Prevalence of antibodies to HTLV-III in AIDS risk groups in West Germany.

The prevalence of antibodies to human T-lymphotropic virus III was determined in acquired immunodeficiency syndrome (AIDS) risk groups by an enzyme linked immunosorbent assay and confirmatory tests in four different areas in West Germany. Twenty-four of 28 homosexual AIDS patients (86%), 24 of 33 homosexual patients with lymphadenopathy syndrome or AIDS related complex (73%), and 44 of 113 asymptomatic homosexuals at risk for AIDS (39%) were seropositive. In three groups of hemophiliacs, 8 of 35 in 1983 (23%), 25 of 65 in early 1984 (39%), and 19 of 23 in late 1984 (83%) showed positive results. Two sera from 36 polytransfused patients were also positive, whereas 36 selected blood donors, and 32 healthy laboratory and clinical personnel were all negative. Also no human T-lymphotropic virus III antibodies were detected in sera of 187 prostitutes in the Munich area.

[Preliminary results of Re-PUVA therapy in cutaneous T-cell lymphomas].

Eight patients with T-cell-lymphoma of the skin (6 mycosis fungoides, 2 parapsoriasis variegata) were treated with Tigason in combination with PUVA-therapy. The daily dose of Tigason was 50-100 mg. There was complete regression of skin lesions in three patients, partial regression in three patients, and an improvement in two patients.

[Sporadic, endemic and AIDS-related Kaposi's sarcoma. Different clinical course variants of the tumor demonstrated by 3 typical cases].

We report on three patients suffering from Kaposi's sarcoma (KS) of the classical, endemic, and AIDS-related variant. We point out the uniformity of this tumor in terms of histomorphology. We found the characteristic findings of angiomatous structures and dense spindle cell formations. All variants of the tumor show histochemical and immunohistological evidence for a histogenesis probably from endothelial cells of venous capillaries. On the basis of our cases and the literature, we discuss to what extent cellular immunity may determine both the tumor biology and therapeutical considerations.

[Marked regression of melanoma metastases under combined chemotherapy].

We report a 60-year-old patient suffering from malignant melanoma and multiple cutaneous, lung, and intraabdominal metastases. After 6 series of combination chemotherapy ( BNCU , Hydroxyurea, Dacarbacin ), the metastases showed a remarkable regression. At present, the patient is in a nearly complete remission.

Quantitative determination of complement components produced by purified hepatocytes.

In this report we describe, on a quantitative basis, the secretion of complement components by hepatocytes. Primary cultures were established after isolation of the cells from guinea-pig liver and the synthesis of C3, C5, C4 and C2 was measured. The cells were isolated by collagenase perfusion of the liver followed by differential centrifugation. The contamination of the hepatocyte suspension with non-parenchymal cells was less than 1%. At 24 h after plating the cells the kinetics of complement production were measured. C3 and C5 content in the culture medium harvested at different time intervals was determined by a sensitive ELISA. Secretion of C2 and C4 was measured haemolytically using C2 or C4 deficient guinea-pig serum. Under the conditions used hepatocytes secreted C3 at a rate of about 100 ng/10(6) cells/h with a plateau of secretion after 24 h of culture corresponding to about 350,000 molecules/cell/h. C5 secretion was detectable after 3-6 h of culture. The C5 secretion rate was about 15 ng/10(6) cells/24 h. The functional activity of C4 and C2 in the supernatants amounted to about 80 SFU/cell/h if the culture medium was changed every 3 h but dropped significantly if the medium was changed every 12 h. The decrease of the haemolytic activity became stronger if the medium was changed every 24 h. Cycloheximide reversibly inhibited the complement production. Our results show that guinea-pig hepatocytes synthesize considerably more C3 and C5 compared to peritoneal macrophages supporting the hypothesis that hepatocytes provide the major source of plasma complement.

C3a induced activation and stimulus specific reversible desensitization of guinea pig platelets.

C3a and its C-terminal hexapeptide lead to a dose dependent release of biogenic amines and nucleotides stored in platelet's granules. The release reaction can be measured by tritiated serotonin or by ATP, indicated by an ATP specific bioluminescence assay. We tested the capability of C3a to induce aggregation of washed platelets. The recently described phenomenon of low dose, stimulus specific desensitization of platelets to the anaphylatoxic peptides C3a and C5a could be shown by measuring the release reaction as well as the aggregation. Further we could demonstrate the reversibility of the stimulus specific desensitization within 2 to 3 hours. The desensitization was proven to be temperature dependent. The recovery of function was independent of newly synthetized protein and is discussed as the result of receptor-recycling.
