[Successful collection of peripheral blood stem cells mobilized by a combination of G-CSF with high-dose Ara-C or high-dose etoposide].

The author evaluated the effectiveness of a combination of G-CSF with high-dose Ara-C (HD Ara-C) or high-dose etoposide based on the collection of peripheral blood stem cells (PBSC) during the recovery phase from myelosuppression in 17 patients with various types of hematopoietic malignancies. Following therapy with HD Ara-C or HD etoposide, G-CSF (250 micrograms/body) was administered from the nadir state and blood mononuclear cells were collected using a CS-3000 by processing 15 litres of blood at each apheresis. Stem cell-enriched fractions obtained from discontinuous Percoll gradients were collected and stored in liquid nitrogen. The mean numbers of granulocyte-macrophage colony-forming units (CFU-GM) obtained by the first apheresis were 10.9 x 10(5)/kg after chemotherapy with HD Ara-C and 13.4 x 10(5)/kg after HD etoposide therapy, respectively. In 14 out of the 17 first apheresis, more than 2 x 10(5) CFU-GM/kg were collected. Serial apheresis was performed in seven patients, at intervals ranging from 21-79 days, and the progenitor yield in the second apheresis decreased in five patients. These results suggest that the use of either HD Ara-C or HD etoposide induces an effective mobilization of PBSC and enables large scale collection of PBSC for autotransplantation.

Effects of fluorocarbon infusion on microcirculation during cardiopulmonary bypass.

A pulsatile pump was used to effect a three-hour cardiopulmonary bypass in 15 mongrel dogs. During the bypass, Fluosol-DA (35%) infusion was used in lieu of homologous blood transfusions. The greater omentum was chosen as a model of extraorganic microcirculation, and its reaction to the infusion was observed. The omentum's microcirculation was not adversely affected by the Fluosol-DA (35%): no sludging, microthrombi, arteriovenous fistulas, plasma skimming, stasis, or other adverse findings were observed. The animals' oxygen consumption did not decrease, and the resistance of the systemic blood vessels, did not increase during the experiment, suggesting that the fluorocarbon infusion had no untoward effects on the microcirculation.

Carcinogenicity of benzyl chloride, benzal chloride, benzotrichloride and benzoyl chloride in mice by skin application.

The carcinogenicity of benzyl chloride (BYC), benzal chloride (BAC), benzotrichloride (BTC) and benzoyl chloride (BOC), which were suspected as causative agents of lung cancer and maxillary malignant lymphoma of workers employed in factories producing BOC, was examined by skin applications in female ICR mice. After rather high dose exposure, BTC exhibited leukemogenic and pulmonary tumorigenic activities as well as potent dermal carcinogenic activity. After administration of the chemicals at the dose of 2.3 microliter/animal, twice a week for 50 weeks, BTC induced 68% incidence of skin cancers and 58% incidence of pulmonary tumors (including 10% of lung carcinomas) within 399 days. Incidence of skin cancers was 58% for BAC, 15% for BYC and 10% for BOC within 560 days. Considering the extent of possible exposure of the workers to these chemicals in the working environment and the carcinogenic potency of the chemicals tested, it can be concluded that BTC was very probably responsible for causing the cancers seen int he workers employed in manufacturing BOC.
