[Activation of the macrophage/lymphocyte system in AIDS, ARC and AIDS risk groups].

Elevated neopterin levels are indicative of activation of the cellular immune system. Studies on excretion of neopterin in patients with AIDS and ARC, as well as in members of risk groups have demonstrated repeated or permanent stimulation of the immune system. This stimulation can be observed in all risk groups independent of LAV/HTLV-III infection. Additionally, in vitro replication of LAV/HTLV-III has been observed to be quantitatively greatest in activated CD4+-lymphocytes. Hence, we conclude that activation of the cellular immune system represents the central cofactor for progressive LAV/HTLV-III infection. These findings seem to contrast with most reports in the literature to date, but observations of other authors corroborate them. As consequence of these studies, therapeutic regimens using immunostimulatory strategies should be prevented in AIDS and ARC patients. Immunosuppressive treatment should be considered.

Structural components of bovine leukemia virus: further biochemical and immunological characterization of major structural proteins and glycoproteins.

Proteins from bovine leukemia virus (BLV) were investigated by sodium dodecyl sulphate-polyacrylamide gel electrophoresis in the presence and absence of 14C amino acids. Virus grown either in fetal lamb kidney (FLK) or bat lung (BL) cells revealed seven major proteins designated p10, p12, p15(1), p15(2), p24, gp30, and gp64. By tryptic peptide analysis, N-terminal amino acid analysis and radioimmunoassay it could be shown that p10, a basic protein with hydrophobic properties, is a cleavage product of p15(2), the acidic and slightly hydrophobic phosphoprotein of BLV. The structural protein p12 was shown to be a phosphorylated protein identifying it as a second major viral protein to contain phosphorous. Investigation of [3H]glucosamine incorporation, N-terminal amino acid analysis and hydrophobic properties by chloroform-methanol extraction confirmed properties of gp30 and gp64 predicted by nucleotide sequence data. The two p15 proteins have been characterized in more detail.

Influence of different culture conditions on BLV expression in permanently infected FLK cell lines.

The permanently BLV-infected FLK cell line 44/2 and FLK sublines were tested for the stability of their BLV and antigen synthesis by three virus markers, p24, gp51, and activity of reverse transcriptase. The extent of BLV production in the FLK line correlated directly with the surface for cell growth in roller and stationary cultures. The synthesis and secretion of non-virus-associated gp51 is especially stimulated in the roller culture, and is largely independent of the quality of the culture medium. The roller culture allows considerable economy of the medium, at the same time retaining its full biological activity. As much as 2 mg of gp51 per litre of culture supernatant was obtained by this procedure. Appropriate markers for estimation of BLV production are p24 and gp51. For this purpose, the activity of reverse transcriptase on its own was not sufficient. The BLV yield differed by one order of magnitude among the 18 cloned FLK sublines. Three sublines have been identified, which showed a comparatively high virus production, under conditions of stationary cultures. The amount or activity of viral markers could not be correlated with the number of BLV proviruses.

Isolation and characterization of cell clones producing various amounts of bovine leukosis virus.

Several single-cell clones were isolated from lamb kidney cell line (FLK) persistently infected with bovine leukosis virus (BLV). The clones differed in the amount of produced virus, some clones were highly virus productive, others produced less virus. The restriction analysis of cell DNA from clones revealed the existence of several numbers of integrated BLV-DNA proviruses in various cell clones. In some cell clones the unintegrated proviral DNA was observed. The proviral BLV sequences were found to be integrated in reiterated DNA in some cell clones. There was no apparent relationship between the number of integrated BLV proviruses and the extent of virus production. The origin of the different numbers of integrated BLV proviruses in various cell clones is discussed.

Bracing and supporting of the lumbar spine.

The orthopaedic surgeon should be familiar with various supports and braces for the treatment of low back pain. Severe cases of spinal instability always need a Hohmann overbridging brace, whereas the milder form of motion-segment instability is treated with one of the elastic supports. In cases of osteoporosis of the spine and insufficiency of the lumbosacral junction the Lindemann 2/3 semi-elastic brace is prescribed.

[Improved test for the detection of reverse transcriptase of bovine leukosis virus].

The revertase test with exogenous matrix, poly-(rA) . oligo-(dT), for the detection of bovine leukemia virus from crude virus sediments was standardised and miniaturised. An amount of 10 ml of cell culture supernatant of short-term cultured lymphocytes (5 . 10(6) cells/ml) is quite sufficient for testing one cattle sample. The lower sensitivity limit of the test was found to be 10(6) virus particles. The test is properly reproducible, within tolerance limits of +/- 20--30 per cent, provided that optimum lysis conditions be maintained (0.01 per cent triton X-100, 20 minutes, 0--4 degrees C incubation) and under the condition that the protein quantity in 100 microliter test solution does not exceed the threshold of 3--15 micrograms. The specificity of the test is based on the use of free viruses from cell culture supernatant, the optimum temperature of the revertase reaction at 25 degrees C, which actually deviates from that for cellular DNA polymerases, that is 37 degrees C, and magnesium ion concentration which has to be optimum for bovine leucosis virus revertase. Two-hundred heads of cattle, differing by haematological status, were examined, and 56 per cent of them were, clearly, virus producers, among them 95 per cent of all animals with positively established leucosis and 36 per cent of the haematologically intact animals. Examinations of individuals have shown that in repetitive checks, carried out in intervals between two months and one week, the revertase activities varied by something between 0.5 and two magnitudes.

Familial occurrence of syngnathia congenita syndrome.

A case of familial occurrence of syngnathia congenita in mother and child is described; this syndrome has been seen only sporadically until now. Due to the distinct appearance of the intraoral chords--they were situated on both sides between the ridges of the jaw and also between the left half of the bifid uvula and the retrolingual region--a relationship to the autosomal-dominantly transmitted Cleft Palate Lateral Synechiae Syndrome (CPLS-Syndrome) is suspected. Complete penetrance with different expressivity may explain the different clinical manifestations of the cases seen up to now and the absence of bifid uvula in the mother of our patient. The probable pathogenesis during embryological development is discussed.

Purification and characterization of bovine leukemia virus DNA polymerase.

An RNA-directed DNA polymerase was purified from bovine leukemia virus (BLV) by successive glycerol gradient centrifugation, column chromatography on phosphocellulose and gel filtration on Sephadex G-200. The purified DNA polymerase transcribes heteropolymeric regions of 30--40 S RNA isolated from avian myeloblastosis virus. The enzyme differs from other known DNA polymerases of mammalian type-C RNA tumor viruses by the following properties: 1. Its apparent molecular weight as estimated by velocity sedimentation data is 58,000 at 0.12 M KCl and 43,000 in the presence of 0.50 M KCl. 2. It has a Mg2+ optimum of 10 mM, and a Mn2+ optimum of 0.25 mM with (rA)n-(dT)10 as template. 3. At 50 mM KCl it is inhibited more than 70%, but it is not inhibited by phosphate ions at 2 mM. These properties confirm the peculiar position of BLV within the family Retraviridae.

The biosynthesis of glycolipids during the differentiation of the slime mold Dictyostelium discoideum.

In the slime mold Dictyostelium discoideum polysioprenylphosphomannosides are substrates for membrane bound mannosyltransferases; the isolated and purified isoprenyl derivatives transfer mannose to protein in vitro in presence of membrane fractions. The biosynthesis of the mannolipids as well as the biosynthesis of a glucose containing cerebroside, which becomes synthesized in an early stage of the cell development proceeds under control of the cell differentiation. The isolation procedure and the properties of the glycolipids are described, and their functions for the cellular development are discussed.

[Studies on the virus demonstration in enzootic bovine leukosis using the revertase test].

This exogenic test for the detection of enzyme activities (revertase) is helpful in differentiating between leucocytes of leukaemic cattle, on the one hand, and those of intact animals, on the other. The mechanism of such identification is based on differences in enzyme activity in the presence of poly rA/oligo dT and poly dA/oligo dT. The best results were obtained from short-time cultured leucocytes which gave ten times higher margins of incorporation by comparison between intact and preleucotic cattle. The morphological findings obtained by electron microscopy confirmed the results by other authors and enlarged them by determination, for the first time, of RNA filaments and protein rings.

[Orthopedic problems in hemophilia. Pathophysiology and results of operations (author's transl)].

During the last 3 years we have treated over 400 patients with severe hemophilia A or B of all ages. They were looked after by the Institute for Experimental Hematology and Blood-Transfusion of the University at Bonn, for prevention, rehabilitation and operation. The specific pathophysiology of disability of the hemophiliac is shortly outlined, the chances of operative rehabilitation and their indications are discussed. The results show that--with experience--extensive operative rehabilitation can be achieved also in hemophiliacs.

[Ruptures in the rotator aponeurosis (author's transl)].

Ruptures in the rotator aponeurosis occur frequently. Minor defects are usually not discovered and are classified in the "painful shoulder" syndrome. Depending on the localization and extent, extensive destruction, which occurs suddenly under particular circumstances, leads to various functional disturbances including the so-called pseudoparalysis of the arm. Smaller ruptures generally respond to the usual conservative measures used for the supraspinous syndrome. Surgical treatment of larger ruptures is promising and good results can be achieved with the appropriate technique and adequate follow-up treatment. In the case of extensive defects, they represent the only chance for restoring satisfactory function. Even with long standing, unsuccessful treatment for "periarthritis humeroscapularis", the necessary diagnostic and/or surgical consequences, should be carried out.
