Isolation, characterization and sequence of a cDNA encoding gamma-tubulin protein from the fern Anemia phyllitidis L. Sw.

A complete cDNA encoding gamma-tubulin protein from Anemia phyllitidis is presented. The deduced amino acid sequence shows an average similarity of 81-86% to known gamma-tubulin genes from insects, mammals and fungi. Northern blots indicate the existence of transcripts of ca. 1.9 kb which are expressed during gametogenesis of A. phyllitidis. The gene described is the first cloned and entirely sequenced gamma-tubulin gene in any plant.

PCR-dependent amplification and sequence characterization of partial cDNAs encoding myosin-like proteins in Anemia phyllitidis (L.) Sw. and Arabidopsis thaliana (L.) Heynh.

Partial cDNAs encoding for myosin-like proteins from Anemia phyllitidis and Arabidopsis thaliana have been isolated using PCR technology. The deduced amino acid sequences show an average similarity up to 62% with known myosin heavy chain genes. From northern blot analysis we were able to estimate that transcripts of ca. 6.1 kb size are expressed in A. phyllitidis.

Detection and partial sequence of phytochrome genes in the ferns Anemia phyllitidis (L.)Sw (Schizaeaceae) and Dryopteris filix-mas L. (Polypodiaceae) by using polymerase-chain reaction technology.

Phytochrome controls several developmental steps during formation and differentiation of the fern gametophyte, including spore germination, morphogenesis of the gametophyte or differentiation of the sexual cells. To obtain information about the amino acid sequence and the regulation of phytochrome expression at the gene level, two degenerated oligonucleotides against well conserved amino acid regions were designed after an optimal alignment of the known phytochrome sequences. These primers were tested against DNA isolated from Arabidopsis thaliana, and the polymerase-chain reaction (PCR) products were cloned and sequenced. The DNA fragment produced with this method proved to be identical with a phytochrome-A-gene fragment from A. thaliana, and hence this fragment was used in further experiments to prove whether amplified DNA from fern species contains phytochrome-like DNA. With this procedure we successfully detected and cloned gene fragments both from gametophytes of Anemia phyllitidis and Dryopteris filix-mas, cultured for 7 days under vegetative conditions. In addition, poly(A)+ RNA was prepared from 7-day-old gametophytes of A. phyllitidis, induced to differentiate antheridia under generative conditions. This poly(A)+ RNA was transcribed into complementary DNA and used together with both phytochrome specific primers in a PCR experiment. We thereby obtained another DNA fragment. These data strongly suggest that A. phyllitidis has at least two phytochrome genes, and that at least one of them is expressed in light-grown gametophytes.

Aphidicolin Inhibition of DNA Synthesis and Germination in Spores of Anemia phyllitidis L. Sw.

Aphidicolin inhibits DNA synthesis and nuclear division in spores of Anemia phyllitidis. In spite of blocked DNA replication, spores germinate under continuous dark conditions, if induced by addition of 5 x 10(-5) grams per milliliter gibberellic acid. Differentiation of aphidicolin-treated prothallia indicate the existence of a prepattern in the dry spore which is realized independent of cell division during early events of spore germination.

In vivo phytochrome difference spectrum from dark grown gametophytes of anemia phyllitidis L. Sw. Treated with norflurazon.

An in vivo phytochrome difference spectrum of dark grown Anemia phyllitidis L. Sw. gametophytes has been measured. The spectral characteristics estimated from the difference spectrum were as follows: red maximum at 662 nanometers, far red maximum at 737 nanometers, isosbestic point at 695 nanometers, and a shoulder from 620 to 630 nanometers. To diminish the influence of chlorophyll on phytochrome measurements in dark grown gametophytes, total chlorophyll content could be reduced by about 65% by treatment with 1 micromolar Norflurazon. In gametophytes grown in red light, the inhibition caused by 1 micromolar Norflurazon was 97.5%, based on g spores. Total carotenoid content of dark grown gametophytes was not appreciably different from that already present in dry spores. Herbicide treatment reduced this level to about 60%. Gametophytes grown in red light showed a nearly 7-fold increase in total carotenoids compared with that of the dark control. In treated light grown protonemata, no carotenoid accumulation took place, and the level remaining corresponded with that found in treated dark grown protonemata.

Effects of 5-bromo-deoxyuridine on chloroplast structure in gametophytes of Anemia phyllitidis L. Sw.

Application of 5-bromo-2-deoxyuridine causes servere changes in the ultrastructure of chloroplasts. These modifications include an increase of thylakoid layers ("giant"-grana), coiling of thylakoids, and lastly vacuolization of the entire membrane system of the chloroplast. Such observations can be correlated with types of membrane aberrations reported for natural mutants.
