bound transporters, raising the possibility that the GCN1/GCN20 complex regulates GCN2 indirectly as an amino acid transporter. At odds with this idea, indirect immunofluorescence revealed cytoplasmic localization of GCN1 and no obvious association with plasma or vacuolar membranes. In addition, a fraction of GCN1 and GCN20 cosedimented with polysomes and 80S ribosomes, and the ribosome association of GCN20 was largely dependent on GCN1. The C-terminal 84% of GCN20 containing the ABCs was found to be dispensable for complex formation with GCN1 and for the stimulation of GCN2 kinase function. Because ABCs provide the energy-coupling mechanism for ABC transporters, these results also contradict the idea that GCN20 regulates GCN2 as an amino acid transporter. The N-terminal 15 to 25% of GCN20, which is critically required for its regulatory function, was found to interact with an internal segment of GCN1 similar in sequence to translation elongation factor 3 (EF3). Based on these findings, we propose that GCN1 performs an EF3-related function in facilitating the activation of GCN2 by uncharged tRNA on translating ribosomes. The physical interaction between GCN20 and the EF3-like domain in GCN1 could allow for modulation of GCN1 activity, and the ABC domains in GCN20 may be involved in this regulatory function. A human homolog of GCN1 has been identified, and the portion of this protein most highly conserved with yeast GCN1 has sequence similarity to EF3. Thus, similar mechanisms for the detection of uncharged tRNA on translating ribosomes may operate in yeast and human cells.

Reduced transdermal absorption of N,N-diethyl-m-toluamide from a new topical insect repellent formulation.

Extensive absorption of the topical insect repellent N, N-diethyl-m-toluamide (DEET) causes systemic and local toxicities. This report describes the preparation and characterization of a new insect repellent formulation (FA), a PEG-polyacrylic acid polymer system, for its DEET release, in vitro skin permeation, and in vivo transdermal absorption properties; and for its relative repellency against Aedes aegypti mosquitoes. DEET release and skin permeation were studied in Franz diffusion cells. DEET transdermal absorption and relative repellency of FA were assessed in beagle dogs. A commercial DEET lotion (FB) and technical DEET (FC) were used as references. FA exhibited 19.5% and 61.7% decrease in DEET steady-state skin flux compared with FB and FC, respectively. At 15 mg DEET/kg, the absolute DEET transdermal bioavailability and Cmax were 13.4% and 154.3 ng/ml, respectively, for FA; and were 17.5% and 196.5 ng/ml, respectively, for FB. DEET half-lives (t1/2) for FA (2.52 hr) and FB (2.73 hr) were similar, while MRT for FA (4.99 hr) was significantly greater (p < 0.05) than that for FB (4.38 hr). FA showed lower mosquito biting rates at 1, 2, 3, 4, 5, and 6 hr postdose at 0.5 mg DEET/cm2. FA exhibited reduced in vitro skin permeation and in vivo transdermal absorption of DEET as well as superior repellency compared with FB. The PEG-polyacrylic acid polymer system is of value in the formulation of DEET lotions.

[Clinical study of systemic inflammatory response syndrome and multiple organ dysfunction syndrome in critically patients].

We defined the epidemiology of systemic inflammatory response syndrome (SIRS) and multiple organ dysfunction syndrome (MODS) in critically ill patients, and evaluated the procession from SIRS to MODS and the therapeutic strategies. 230 patients were studied prospectively until discharge or death. On admission, the morbidity rate of SIRS was 71.3%. The mortality rate of the patients with SIRS was 18.9%. MODS was developed in 65 patients (28.3%), and 33 patients dided (50.8%). In SIRS patients with non-infectious SIRS, sepsis and septic shock, the morbidity rates of MODS were 22.8%, 61.1% and 85.7%, and the mortality rates were 11.4%, 30.6% and 50.0% respectively. The outcome of critically ill patients may be improved if SIRS is early diagnosed and the body inflammatory response is reglulated properly.

Engineering factors affecting the granulation of sludge in the UASB reactor.

The influence of inoculum sludge involving different microbial populations, different modes of stream flow and different flow rates in a bioreactor on the granulation of sludge was studied. In the granulation process there were four periods: formation of microbial floccus, formation of subcore, growth of subcore, and maturity of granules. The production of microfloccus could be attributed to acid-forming bacteria. The hydrodynamic momentum transfer caused by moving phases and the hydraulic shear effect are the crucial factors affecting the formation of the subcore. For this a new concept-the lowest limited flow rate-is proposed, i.e., the lowest flow rate to form an expanded sludge bed. A sufficient feed rate (higher than the lowest flow rate), an appropriate sludge load, and an equal distribution of feed, as well as a suitable alkalinity in the medium are the essential factors for the scaling-up and process-control of the UASB reactor.

[Toxoplasmosis infection in pregnant women in Lanzhou].

OBJECTIVE: To investigate toxoplasmosis (TOX) infection in pregnant women. METHODS: The blood samples from 1,250 pregnant women were tested by indirect hemaglutination (IHA) for TOX. 1,327 normal cases were included in control group. RESULTS: The infection rate of TOX was 7.28% and 8.14% in the study and control group respectivity. There was no significant difference between the study group and control group (P > 0.05). There was no marked relationship between various age groups. (P > 0.05). No significant difference was noted in three gestational periods. Most pregnant women with of TOX infection were active and recent. CONCLUSION: Pregnancy did not increase the chance of TOX infection.

[The new parameters of motor unit potential in the diagnosis of neurogenic lesions in spike-triggered averaging electromyography].

OBJECTIVE: To evaluate the clinical significance of the new parameters of motor unit potential (MUP) to the diagnosis of neurogenic lesions. METHODS: Three groups of muscles, anterior tibialis, abductor pollicis brevis and vastus medialis were studied with spike-triggered averaging electromyography (EMG). A total of 3,646 MUPs, 1,903 from normal group and 1,743 from neurogenic group, were recorded under slight muscle contraction. RESULTS: The area, as a single parameter, produced the most significant result in discriminant analysis, about 80% MUPs from neurogenic muscles being judged to be abnormal. Compared with the area, the amplitude and duration were less sensitive, being able to assign about 77% and 70% of MUPs in the neurogenic group as abnormal. A combination of the area/amplitude ratio (thickness) and the amplitude, and a combination of the duration (width) and the amplitude could further improve the discriminating ability, around 80% and 81% MUPs in the neurogenic muscles being classified as abnormal by the combinations. The simplified discriminant functions, 2 x Log10 (amplitude) +Log10 (area/amplitude) -4 for size index, and 5 x Log10 (duration) + 3 x Log10 (amplitude) -10 for width-height index had same discriminating ability as the real discriminant function. CONCLUSIONS: The new parameters, SI, WHI and area in spike-triggered EMG seemed to be promising, since they produced a better yield in the diagnosis of neurogenic lesions than amplitude or duration alone in the discriminant analysis.

Thermodynamics of folding of the RNA pseudoknot of the T4 gene 32 autoregulatory messenger RNA.

Nucleotides U(-67) to C(-40) at the extreme 5' end of the gene 32 mRNA in bacteriophage T4 have been shown to fold into an RNA pseudoknot proposed to be important for translational autoregulation. The thermal denaturation of three in vitro transcribed RNAs corresponding to the pseudoknot region has been investigated as a function of Mg2+ concentration to begin to elucidate the determinants of the structure and stability of this conformation. T4-35 is a 35-nucleotide RNA containing a 5' G followed by the natural T4 sequence starting with the mature 5' end of the mRNA, nucleotides A(-71) to C(-38). A 32-nucleotide RNA, termed T4-32, contains the native sequence form U(-67) to C(40) with 5'GC and 5'CA single-stranded regions appended to the 5' and 3' ends of the core sequence, respectively. T4-28 contains only the 28 core nucleotides, and the predicted closing U(-67)-A(-52) base pair in stem 1 has been replaced with a phylogenetically allowed G(-67)-C(-52) base pair. Ribonuclease mapping of T4-32 and imino proton NMR experiments of T4-35 show that both sequences adopt a pseudoknotted conformation. At pH 6.9 and 50 mM NaCl, T4-35 and T4-32 RNAs are characterized by a single major melting transition over a wide range of [Mg2+] (0-6 mM). The delta H degree of unfolding for T4-35 and T4-32 shows a large dependence on Mg2+ concentration; the maximum delta H degree occurs at about 2.0 mM Mg2+ with further addition of Mg2+ simply increasing the tm. Investigation of the [Mg2+] dependence of the tm suggests that a net of one Mg2+ ion is released upon denaturation of T4-35 and T4-32 RNAs. Over the entire [Mg2+] range, the delta G degree (37 degrees C) for the folding of T4-35 is consistently 1-1.5 kcal mol(-1) more negative than T4-32 due to a higher stabilization enthalpy for the natural sequence molecule. In contrast to this behavior, T4-28 gives consistently higher tm's but less negative enthalpies and is destabilized (at 37 degrees C) by about 0.5-1.5 kcal mol(-1) relative to T4-32 and by about 2-3 kcal mol(-1) relative to T4-35, depending upon cation concentration. (1)H NMR experiments suggest that, even in the presence of 4.0 mM Mg2+, T4-28 RNA does not adopt a stable pseudoknotted conformation. These data show that the stability of the pseudoknot in the gene 32 mRNA encoded by the 28-nucleotide core sequence is significantly influenced by the number and nature of the immediately adjacent "single-stranded" 5' and/or 3' nucleotides appended to the core structure. These findings are discussed within the context of the structural model for the evolutionarily related phage T2 and T6 gene 32 mRNA pseudoknots presented in the following paper [Du, Z., Giedroc, D. P., & Hoffman, D. W. (1996) Biochemistry 35, 4187-4198].

Comparison of adjuvant formulations for cytotoxic T cell induction using synthetic peptides.

We have investigated the capacity of synthetic peptides delivered in different adjuvant formulations to induce cytotoxic T lymphocyte (CTL) responses to a class I H-2Kd-restricted Plasmodium berghei circumsporozoite epitope, CS 252-260. Using three immunogen formulations: soybean emulsion; Montanide ISA720; and lipopeptide (P3-CS), we first evaluated the effects of immunization routes on CTL induction. No CTL response was induced in mice immunized s.c. or i.p. with CS peptide formulated in soybean emulsion. In contrast, immunization with lipopeptide P3-CS either s.c. or i.p. effectively primed for CTL. Interestingly, CS peptide emulsified in Montanide ISA720 induced a CTL response only when delivered s.c. and not i.p., indicating the critical influence of immunization routes on CTL induction. We then compared the effectiveness of eight adjuvant formulations to induce CTL response following a single s.c. immunization. Notably, lipopeptide P3-CS and CS peptide admixed with P3 or POE lipid molecules stimulated a vigorous CTL response. However, only mice immunized with P3-CS and CS peptide admixed with P3 molecule generated long-lived CTL which persisted in vivo for 5 months. Thus, based on a simultaneous comparison of the different adjuvant formulations, we demonstrated that the conjugated and unconjugated P3 lipopeptides were the most effective immunogens for eliciting primary and memory CTL in mice.

Solid-phase extraction and liquid chromatographic quantitation of insect repellent N,N-diethyl-m-toluamide in plasma.

A sensitive and reliable method based on solid-phase extraction and reversed-phase liquid chromatography was developed and validated for the quantitation of insect repellent N,N-diethyl-m-toluamide (DEET) in plasma. N,N-diethyl-2-phenylacetamide was used as internal standard in the extraction which employed C18 solid-phase extraction cartridges. The wash solvent was 3 ml acetonitrile-ammonium acetate (pH 4.5; 0.03 M) (10:90, v/v), and the eluting solvent was 1 ml acetonitrile-ammonium acetate (pH 4.5; 0.03 M) (40:60, v/v). The eluent obtained from the extraction cartridge was directly analyzed on a reversed-phase C8 column with UV detection at 220 nm. A clean chromatogram and high sensitivity were achieved at this wavelength. The mobile phase was acetonitrile-ammonium acetate (pH 4.5; 0.03 M) (36:64 v/v). The retention time was 7.9 min for the internal standard and 9.6 min for DEET when the mobile phase was delivered at 1.0 ml min-1. The overall absolute recovery was 97.7% with a standard deviation (SD) of 3.9 (n = 9) for DEET and 100.2% with a SD of 3.4 (n = 3) for the internal standard. The limit of quantitation was found to be 15 ng ml-1 with a relative standard deviation of 12%. For the analyses of DEET-spiked plasma samples with five replicates each at 50, 500 and 1500 ng ml-1, the overall intra- and inter-day precisions were 5.7% and 5.5% respectively, and the overall intra- and inter-day accuracies were 2.0% and 2.4% respectively. The correlation coefficient for calibration plots in the concentration range 15-1500 ng ml-1 was typically 0.999. The method was applicable to both dog and human plasma samples and was successfully used in pharmacokinetic studies of DEET in beagle dogs after intravenous bolus and topical routes of administration.

Synthetic peptides entrapped in microparticles can elicit cytotoxic T cell activity.

Peptides from Plasmodium berghei circumsporozoite protein (CS) and influenza A virus nucleoprotein (NP) were entrapped in microparticles prepared from poly (lactide-co-glycolide) polymers, and the microparticles were administered parenterally to mice. After immunization with single or multiple doses, splenocytes were tested for a cytotoxic T cell (CTL) response and high levels of CTL activity were detected. The CTL induced were CD8+, MHC class I restricted, and could recognize virus infected cells. Peptide entrapped in microparticles of mean size < 500 nm were better inducers of CTL than larger microparticles (mean > 2 microns and above). Microparticles could also be used to deliver lipid modified peptides (lipopeptides) and elicited higher levels of cytolytic activity than either free peptide in microparticles or lipopeptide alone. Microparticles provide a novel way of inducing a CTL response using synthetic peptides.

[Linkage analysis between milk protein loci and lactation trait loci].

The linkage relationship between milk protein loci and lactation trait loci, such as milk yield, fat percent, protein percent and dry matter percent, were fully investigated by the methods of heterozygosity analysis and linkage group analysis. The results showed that each single protein locus showed a higher homozygosity while the three easein loci showed a lower homozygosity when they were investigated simultaneously. There was no significant regression of deviations in lactation traits on overall marker heterozygosity. Significant linkage between kappa-CN locus and milk fat percent and protein percent, and that between alpha sl-CN locus and 305-d milk yield were found.

[The role of TNF alpha, IL-1 beta and MIP-1 alpha in LPS-induced organ injury].

OBJECTIVE: To investigate the role of inflammatory cytokines in endotoxemia models and to explore the possible anti-cytokine therapy of endotoxemia. METHODS: TNF alpha in plasma was measured by ELISA, and the mRNA of cytokines was assessed by slot blot analysis. RESULTS: LPS-induced TNF alpha release was in a dose:dependent manner in human whole blood. Dexamethasone (> or = 10(-8)mol/L) exhibited inhibitory effect on TNFa release. Ibuprofen 10(-7)-10(-9) mol/L had inhibitory effect on TNFa production, whereas 10(-3)-10(-4)mol/L stimulated TNFa release. The rat model of acute lung injury was made by LPS intraperitoneal injection. Both dexamethasone and ibuprofen that injected at 1 hour before LPS injection could decrease the contents of Evans blue in the lung (t 2.80 and 7.31 respectively, P < 0.05 vs LPS control). After LPS administered, there was a progressive up regulation in transcripts of TNF alpha, IL-1 beta and MIP-1 alpha in whole lung homogenates of rats. The mRNA peaked at 2, 6, 12 hours respectively. The TNF alpha, IL-1 beta and MIP-1 alpha mRNA levels decreased markedly when dexamethasone or ibuprofen given at 1 hour before LPS injection. CONCLUSION: TNF alpha, IL-1 beta and MIP-1 alpha play an essential role in the inflammatory response. LPS-induced acute lung injury may be prevented by dexamethasone and ibuprofen which have inhibitory effect on the gene expression of cytokines in the lung.

[Pedigree investigation of familial non-insulin dependent diabetes mellitus].

UNLABELLED: To discuss the inheritant mode of familial NIDDM. METHODS: According to WHO criteria of DM, 100 NIDDM Probands with family history of DM were diagnosed and 100 persons were chosen at random for controls. The survey of DM was performed in both groups, including FBG, HbAlc, FINS. Some members had insulin release test. RESULTS: The prevalance rates of DM in familial DM group were 26 times of the control group (34.3% and 1.3%). The prevalance rate of DM among first-degree relation was 18 times higher than that in general population (28.3% and 1.5%). The rate of diabetes in the siblings and in the off-springs was 44.4% and 9.7% respectively. The pedigree analyses showed that 83.9% affected families had one diabetic parent, one half siblings had DM, and there was a successive transmission of DM through at least three generations in sixteen large families. Besides, the incidence of DM was much higher in females than in males (40% and 28%). Among affected parents, diabetlic mothers were much more than diabetic fathers (50.8% and 27.6% P < 0.01). CONCLUSION: Familial NIDDM had a familial aggregation. It was inherited in the manner of Mendelian autosomal dominant inheritance. The difference between the rate in DM mothers and fathers was probably due to unequal prevalance rate in females and males.

[Effects of selective nitric oxide synthase inhibitor in sheep with endotoxic shock].

OBJECTIVE: To investigate the effects of S-methylisothiourea sulfate (SMT), a selective inducible nitric oxide synthase (iNOS) inhibitor, on hyperdynamic endotoxic shock sheep. METHODS: Endotoxic shock was induced by Escherichia coli endotoxin in both control (n = 8) and SMT groups (n = 8). SMT was given intravenously. Hemodynamic data, oxygen delivery derived parameters and intramucosal pH (pHi) were measured. RESULTS: The control group had a hyperdynamic state, similar to that of human septic shock. In the SMT group, blood pressure was maintained at baseline, and cardiac index (CI) was lower than that in the control group (P < 0.05). Oxygen extraction ratio (O2 ext) was increased up to 40% +/- 5% and was much higher than that of the control group (P < 0.01). Pulmonary artery pressure (PAP) was higher than that of the control group (P < 0.01), and pHi decreased gradually similarly to the control group. CONCLUSION: SMT restored the blood pressure and increased O2 extespecially in the gut, but decreased CI and oxygen delivery and increased PAP. So over inhibition of iNOS should be cautiously considered.

[Minilaparotomy cholecystectomy with clinical analysis of 50 cases].

Fifty cases of gallbladder disease operated with minilaparotomy cholecystectomy (MC) and fifty cases operated with conventional cholecystectomy (CC) were reviewed and compared in terms of their need of postoperative analgesic and hospitalization days. The results showed that the MC was better than the CC. The merits and demerits of MC. CC and LC were discussed. The operative indexes of MC were presented by author.

[Experimental study on homoiothermic extracorporeal liver preservation].

To prove that perfused liver can be preserved at room temperature (22 degrees C), we made the experiment, in which HTK was basic solution, perfluorocarbon acted as oxygen carrier and lipid acted as energy substrate in the homoiothermy condition, pig liver organs were perfused extracorporeally through the V. portal with perfusion solution. In fixed period of time the perfusion solution from the liver was taken and analysed to determine for liver biochemical function and observe the concentration and size of hepatocellular mitochondria. In oxygen carrier perfusion solution the ammonia concentration was low, urea concentration was high, damage to mitochondria was minimum and by addition of lipid emulsion the concentration of glucose can be maintained. The experimental and control data were obviously significant. The result demonstrated that oxygen carrier (perfluorocarbon) as oxygen supplier can provides enough oxygen to liver cell, at the sametime, lipid emulsion intralipid) provides energy so as the perfused liver can be preserved at high temperature, i.e. 22 degrees C at relative long time (40 hours) and still has the function of removing toxic substance such as ammonia and converting it to urea, meanwhile maintain the normal structure of mitochondria. The preservation of perfused liver at homoiothermy is possible.

[Clinical study of imposed work of breathing caused by the breathing apparatus].

OBJECTIVE: The imposed work of breathing caused by the breathing apparatus (endotracheal tube, breathing circuit tubing, and ventilator's valves) may increase patient's work of breathing. To evaluate the imposed work may be useful to determine weaning. METHODS: Work of breathing was observed by Bicore CP-100 breathing monitor at different levels pressure support (PS), T piece and after extubation, and the PS level to compensate imposed work of breathing was computed. RESULTS: When PS level decreased, patient's work of breathing increased significantly. At PS O kPa, the patient's work of breathing increased 57%, which was caused by imposed work of apparatus. Because of the resistance of endotracheal tube, the patient's work of breathing increased 41% at T piece. The PS level that compensated for the imposed work was 0.7 kPa. CONCLUSIONS: When PS level was too low or T piece was applied, the imposed work of breathing caused by apparatus could markedly increase patient's work of breathing.
