A database designed to computationally aid an experimental approach to alternative splicing.

A unique microarray approach has been developed to profile alternative splicing in the cell. To support the development of this approach, we have developed the Manually Annotated Alternatively Spliced Events (MAASE) database system, which is a unique alternative splicing information resource designed specifically with experimentalists in mind. MAASE is an online resource for the convenient access, identification, and annotation of alternative splicing events (ASEs). MAASE consists of two components: an annotation system and a curated database. The annotation system is a web-based workspace that combines manual and computational approaches to identifying and annotating ASEs, a combination that is vital if a comprehensive collection is to be obtained. The annotation system is publicly available and provides a scalable solution to acquiring as well as contributing to annotated ASEs. MAASE annotated ASEs are deposited into the database component, which can either be queried one entry at a time or multiple entries at a time with convenient access to alternatively spliced junctional and surrounding sequences to facilitate the design of microarray experiments.

[Study of apoptosis and expression of apoptotic related genes P53 bcl-2 C-myc in oral lichen planus].

OBJECTIVE: To investigate the difference of apoptosis and expression of apoptotic related genes p53 bcl-2 C-myc in the development of oral lichen planus(OLP). METHODS: Apoptotic cells were detected by TUNEL and immunohistochemical method was used to study P53 bcl-2 C-myc expression in 45 samples with OLP(23 cases of erosive OLP group and 22 cases of non erosive OLP group). RESULTS: Apoptosis occurred in 100% of the cases and apoptotic index(AI) was 24.3%+/-12.0% (1.9%-50.5%). AI of erosive OLP group was 19.0%+/-10.4% (1.9%-50.5%), and AI of non erosive OLP was 29.4%+/-11.2% (10.4%-49.3%). The obvious differences existed between erosive group and non erosive group (P<0.05). The expression of p53 bcl-2 C-myc positive rates were 31.1% 35.5% 33.3% respectively. The p53 bcl-2 C-myc positive rates in erosive OLP group were 39.1% 52.2% 43.2%and that with non-erosive OLP group were 22.7% 18.2% 22.7% respectively. There was only statistically significance between erosive group and non-erosive group (P<0.05) in expression of bcl-2. CONCLUSION: Apoptosis and apoptotic related genes p53 bcl-2 C-myc participate in clinical development of OLP. The changes of AI are related to the chronic inflammation of lesion with OLP. We should attach importance to over expression of bcl-2 in erosive OLP.

Proton-induced cation current in embryonic rat spinal cord neurons changes ion dependency over time in vitro.

A rapid increase in proton concentration [H+]0 induces Na+ conductance in a variety of cell types. Here we report that H+ trigger a cation-selective channel whose ion dependency changes over time in culture. Whole-cell recordings of ventral spinal cord neurons dissociated at E15 and cultured for up to 14 days revealed that more than 80% had H(+)-induced inward current responses exhibiting a rapid decay phase. The current response was activated beginning about pH 6.8. Following decay, several minutes were required for complete recovery. More modest decreases in pH, which by themselves failed to activate this current, depressed those triggered by effective changes in pH. The currents recorded from cells in culture for less than 7 days could be abolished completely in the absence of Ca2+ and persisted in Na(+)-free and Ba(2+)-containing solutions. Ensemble analysis of current fluctuations recorded at the peak of the current allowed us to estimate a unitary channel conductance of 7.0 pS and a mean open time of 4.1 ms. In neurons cultured 2 weeks or more, protons induced an inward current response with similar kinetic properties, but with [Na+]0 dependency. Thus, proton-activated cation conductance in embryonic rat spinal cord neurons is self-limiting and involves brief openings of cation-selective channels whose ion dependency changes over time in culture.

Randomized, controlled trial of high-dose intravenous pyridoxine in the treatment of recurrent seizures in children.

To determine the efficacy of pyridoxine in treating seizures, 90 infants and children with recurrent convulsions primarily due to acute infectious diseases were enrolled in the present study. Forty patients were treated with high-dose pyridoxine (30 or 50 mg/kg/day) by intravenous infusion, and 50 subjects served as controls. Antiepileptic drugs and other therapies were similar in the two groups except for pyridoxine. Clinical efficacy criteria were based on the frequency of convulsions per day and on the duration of individual seizures after therapy was initiated. The results indicated that total response rates in the pyridoxine group and control group were 92.5% and 64%, respectively (chi-square = 14.68, P < .001). After initiation of therapy, seizures resolved after 2.4 +/- 1.4 days in the pyridoxine group and after 3.7 +/- 2.0 days in the control group (t = 3.67, P < .001). No adverse effects of pyridoxine were apparent during the observation period. We conclude that pyridoxine is an effective, safe, well-tolerated, and relatively inexpensive adjunct to routine antiepileptic drugs for treatment of recurrent seizures in children.

Mitochondrial gene transfer ribonucleic acid (tRNA)Leu(UUR) 3243 and tRNA(Lys) 8344 mutations and diabetes mellitus in Korea.

The high prevalence of diabetic patients with a mutation in the mitochondrial gene (the 3243 and 8344 bp mutations) has been identified in Japan. To determine the prevalence of diabetic patients with those mutations in Korea, we randomly screened selected 503 clinical diabetic patients regardless of their diabetes types. We found only 1 patient with the mitochondrial DNA mutation at position 3243 (percent mutation, 32%), and the mitochondrial DNA mutation at position 8344 was not found in any of the patients. The affected subject was a 22-yr-old man with a history of myoclonic epilepsy and mild sensorineural hearing loss, a 1-yr duration of diabetes mellitus, and a low level C peptide response to oral glucose. Because of the low frequency of these mutations in the Korean diabetic population, we concluded that these particular mutations of mitochondrial DNA may not be a common contributor to diabetes mellitus in Korea.

[A longitudinal study of growth and development of low birth weight infants].

In order to investigate the growth and development of low birth weight infant (LBWI), 132 low birth weight infants (BW < 2500g) born in 1983 living in Xi Hui district were followed up from birth to three years. And 49 full-term infants with normal birth weight were selected as controls. The weight, length, and head circumference of infants of two groups were measured periodically. Their development abilities were assessed by using Gesell Development Diagnosis. Results showed the weight, length, and head circumference of LBWI were obviously less than that of the controls. The developmental quotient (DQ) of LBWI were lower that that of controls, particularly in gross motor field, Therefore, it is important to reduce the rate of IBWI and to give scientific guidance on child care and early training for promoting growth and development of infants.

Biomechanical evaluation of cervical spine instability after multiple level laminectomy.

Four fresh human cervical spine specimens (C2-T1) were tested both intact and with C5-C6 laminectomies to evaluate the biomechanical effects of multiple level laminectomy. The loads applied to the specimens were physiological and clinically relevant motion patterns were simulated. The results showed that C5 vertical displacements increased by 83.33% in axial compression, 168.75% in flexion, 106.09% in extension, and 35.14% in left bending after C5-C6 laminectomies compared with intact specimens. The increased rates of C6 vertical displacements after laminectomy were slightly lower than C5. The anterior horizontal bulging of C5-6 discs increased by 29.69% in axial compression, 13.86% in flexion, 61.79% in extension, and 13.40% in left bending after laminectomy. The rotational angles of whole specimens had an increase of 15% after laminectomy. The strains in the anterior vertebral bodies and posterior laminae near the articular processes of C5 and C6 were increased significantly after laminectomy. The data indicated that multiple level laminectomy can lead to biomechanical instability of the cervical spine.

Purification of bovine cholecystokinin-58 and sequencing of its N-terminus.

Molecular cloning of cholecystokinin (CCK) mRNA from porcine brain and gut has demonstrated that CCK is synthesized as an identical precursor in both tissues. The sequence for porcine CCK-58 predicted from CCK cDNA was identical with the amino acid sequence of the peptide purified from different lots of animals. However one group did report that there were differences in the N-terminus of CCK-58 purified from the intestines of two different lots of mongrel dogs. In the current report it is demonstrated that the amino acid sequences of CCK-58 purified separately from three bovine brains are identical through the first 19 N-terminal amino acid residues. The peptides were sequenced for ten additional steps and were shown to be identical with the previously reported sequences for the N-terminus of CCK-39. The N-terminus of bovine CCK-58 has the following sequence: AVPRVDDEPRAQLGALLAR.

Circulatory crisis in free toe-to-hand transfer and its management: I. Clinical experience.

On the basis of experience with 226 patients undergoing free toe transfer for reconstruction of the thumb and fingers, circulatory crises during and after surgery are analyzed and classified according to the time of crisis occurrence. Prompt re-exploration is indicated when the crisis occurs within 24 hr postoperative. Crises occurring 48 hr and more postoperative are usually due to vasospasm, and antispasmodic therapy should be instituted. Surgical intervention in these later cases may prove fruitless.

Microsurgical anatomy of the lateral skin flap of the leg.

The blood supply, venous drainage, and innervation of the lateral skin flap of the leg were studied in 52 fresh specimens by dissection, angiography, and selected injection of cutaneous arteries with india ink. This flap has a long vascular pedicle that contains the peroneal artery and vein and their branches. The diameter of the peroneal artery is about 3.7 mm. There are 4 to 7 cutaneous branches that penetrate the posterior intermuscular septum and widely anastomose with each other in the subcutaneous tissue. The skin area supplied by the peroneal artery measures approximately 32 X 15 cm. There are 2 sets of veins in the flap: (1) 2 deep veins, the peroneal veins, are about 4 mm in diameter, and (2) a superficial vein, the small saphenous vein, is about 3.3 mm in diameter. The cutaneous nerve is the lateral sural cutaneous nerve. This new lateral skin flap of the leg may be used either as a free flap or as a cutaneous flap and has been proven successful clinically.
